Abstract

Screening of indigenous bacteria from sago waste based on amylolytic and cellulolytic activity was done to obtain bacterial isolate having double activity, i.e. could to hydrolize of starch (amylolytic) and cellulose (cellulolytic). Screening amylolytic and cellulolytic bacteria was done based on amylolytic and cellulolytic activity on agar media. Determination of amylolytic activity on starch agar media was based on the presence of clear zone around the bacterial colony upon flooding with lugol’s iodine solution. Cellulolytic activity was determine based on the presence of clear zone around the bacterial colony on Carboxy methyl cellulose (CMC) agar upon flooding with congo red solution. Presence of a clear zone around the colony indicated starch and cellulose hydrolysis. The diameters of clear zone produced on CMC and starch agar were measured and used as an indication of the amylolytic and cellulolytic activities of the bacteria. The results of the screening based on amylolytic and cellulolytic activity showed that a number of 21 bacterial isolates that having both activities. LCA2 was the bacterial isolate with the highest amylolytic and cellulolytic activity as revealed by the size of clearing zone on both types of agar plates. The diameters of clear zone on starch and CMC agar were 4,98   and 3,65 cm2, respectively. Therefore, LCA2 isolate was bacterial isolate that potent for biconvertion sago hampas into value-added products. Keywords : Bacteria, Amylolytic, Cellulolytic, Sago waste.

Abstract

Abstract. This study investigated a natural acid-base indicator which is extracted from plants in Kupang city. There are 14 plants that potential as a source of natural indicator for acid base titration, i.e, KolUngu (Brassica oleracea Capitata Group), Turi Merah flower (Sesbania grandiflora L. Pers), BelimbingWuluh flower (Averhoa bilimbi L), Kaktus Merah fruit (Opuntia vulgaris Mill), Ruelia flower (Ruellia simplex), Flamboyan flower (Delonix regia), bugenvil flower (Bougainvillea spectabilisWilld.), Bayam Merah leaves (Amaranthus tricolor L.) Jamblang fruit (Syzygiumcumini L.), Murbey fruit (Morus alba L.), Pinang fruit (Areca catechu L.), Sirih fruit (Piper betle L.), Kunyit (Curcuma longa Linn), and Nanas Kerang leaves (Rhoeo discolor). Plants extract shows a sharp color change in acid and base solution. Promising results as a natural indicator also shown in acid base titration which is have similar equivalent point to synthetic indicator. We can use these natural indicators as an alternative to synthetic indicator because they are found to be simple, very useful, cheap, easy to extract, accurate, and eco-friendly. Abstract

The research objective was to determine the effectiveness of INoSIT (Integrated Nature of Science Inquiry with Technology) learning model to improve the scientific literacy competence of class IX2 students of SMPN 4 Kendari. Through the design of one group pretest-posttest which involved three science teachers as model teacher and observers which were carried out in odd semesters of the 2019/2020 school year. Data collection used the observation sheet for the implementation of learning, student activities, science literacy tests and student response questionnaires. The results of the analysis of average n-gain mean on scientific literacy competence were obtained on the indicators explaining scientific phenomena that were in the high category; while indicators evaluate and design scientific inquiry and interpretation of data and scientific evidence are still in the medium category. Furthermore, the results of hypothesis testing show that there is a significant difference between the scientific literacy competencies of students through the pretest and posttest on . In conclusion, the implementation of the INoSIT learning model in science learning is effective in increasing the scientific literacy competence of class IX2 students of SMPN 4 Kendari.
