Abstract

Catechin content in tea plant is a major character in tea breeding program for a healthy beverage. A tea breeding program for high catechin content maybe will performed by indirect selecting a trait or traits that significantly correlated with high catechin content. This research was performed to identify morphological traits which maybe significantly correlated with high catechin content in tea plant. A randomized complete block design with three replications was used in this experiment using 10 selected clones. The experiment was performed in Gambung Experimental Station ot the Research Institute for Tea and Cinchona, Gambung, West Java. Catechin content, trichoma density of young leaves, weight of p+2, size of mother leaves, chlorophyll content of mother leaf, stomatal density of mother leaf, mother leaf thickness, and mother leaf angle, were measured. The results indicated that stomatal density and mother leaf angle were significantly correlated. Based on path analysis indirect selection for high catechin content could be performed by selecting stomatal density considering to trichoma density, mother leaf angle and chlorophyll content. In addition, indirect selection for high catechin content could be performed by selecting other leaf angle independently without considering other morphological traits.

Abstract

Tea is used in the food, cosmetic, and herbal drug industries. White tea is a kind of tea containing a relatively high polyphenol content compared to other kind of tea, and thus has the potential of being used as a natural antibacterial. The objective of this research was to study the antibacterial activity of of  white tea extract in several variation of  13 % (w/v) concentration of solvents to the activity of gram-positive bacteria (Bacillus cereus) and Gram-negative bacteria (Shigella dysenteriae). Laboratorium experimental method was used in this descriptive study. The antibacterial activity study was comprised of estimating inhibition area and total of bacterial growth. The results revealed that white tea extract inhibited bacterial growth, indicating that white tea extract from the n-hexane, ethyl acetate and 96% ethanol solvents could be developed as an antibacterial material. The diameter of the inhibition area to Bacillus cereus bacteria for n - hexane , ethyl acetate, and  ethanol 13 % (w/ v)  solvents were 0.6 mm, 10.5 mm and 1.75 mm, respectively; while for Shigella dysenteriae bacteria were 0,5 mm, 9,86 mm  and 10,6 mm, respectively. The total growth of Bacillus cereus bacteria for n-hexane, ethyl acetate, and ethanol 13 % (w/v) solvent were 1.3 x 107, 6.0 x 106, and  1.1 x 104 CFU / ml , and of Shigella dysenteriae bacteria were  1.0 x 107,  6.5 x 106 and 4.6 x 106 CFU / ml, respectively.

Abstract

In order to increase the income of tea smallholder and maintain the sustainability of tea, Dewan Teh Indonesia collaborated with Business Watch Indonesia in establishing a local certification scheme that is called as Lestari Tea Certification Program. This certification program has been conducted at some areas since 2011 until 2013 and one of the areas was Campaka sub-district, Cianjur. This research aimed at analysing the sustainability impact of the certification program by questionnaire and interview data collection. Analysing sustainability is conducted by using Mann-Whitney test by comparing the perception of the tea farmers who participated and those who didn’t participate on the certification program. Independent sample t test is also used to test the differentiation of the mean the income and transportation vehicle and communication asset between member of Tea Lestari Certification Program and those who are not. Moreover, logistic regression is used to know which variable more influence the probability for the tea farmers to join the certification program. The output of Mann-Whitney test shows that there is significant difference on the aspect of environment, economy, and social between tea farmers that participated and those who didn’t participate on the certification program. Logistic regression output shows that education level, size of plantation land, and location of village influence the probability of the tea farmers whether to join or not join the Lestari Tea Cerfication Program.

Abstract

Theaflavin (TF) is a product of catechin oxidation that give contribution to golden yellow color, brightness, briskness of black tea and has a potential as natural antioxidant activity. TF has a higher superoxide radical capture rate than EGCG (Epigallocatechin gallate). The study was to synthesize TF components in the form of extracts that have high antioxidants. Materials used in this research is bud+3 leaves from RITC clone (GMB 7). The research were caried out used 15 liters of tea leaves pulp per treatment, fermentation using fermentor with 15 L/min, 20 L/min and 25 L/min airflow treatment, separation of TF from thearubigin (TR) used 2.5% NaHCO3, and isolation of TF from impurity compounds used silica gel column with ethyl acetate/hexane as eluent, and DPPH (1,1-diphenyl-2-picryl hydrazyl) for antioxidant activity. The result showed that TF synthesis of fresh tea leaf catechin at water-air rate of 25 liters/minute could produced TF extract and yield TF purity extract with 1.01% and 39.15% in dry weight respectively after isolation by silica gel column. Antioxidant activity by DPPH from TF extract showed the smallest value of EC50 is 12.16 mg/L at 150 mg dose and its activity increased with the addition of concentration of TF extract.

Abstract

Bio-imunizer contains an active compound of  Chryseobacterium sp. and Bacillus sp. has been developed by PPTK Gambung. This formula has positive effect on the growth of tea plants also potentially increasing resistance of the plant. The purpose of this study was to determine the effects of bacteria in Bio-imunizer to the rhizosphere bacterial communities as well as the consistency of its existence after application on tea plants at the nursery stage. The technique used in this research is Terminal Restriction Fragment Length Polymorphism based on metagenomic and culture dependent approaches. The value of relative abundance, Shannon diversity index, Pielous evenness index, and Simpson dominance index were calculated. Based on the T-RF profiles of rhizosphere bacterial communities show that Chryseobacterium sp. and Bacillus sp. which is the active compound of  Bio-imunizer consistently found in the tea plant rhizosphere. Application of Bio-imunizer can increase the diversity of rhizosphere bacterial community without affecting the communities that already exist.
