Abstract

Terapi suatu obat yang bersamaan dengan konsumsi makanan dapat mempengaruhi efektivitas obat tersebut. Penelitian ini bertujuan untuk mengetahui pengaruh praperlakuan jus kubis bunga (Brassica oleracea L. var. botrytis L.) terhadap aktivitas diklofenak dalam terapi inflamasi pada tikus putih jantan galur Wistar. Tiga puluh ekor tikus dibagi enam kelompok secara acak. Kelompok I (kontrol negatif) hanya diberi aquadest, kelompok II (kontrol positif) diberikan perlakuan natrium diklofenak dosis 5,04 mg/kg BB secara peroral. Kelompok III diberi praperlakuan jus kubis bunga dosis 20 ml/kg BB 1 jam sebelum perlakuan natrium diklofenak. Kelompok IV diberi praperlakuan jus kubis bunga dosis 20 ml/kg BB 1 hari sebelum perlakuan natrium diklofenak. Kelompok V diberi praperlakuan jus kubis bunga dosis tunggal 20 ml/kg BB selama 3 hari sebelum perlakuan natrium diklofenak dan kelompok VI diberi praperlakuan jus kubis bunga dosis tunggal 20 ml/kg BB selama 5 hari sebelum perlakuan natrium diklofenak. Tiga puluh menit setelah perlakuan natrium diklofenak, kemudian telapak kaki tikus diinjeksi dengan karagenin 1% sebanyak 0,10 ml secara subplantar. Volume udema diukur dari jam ke-0 sampai jam ke-5 setelah injeksi karagenin menggunakan pletismometer. Hasil analisa statistik data AUC dan DAI menunjukkan bahwa praperlakuan jus kubis bunga yang diberikan 1 jam sebelum pemberian natrium diklofenak tidak memperlihatkan perubahan yang bermakna terhadap kontrol positif. Hasil penelitian ini menunjukkan bahwa praperlakuan jus kubis bunga selama 3 hari dan 5 hari mampu menurunkan daya antiinflamasi dari diklofenak, sehingga terapi inflamasi dengan diklofenak menjadi tidak maksimal.

Abstract

Cytochrome  P-450  as  a  major  component  enzyme  system  in  drug metabolism. Activity  enzyme  of  cytochrome  P-450  was  influenced  by  internal and external factors. Vegetables of Brassicaceae such as cauli flower were often consumed  in  the  long  term,  its   has  inductor  activity  of  oxidation  enzyme systems and conjugation reactions. Theophylline as a bronchodilator drugs have the unknown effects on hepatic microsomal enzyme, such as cytochrome P-450 and   have  not  been  many  studies  that  tried  to  prove  it.  The  purpose  of  this study  was  to  prove  the  effects  of Cauliflower  and  indol  on  level  cytochrome  P-450  enzyme.  90  rats  were  divided  into  3  groups.  Group   I,  were  given Theophylline  20  mg/kg  BW.  In  group  II  30  rats  were  treated  with  indole  1,2; 2,4; 3,6 mg/kg BW, and group III 30 rats were treated with cauliflower extract respectively  doses  100,  200,  300  g/kg  BW.  Each  dose  was  given  on  10  rats, each group were divided 2 sub-groups were treated for 5, 10 days. On the last day  of  treatment  were  given  Theophylline  20  mg/kg  BW.   Cytochrome  P-450 enzyme  levels  were  determined  by  the  method  of  Omura  and  Sato(Snell,  and Mullock,1987). An induction cauliflower and indole did not increased levels of hepatic  cytochrome  P-450.  The  long  treatment  and  the   increased  of administered  dose  did  not  enhanced  the  levels  of  hepatic  cytochrome  P-450 enzyme. cauliflower and indole contained in vegetables when consumed together with theophylline drug, would not affect the metabolism of theophyllineKey words: Cauli flower, cytochrome P-450, theophylline, indole, spectrofotometric.

Abstract

The purpose of this study was to explore the role of Affective Commitment (AC) as a mediating variable in encouraging organizational citizenship behavior in Private Universities in Central Java, Indonesia. The study explains whether Affective Commitment (AC) is able to mediate the relationship between Leadership Style (LS) and Subjective Well-being (SWB)  with Organizational Citizenship Behavior (OCB). The sample in this study is permanent staff and lecturers who work in Private Universities in Central Java. Data from 124 respondents were collected by utilizing the questionnaire.The sample sample used in this study is 150 respondents. It was taken by applying proportional random sampling.  Only the 150 questionnaires distributed, 124 were returned, for a response rate of 82%.After processing and modifying the data, only 124 respondents used as the main resources analyzed by using Structural Equation Model (SEM). Statistical analysis reveals that there exists significant relationship between Subjective well-being, Leadership Style, and Organizational Citizenship Behavior; likewise, Affective commitment  mediates the relationship between Subjective well-being, Leadership Style, and Organizational Citizenship Behavior. Limitations and future implementations of this research are also discussed

Abstract

This paper main objective was to investigate whether  career development has impact on employees performance. This paper is quantitative in nature. Data for the paper have been collected through primary source that are from questionnaires surveys. The data have been checked through statistical software to find the impact of career counseling on worker efficiency, and career mentoring and skill assessment on employees performance. The population of this study was 1,116 lecturers and administrative staff  of 10 Private Higher Education in Central Java, Indonesia., and the taken sample was 250 respondents. Descriptive statistic tools SPSS were applied on the questionnaire to see the reliability and consistency. The result showed that career counseling had a positive and significant effect  on worker efficiency, and career mentoring and skill assessment had a positive and significant effect  on employees performance. The study recommends that organizations should encourage career counseling activities as a leeway of providing a sustainable framework upon which employees acquire the needed operational skills that will enhance their job performance in a typical organization.

Abstract

ABSTRACTActive compounds of cruciferae family and the change of total cholesterol level and vitamine C in hipercholesterolaemic ratsBackground: Cauli flower is a member of the Cruciferae family. One of the active compound content of indole-3-carbinol, is used to prevent and treat cancer. Vitamine C in the cauli flower as an antioxidant is able to avert free radicals that are formed in aerobic organisms and hipercholesterolaemic condition. The study aimed to determine the active compounds contained in the of cauli flower and its role in changing of the lipid profile and vitamine C levels in hipercholesterolaemic rats.Methods: To identify up the active compounds of cauli flower by maceration repeately, with alcohol: ammoniak (19:1). The results were determinated by thin layer chromatography (TLC). Spotting results obtained were compared with standard spots in its color reaction and Rf spots with Rf standard samples. Twenty rats were divided into 4 groups, and made hipercholesterolaemic, group I as a negative control, group II as a positive control were given green tea 0.605 g/kgBW, group III-IV were given cauli flower juice for 14 days dose of 10 and 15 ml/kgBW, the blood were analyzed for total cholesterol level and vitamine C plasma.Results: Cauli flower active compounds are indole-3-carbinol, indole-1-3-carbinol, indole-3-carboxylic acid and sulforafan they were capable of decreasing total cholesterol levels to 45%, and increasing plasma vitamine C level by 55% compared to before treatment.Conclusion: Cauli flower contains 4 indoles, and is able to decrease total cholesterol and increase vitamine C plasma in hipercholesterolaemic rats.Keywords: Cruciferae, indole, TLC, cholesterol, vitamine CABSTRAKLatar belakang: Bunga kubis, termasuk keluarga Cruciferae. Salah satu kandungan senyawa aktifnya indol-3-karbinol, mampu mencegah dan mengobati penyakit kanker. Vitamin C yang terdapat di dalam bunga kubis sebagai antioksidan mampu menangkal radikal bebas yang terbentuk pada organisme aerobik, kondisi hiperkolesterolemia. Penelitian ini bertujuan untuk mengetahui senyawa aktif yang terkandung dalam bunga kubis (Brassica Oleracea var Botrytis L) dan perannya dalam mengubah profil lipid dankadar vitamin C pada tikus hiperkolesterolemia.Metode: Untuk menyari senyawa aktif bunga kubis dilakukan macerasi berulang, dengan alkohol: ammoniak (19:1). Hasil penyarian dilakukan penetapan senyawa aktif secara kromatografi lapis tipis (KLT). Hasil bercak yang diperoleh dibandingkan dengan bercak standar dengan membandingkan reaksi warna dan Rf bercak sampel dengan Rf standar.Untuk mengamati efek farmakologi, 20 ekor tikus dibagi menjadi 4 kelompok, dibuat hiperkolesterolemia, kelompok I sebagai kontrol negatif, kelompok II sebagai kontrol positif diberi teh hijau 0,605 g/kgBB, kelompok III-IV diberi jus bunga kubis bunga selama 14 hari dosis 10 ml/kgBB dan 15 ml/kgBB, data darah dianalisis kadar kholesterol total serum dan vitamin C plasma.Hasil: Bunga kubis (Brassica Oleracea var Botrytis L) mengandung senyawa aktif: indol-3-karbinol, indol-1-3-karbinol, indol-3- asam karboksilat dan sulforafan yang mampu menurunkan kadar kolesterol total serum sampai 45% dan meningkatkan kadar vitamin C plasma sampai 55% dibanding sebelum perlakuan.Simpulan: Bunga kubis mengandung 4 senyawa indol dan mampu menurunkan kolesterol total serum dan meningkatkan vitamin C plasma pada tikus hiperkolesterolemia

Abstract

A drug therapy together with food consumption could influence the drug effectiveness. This research aimed to know the influence of cauliflower (Brassica oleracea L. var botrytis L.) juice pretreatment on diclofenac activity in inflammation therapy to wistar white male rats. Thirty rats divided into 6 groups randomized design. Group I (negative control) was only given aquadest, while group II (positive control) was given treatment with sodium diclofenac orally and the dose was 5,04 mg/kg BW. Group III was given pretreatment cauliflower juice with the dose of 20 ml/kg BW. It was given one hour before treatment with sodium diclofenac. Group IV was given pretreatment cauliflower juice with the dose of 20 ml/kg BW and it was given one day before treatment with sodium diclofenac. Group V was given pretreatment cauliflower juice with single dose 20 ml/kg BW for 3 days before treatment with sodium diclofenac,  and group VI was given pretreatment cauliflower juice with single dose 20 ml/kg BW for 5 days before treatment with sodium diclofenac. Thirty minutes after treatment with sodium diclofenac, the rat’s paw was injected with 1% carrageenan 0,10 ml subplantary. Edema volume was measured from 0th hour to 5th hour after carrageenan injection used the pletismometer. The result of statistic analysis of AUC and DAI data have shown that cauliflower juice pretreatment given one hour before treatment with sodium diclofenac did not show significant different with positive control. The research showed that cauliflower juice pretreatment for 3 and 5 days was capable to reduce antiinflamation activity of diclofenac, therefore inflammation therapy with diclofenac was not maximum.

Abstract

Traditional herba medicine, Gadung (Dioscorea hispida Dennst) has already known can be used for oral anti diabetic drug. But it have not be proved experimentally. The purpose of experiment want to know the influence of Gadung (Dioscorea hispida Dennst) corm infuse, can decrease blood glucose consentration diabetic aloxan inducted rats.This experiment use 20 rats by randomized design at 2-3 months, 140-280 gram BW, Aloxan inducted diabetic male albino Wistar strain. The dose of aloxan 150 mg/BW given by intraperitonial. The rats divided in 4 groups, each group consist of 5 rats. First group : positif controle which is given insulin 12,6 UI/BW subcutanly; Second group : negative controle which is given aqua dest 5 mL/BW orally; Third group is given Gadung (Dioscorea hispida Dennst) corm infuse 630 mg/BW orally and Forth group is given Gadung (Dioscorea hispida Dennst ) corm infuse 1260 mg/BW dose orally. The experiment results of blood glucose level, was analyzed by 2 way ANOVA (p<0,05).The conclusion of the experiment is the rats has given Gadung (Dioscorea hispida Dennst) corm infuse 630 mg/BW and 1260 mg/BW can decrease blood glucose consentration proporsionally with insulin effect (p<0,05).Key words : Gadung (Dioscorea hispida Dennst), Aloksan, antidiabetic drug.

Abstract

The unbalance of oxidant and antioxidant can effect of various diseases, for example atherosclerosis. High total cholesterol level was given the risk factor to atherosclerosis. This research aims to know the effect of cauliflower (Brassica oleracea var. botrytis L.) juice to change of total cholesterol level and peroxide of lipid (MDA level). A twenty white rats Wistar strain were treated as hiperkolesterolemia with consumed a high fat meal for 14 days, then devided in to 4 groups. Group I as positive control was given a green tea leaf infuse through a dosage 0,605 g/ kg BW, group II as negative control was given an aquadestilata. Then Group III and IV as were given cauliflower juice through a dosage 15 mL/ kg BW and 20 mL/ kg BW for 14 days. The result of this research were concluded that cauliflower juice through a dosage 15 mL/ kg BW and 20 mL/ kg BW was decreased of total cholesterol level and lipid peroxide (MDA level). Cauliflower juice through a dosage 20 mL/kg BW was found as an effective dosage to decreased of total cholesterol level and lipid peroxide (MDA level) of male white rats Wistar strain with high fat diet treatment.Key words : Juice, cauliflower, total cholesterol, peroxide of lipid, MDA.

Abstract

BackgroundIndole and its derivatives, such as indole acetic acid are active constituents of cauliflower (Brassica oleracea var. botrytis L.) with are known to function as plant growth hormones. Drug metabolism mainly occurs in the liver, so that the possibility of damage to this organ is considerable. The purpose of this study was to evaluate the effect of cauliflower extract on alanine transaminase (ALT) levels and on the hepatocyte morphology induced by theophylline in Wistar rats.MethodsTwenty Wistar rats were divided randomly into four groups. Group I was given only theophylline 20 mg/kg BW. Groups II, III, and IV were given cauliflower extract 200 g/kg BW, for 5, 10, 15 days, and on the last day were given theophylline 20 mg/kg BW. On days 5, 10, and 15 the animals were terminated, their livers taken and observed for microscopic appearance, while blood samples were taken for measuring ALT levels.ResultsCauliflower affected a reduction in ALT levels, mean serum ALT concentration in rats given cauliflower for 5 days (109.86 ± 2.20 IU/L was significant higher compared to rats given cauliflower for 15 days (71.58 ± 3.17 IU/L) (p=0.000). Mean values of histopathologic liver cells in rats given cauliflower for 15 days (1.17 ± 0.01) significant lower compared to rats given cauliflower for 5 days (1.45 ± 0.06) (p=0.000).ConclusionSCauliflower extract reduced the level of ALT and improved the microscopic appearance of hepatic cells in the rats. Cauliflower extract could lead to the development of potent anti-tumor agents for hepatic cancer treatment.

Abstract

ROS increase can causes damage to the kidney so that urea and creatinine level increase in the blood and can be used as a marker of decreased kidney function. Excessive free radicals can cause a decrease in endogenous antioxidant activity, namely the catalase enzyme. This condition can be overcome by given exogenous antioxidants such as strawberry juice (Fragaria ananassa Duchessne). This study aimed to determine the effect of strawberry juice treatment against urea and creatinine levels in male Wistar rats Isoniazid-induced for 14 days divided into groups, namely normal control, negative control, positive control, strawberry juice dose of 3 g/kgBW, 6 g/ kgBW, and 9 g/kgBW. Data were collected on 1st, 15th, and 29th day. The results of the study concluded that strawberry juice had an effect in reducing levels of urea, creatinine and catalase enzyme activity in isoniazid-induced rats with an effective dose of 3g / kgBW
