Abstract

ABSTRAK Telah dilakukan penelitian uji antidiabetika infus biji alpokat (Punica granatum L.) terhadap kelinci jantan yang telah dibebani glukosa serta profil KLTnya. Tujuan penelitian ini untuk mengetahui efek antidiabetika infusa biji alpokat dan kandungan kimianya yang terdapat pada infus biji alpokat. Penelitian ini menggunakan 15 ekor kelinci jantan yang dikelompokkan menjadi 5 kelompok. Kelompok 1 (kontrol negatif) diberi perlakuan aquadest, Kelompok 2 (kontrol positif) diberi perlakuan suspensi glibenklamid dalam CMC-Na 0,5%. Kelompok 3, 4, dan 5 diberi perlakuan infus biji alpokat (Punica granatum L.) dengan dosis 0,3289 g/kgBB, 0,6571 g/kgBB, dan 0,9856 g/kgBB. 30 menit kemudian masing-masing kelompok diberi glukosa dengan dosis 1 g/kgBB. Penurunan efek kadar glukosa darah ini menggunakan metode enzimatik. Kadar glukosa darah yang diperoleh dianalisis dengan menggunakan uji ANAVA satu arah dengan taraf kepercayaan 95 % dilanjutkan dengan uji LSD. Hasil penelitian menunjukkan bahwa infus biji alpokat dengan dosis 0,3289 g/kgBB, 0,6571 g/kgBB, dan 0,9856 g/kgBB dapat menurunkan kadar glukosa darah dimana infusa biji alpokat dosis 0,9856 g/kgBB memberikan aktivitas yang lebih tinggi dalam menurunkan kadar glukosa darah. Hasil pengujian KLT menunjukkan bahwa infusa biji alpokat mengandung flavonoid, alkaloid dan saponin. Kata kunci : Biji alpokat, antidiabetika, KLT ABSTRACT A research had been conducted to test the antidiabetic effect of the avocado fructus infusa (Punica granatum L.) to male rabbits preloaded glucose and also TLC profiles. The aim of the research was to know antidiabetic effect of infusa and its active compounds. This study applied at 15 male rabbits which classified into 5 groups. Group 1 as negative control was given aquadest, group 2 as positive control was given glibenclamide in CMC-Na 0,5 % . Groups 3, 4, and 5 were given avocado fructus infusa with dose 0,3285 g/kg body weight, 0,6571 g/kg body weight, and 0,9856 g/kg body weight. After 30 minutes all rabbits within groups were given glucose solution with dose 1 g/kg body weight. This research used enzymatic method to determine glucose blood level. The blood glucose levels were analyzed statistically by one way ANOVA at confidence level at 95 % and then continued with LSD test. The result of this study shows that avocado fructus infusa with dose 0,3285 g/kg body weight, 0,6571 g/kg body weight, and 0,9856 g/kg body weight had hypoglycemic effect and dose 0,9856 g/kg body weight gave higher activity on decreasing of blood glucose level. The result of TLC test shows that avocado fructus infusa contains flavonoid, alkaloid and saponin. Key word: Avocado fructus, Antidiabetic, TLC
