IN VITRO ANTIOXIDANT ACTIVITY AND GC-MS ANALYSIS OF ETHANOLIC MANGKOKAN LEAVES EXTRACT (Polyscias balfouriana ( Sander ex Andre) L.H.Bailey)

Abstract

Abstrak Daun mangkokan (Polyscias balfouriana (Sander ex Andre) L.H.Bailey) merupakan tanaman hias atau pagar yang banyak tumbuh liar di lingkungan namun belum banyak dimanfaatkan secara maksimal. Penelitian ini dilakukan untuk mengetahui komponen senyawa serta potensi ekstrak etanol daun mangkokan sebagai antioksidan dalam menangkal radikal bebas dengan menggunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl). Tahap penelitian meliputi skrining fitokimia dari kandungan metabolit sekunder, analisis komponen senyawa dengan metode GC-MS serta uji aktivitas antioksidan dengan metode DPPH. Hasil penelitian menunjukkan bahwa ekstrak etanol daun mangkokan positif mengandung tanin, alkaloid, terpenoid, saponin dan flavonoid. Hasil uji analisis GC-MS menunjukkan bahwa kandungan terbanyak daun mangkokan yaitu Hexadecanoic acid dengan luas area 12,28%. Berdasarkan hasil uji dengan penghambatan radikal bebas dengan metode DPPH diperoleh data bahwa ekstrak etanol daun mangkokan memiliki aktivitas antioksidan dengan nilai IC50 sebesar 161,39 ppm. Ekstrak etanol daun mangkokan memiliki aktivitas sebagai antioksidan sehingga sangat potensial untuk dikembangkan dalam pengobatan penyakit yang disebabkan oleh radikal bebas. Kata kunci: antioksidan, ekstrak etanol, daun mangkokan, DPPH, GC-MS Abstract The leaves of mangkokan (Polyscias balfouriana (Sander ex Andre) L.H.Bailey) are ornamental plants or fences that grow wild in the environment but have not been fully utilized. This research was conducted to determine the components of the compound and the potential of mangkokan leaf ethanol extract as an antioxidant in counter free radicals using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. The research stage included phytochemical screening of secondary metabolites, component analysis using the GC-MS method and antioxidant activity testing using the DPPH method. The results showed that the ethanol extract of mangkokan leaves positively contained tannins, alkaloids, terpenoids, saponins and flavonoids. The results of GC-MS analysis showed that the highest content of mangkokan leaves was Hexadecanoic acid with an area of ​​12.28%. Based on the test results with free radical inhibition with the DPPH method, it was found that the ethanol extract of mangkokan leaves had antioxidant activity with an IC50 value of 161,39 ppm. The ethanol extract of mangkokan leaves has antioxidant activity so that it is potential to be developed in the treatment of diseases caused by free radicals.
