Abstract

Background: Blood supplies estimated to be around 2% of the total population1. However, only 1.88 million bags of blood can be met or 40% of the total needs, so there is a 60% lack of blood from the minimum requirement2. Objective: The purpose of this study was to analyze the factors that influence the intention to donate blood regularly at UTD PMI Bantul Regency Yogyakarta using the Theory of Planned Behavior (TPB) approach. Methods: This research was conducted at UTD PMI Bantul Regency, Yogyakarta in September-December 2018. The number of respondents was 35 voluntary blood donors, with sampling techniques using accidental sampling. Results: The results of multiple linear regression analysis show that attitudes, subjective norms, and behavioral control have a significant effect on the intention to donate. While the results of the regression analysis partially, based on a significant level of value, attitudes have a significant influence on one's intention in donating, while subjective norms and behavioral control do not significantly influence one's intentions in donating. Conclusion: The attitude had the most significant influence on the intention to donate regularly on UTD PMI Bantul Regency Yogyakarta using the Theory of Planned Behavior (TPB) approach.

Abstract

Selama proses penyimpanan di bank darah, sel darah merah akan berkurang elastisitasnya, sehingga akan menurunkan kualitas darah dalam menjalankan fungsinya. Vitamin C dan vitamin E adalah antioksidan yang dapat menekan stres oksidatif dan radikal bebas. Tujuan dari penelitian ini untuk membuktikan pengaruh pemberian vitamin C dan vitamin E dalam mempertahankan elastisitas sel darah dalam darah donor yang disimpan di UTD (Unit Tranfusi Darah) dan BDRS (Bank Darah Rumah Sakit). Penelitian dilakukan dengan membuat 4 perlakuan darah donor sebagai berikut: (1) kontrol, (2) perlakuan dengan pemberian vitamin C (10.8 mmol/L), (3) perlakuan dengan pemberian vitamin E (21,34 mg/L), serta (4) perlakuan dengan pemberian kombinasi vitamin C (10.8 mmol/L) dan E (21,34 mg/L). Pengamatan dilakukan pada hari pertama minggu ke 1,2,3 dan 4, penyimpanan darah donor baik pada kelompok perlakuan maupun kontrol. Masing-masing sampel diukur jumlah sel darah merah serta kadar hemolisisnya sebelum dan sesudah sampel dilewatkan pada alat filtrasi. Preparat apus darah dan pengamatan mikroskop dibuat pada saat sebelum dan sesudah filtrasi pada minggu ke-1 dan ke-4. Analisis data dilakukan secara deskriptif. Hasil penelitian menunjukkan bahwa pemberian vitamin C dan E sebagai antioksidan memberikan perlindungan untuk mempertahankan elastisitas sel darah merah selama penyimpanan darah donor. Pemberian kombinasi vitamin C dan E menunjukkan hasil yang lebih baik dari pada hanya pemberian vitamin C atau E saja. Proteksi vitamin C terhadap elastisitas sel darah merah lebih baik dibandingkan vitamin E.

Abstract

Background: Indonesia is a tropical country that is rich in various types of plants that have the potential to be used in the health sector. Basil leaves (Ocimum sanctum L.) is a plant that has antibacterial potential. Antibacterial compounds are compounds that can inhibit bacterial growth. Objective: The objective of this study was to determine the antibacterial activity of basil leaf extract (Ocimum sanctum L.) with varying concentrations of 20%, 40%, 60%, 80% and 100%, and to determine the effective concentration of basil extract which has antibacterial activity against Staphylococcus epidermidis ATCC 12228. Methods: This research was conducted with laboratory experimental research method. Basil leaves ethanol extract was prepared by using maseration extraction method. The method used in the inhibitory test using diffusion of the disc with 5 samples in each treatment group. The sample consisted of treatment groups, ethanol extract of basil leaves with concentration of 20%, 40%, 60%, 80% and 100%. Results: Antibacterial test results showed that basil leaf extracts with various concentrations of 20%, 40%, 60%, 80% and 100% had antibacterial activity against Staphylococcus epidermidis ATCC 12228. The result of this study showed that ethanol extract of basil leaves with 20% concentration had inhibitory power of 10,23 mm, 40% of 10,32 mm, 60% by 10,58 mm, 80% by 13,37 mm, and 100% by 15,83 mm. Conclusion: Basil leaf extract concentrations 100% effective as an antibacterial against Staphylococcus epidermidis ATCC 12228 at 15,83 mm.
