Abstract

Abstrak. Tujuan dari penelitian ini adalah untuk menentukan struktur molekul senyawa metabolit sekunderdari ekstrak diklorometana kulit batang tumbuhan bakau merah (Rhizophora stylosa). Isolasi dimulai dariekstraksi dengan metode maserasi menggunakan pelarut berturut-urut n-heksana dan diklorometana.Fraksinasi ekstrak diklorometana menggunakan metode kromatografi cair vakum (KCV) yang dimonitormelalui kromatografi lapis tipis (KLT), pemisahan menggunakan metode kromatografi kilat dan pemurnianmenggunakan metode rekristalisasi menghasilkan isolat serbuk tak berwarna seberat 0,0623 gram dengantitik leleh 136-137 oC. Penentuan struktur molekul isolat dengan spektroskopi UV-Vis, IR dan GC-MS. Hasiluji kualitatif dari isolat dengan reagen Liebermann-Burchard menunjukkan warna biru kehijauan yangmengindikasikan bahwa isolat merupakan senyawa steroid. Berdasarkan analisis data spektroskopi, isolatmengandung 3 campuran senyawa steroid, terdiri dari kampesterol, stigmasterol, dan β-sitosterol.Kata-kata kunci: Rhizophora stylosa, ekstrak diklorometana, kampesterol, stigmasterol, β-sitosterol.Abstract. The aims of this research is to determine the molecular structure of secondary metabolites fromdichloromethane extract of red mangrove’s stem bark (Rhizophora stylosa). Isolation was started byextraction using maseration method, successively with n-hexane and dichloromethane. Fractionation ofdichloromethane extract were conducted by vacuum liquid chromatography (VLC) monitored by thin layerchromatography (TLC), separation using flash chromatography method and purification by recrystallizationmethod gained colorless powder isolates with 0,0623 gram in weight with melting point of 136-137 oC. Thedetermine molecular structure of isolate by UV-Vis, IR and GC-MS. The results of qualitative test of isolatewith Liebermann-Burchard reagent showed greenish-blue colour indicating that isolate was steroidcompound. According to spectroscopic analysis, isolate contains 3 mixture compounds of steroid consistingof campesterol, stigmasterol and β-sitosterol.Keywords: Rhizophora stylosa, dichloromethane extract, campesterol, stigmasterol, β-sitosterol
