Abstract

BACKGROUND: Ultraviolet (UV)B irradiated-skin cells may respond either by activating protective or apoptotic mechanisms. Several treatments has been reported to prevent apoptosis of the skin cell. To date, despite of the cell, secretome of mesenchymal stem cell (MSC) has been progressively explored for various treatment. Current research was conducted to find out the possible effect of MSC's secretome to protect normal cells from the UVB irradiation.METHODS: For preparation of the conditioned media human umbilical cord blood mesenchymal stem cell (CM-hUCB-MSC), hUCB was collected and separated to collect mononuclear cell (MNC). MNC was cultured in MSC medium until passage 4, then checked for MSC biomarkers. Conditioned media was then produced from the cultured MSC. For induction of apoptosis, NIH3T3 cells were pretreated with/without CM-hUCB-MSC, prior to the UVB irradiation for 5-60 minutes. After 24 hours, apoptosis evaluation was conducted with Sub-G1 assay with hypotonic fluorochrome solution using flow cytometer.RESULTS: The average of apoptotic cells irradiated with UVB for 30 minutes was 94.2%. The 30-minutes-UVB irradiation significantly induced apoptosis in NIH3T3 cells (p=0.000). The averages of apoptotic cells by pretreatment of 0, 5, 10 and 20% CM-hUCB-MSC prior to UVB irradiation were 94.2, 46.8, 31.8 and 31.5 %, respectively. The pretreatment of 5, 10 and 20% CM-hUCB-MSC prior to UVB irradiation could significantly decrease the percentage of apoptosis caused by UVB (p=0.001).CONCLUSION: Taken together, CM-hUCB-MSC inhibited UVB-induced apoptosis in NIH3T3 cells significantly, suggesting that CM-hUCB-MSC might be a potential anti-UVB-induced apoptosis factor. Hence, further research should be explored to disclose its specific intracellular mechanism.KEYWORDS: UVB, stem cell, conditioned media, fibroblast, apoptosis, aging, secretome, NIH3T3 cell

Abstract

The Indonesian nation aspires to realize a just and prosperous national life for that it is necessary to understand and implement the values of Pancasila. The current swift current of globalization has eroded the understanding of future generations about the values contained in Pancasila. This problem of understanding has resulted in the abandonment and oblivion of Pancasila. National identity must be rebuilt by actualizing the values of Pancasila so that it can be a guide in responding to various challenges now and in the future. If we do not succeed in re-actualizing the values of Pancasila, then Pancasila will be alienated from the life of the Indonesian nation. The national values contained in Pancasila are religious values, kinship, harmony, democracy and justice. This paper will discuss the implementation of the value of justice in improving the quality of life in society, nation and the state.

Abstract

ABSTRACT Ultraviolet radiation and Visible light (VL) have biological effects on the skin. VL causes erythema in light-skinned individuals and pigmentation in dark-skinned individuals. Broad-spectrum sunscreen protects skin against ultraviolet radiation but does not adequately protect against VL. The purpose of this study was to analyze the biological effects of visible light (VL) and the choice of sunscreen that can protect it. The research method used in this study is a qualitative descriptive method. The type of data used in this study is qualitative data, which is categorized into two types, namely primary data and secondary data. The protection provided by organic (mineral) and inorganic sunscreen filters can prevent skin damage due to ultraviolet radiation, but does not provide protection against VL. Only tinted sunscreens that use formulations of concentrated iron oxide and titanium dioxide can provide protection against VL. Various steps have been taken to provide photoprotection against VL. There are many sunscreens available that combine percentages level of iron oxide and titanium dioxide pigment to suit all skin types. This tinted sunscreen is beneficial for melasma and post-inflammatory hyperpigmentation and photodermatosis induced by VL. Keywords: Iron Oxide, Photoprotection, Tinted Sunscreen, Titanium Dioxide, Ultraviolet Light, Visible Light

Abstract

The development of stem cells services has grown rapidly throughout the world since 2004, as well as in Indonesia. This is because stem cells have unique properties that can differentiate into various cells types, making it promising to repair tissue damaged by various diseases, especially degenerative conditions. The purpose of this study was to obtain an overview of the regulation of stem cells services so that it can be carried out to the public after the enactment of the Health Law Number 36 of 2009 and to obtain an overview of the implementation of the law in stem cells regulation that is not yet comprehensive. The research methodology used is juridical normative, with a statutory approach, a conceptual approach. Also by using a comparative approach with other countries. This study analyzes substantially the regulation of stem cells and/or cells services based on Minister of Health Regulation number 32 of 2018 concerning stem cells and/or cells services and the implementation of laws against violations in their implementation. The regulations that are not yet comprehensive have in the competence of doctors who carry out stem cells services both as standardized therapy and service-based research that has not been certified, as well as the implementation of the law to use laws and regulations that are generally used for cases of non-specific illegal acts that result in prosecution. which is multi-layered. So that it is necessary to regulate stem cells according to the hierarchy of legislation and need to revise Minister of Health Regulation number 32 of 2018 so that it can be implemented safely and effectively and accommodate the orientation of technology development in the stem cells sector in Indonesia.

Abstract

Dry skin is a problem for millions of people and often causes discomfort and even psychological stress. Increasing the water content in the stratum corneum can be done by applying moisturizer regularly and regularly because it will improve lipid levels and hydrate the epidermis. Saccharide isomers (SI) are one of the answers for the development of Glycobiology. The hypothesis of this study is that the addition of 5% SI in the moisturizing formulation further increases skin hydration and can maintain higher skin hydration even after discontinuation of use compared to ordinary moisturizers. The research subjects were 30 women aged 30-45 years who were not menopausal. Randomly divided into control group (15 people) and treatment group (15 people) by double blind. Moisturizer used for 2 weeks in a row, then discontinued use. Measurement of skin hydration was carried out 3 times a week during the use of moisturizer and after discontinuation. The non-invasive measuring instrument used is the Multi Skin Test Center® MC 750 made in Germany. Giving moisturizer in both groups resulted in an increase in skin hydration after 2 weeks of use (p<0.05). After discontinuation of the moisturizer, the four locations showed significant differences in skin hydration (p<0.05). From the results of this study, it can be concluded that the addition of 5% SI in the moisturizing formulation can increase skin hydration higher and can maintain higher skin hydration even after discontinuation of administration compared to ordinary moisturizers.

Abstract

Euthanasia is an act of intentionally not doing something aimed at prolonging someone's life or intentionally doing something to shorten or also end the life of a patient which is carried out to hasten his death, while enabling a good death without unnecessary suffering. Thus, in practice, in a terminal condition, a doctor may or may not perform an act that may result in the death of a patient in accordance with the written request of the patient/family. This dilemmatic condition requires legal certainty in order to uphold justice and humanity as well as legal protection. The problems are 1) How is passive euthanasia applied for terminal patients? 2) How is legal certainty in the application of euthanasia in Indonesia? 3) How is the implementation of euthanasia that meets the community's sense of justice? To answer these problems, a research using normative juridical methods is needed through primary data collection in the form of empirical data from interviews and filling out questionnaires about knowledge and level of agreement on euthanasia. Secondary data in the form of research on primary, secondary and tertiary legal materials. 8The results of the study show that the implementation of regulations in the Act and Ministerial Regulations that lead to the implementation of euthanasia has so far been applied, especially to terminal patients. However, the legal certainty of the act has not been clearly described because there are rules that are not in line. Euthanasia cannot be equated with ordinary homicide. To fulfill justice and humanity as well as legal protection, euthanasia regulations are needed in a harmonious positive law

Abstract

Trisomy 13 is a serious genetic anomaly in the fetus that is one of the causes of abortion as a result of the chromosomal aneuploidy. Trisomy 13 affects roughly 1 in 10,000 to 20,000 live births, and more than 95% of pregnancies end in prenatal death. Due to the higher prevalence of preeclampsia and the danger of maternal death associated with early birth, abnormalities in these infants also have an impact on the mother's health. According to Law No. 36 of 2009 concerning Health, women who eligble for abortions if there are medical reasons to do so. The problems in this paper are 1) What are the ethical problems found in pregnancies with a fetus diagnosed with a genetic disorder due to trisomy 13? 2) What is the procedure for implementing medical abortion provocation that fulfills ethical aspects and complies with applicable regulations. To answer these problems, a research using normative legal research, accompanied by reports of cases of abortion. This normative legal research focuses on ethical and legal aspects of the implementation of provocative medical abortion following statutory regulations. This normative legal research starts with articles and case reports of fetal pregnancies with genetic disorders from the Gatot Soebroto Army Hospital. The results of the study show that consent to termination of pregnancy with medical indications by the patient and approved by the husband/family. T
