Abstract

The development of analytical UV-Vis spectrophotometric method forthe determination of cholesterol is explained. The assay performed by enzymaticcatalytic reaction ·using cholesterol oxidase (COx) to . oxidise cholesterol into cholestenon and hydrogen peroxide followed by catalytic reduction of hydrogenperoxide with o-dianicidine by using enzyme peroxidase (POx) to produce red colour reduced o-dianicideine that can be measured at ii. 520 nm. The modifiedmethod gave sensitive response to cholesterol, where the detection linearity to cholesterol lies between 0.01-5.0 mM cholesterol, slope 0.158 aulmM cholesterol,and the detection limit of 0~01 mM cholesterol. The method has been applied for thedetermination of cholesterol in traditional food samples, where the concentrations ofcholesterol in food are lies between 0.052-0.799 mM cholestero)

Abstract

The growth of Citrus nobilis Var. Brastepu budding that is free fromCitrus Vein Phloem Degeneration (CVPD) is explained. The method is conductedby taking of bud stick from a selected healthy citrus Brastepu plant and is thengrafted into a sour orange (Citrus auraniium) as mother trees via budding method.The research is conduted in Desa Bukit, Brastagi North Sumatra, Indonesia.Optimum conditions on the budding technique have been obtained to produce goodqualih; seedling of Citrus nobilis Var. Brastepu. Growth characteristics of the plantin various treatment conditions have been studied based on the growth leaves, stem,and branches. Analysis of the DNA using RAPD-PCR method has confirmed thatall new seedling produced in the propagation method are free from CVPD.

Abstract

Community service activities with the title: Utilization of Plastic Waste into Fuel Oil in Namo Bintang Village Deli Serdang Regency was planned in May to November 2019.This activity aim was to convert plastic waste into fuel which has economic value and is environmentally friendly. Community services implementation was by counselling, lectures, explanations of theories and techniques for processing plastic waste which was followed by the Namo Bintang Village Scavenger group, they listened and training to the material delivered by the community service team. Equipment for waste utilization is also very simple hence community groups can make it by themselves. Finally, monitoring and evaluation of the success of work in the field will be carried out by going directly to the field together, monitoring by visiting to find out whether the community can do it themselves.In addition, contact will be made by telephone to overcome the problems that arise.In general, community service activities have been successfully carried out and seen a good cooperative relationship between the Higher Education, especially the Implementation Team, and the community in Namo Bintang Village. Villagers realize the importance of saving the environment and at the same time improving their lives because they can make their fuel.

Abstract

The need for clean water is a challenge in rural areas, especially villages that have not yet been reached by the Regional Water Company (Perusahaan Daerah Air Minum,PDAM). One of the villages experiencing problems with the availability of clean water is Tanjung Selamat Housing Village, Deli Serdang Regency. The people in this village rely solely on ground water that comes from private bore wells. The problem faced is that the water is relatively high in turbidity, and the level of water sterilization is also very low because it contains micro-organisms that cause disease such as dysentery, typhoid diarrhea and so on. An efforts to clean water using citrate have been made but the level of turbidity of the water is still above the threshold. To overcome this problem, appropriate technology is needed for water treatment to produce clean water. The purpose of this activity is to design and implement a technology to process raw water into clean water for public consumption. A design of antimicrobial water purification set with the help of solar power has been provided in Tanjung Selamat Deli Serdang Housing in 2019. The steps taken are surveying the existence of water standards, microbial laboratory testing and acidity test. Low quality raw water has obtained in the target villagewhere the water is high turbidity level, yellowish color, acidity level 6, 23 and contains coliform E.coli bacteria, so it requires processing before it is used as raw material for domestic purposes and drinking water. The application of purifying technology with the help of solar panels is able to convert raw water into clear water, has an acidity of 8.6 and is free of E. coli bacteria. The water produced is also alkaline so it is good for domestic consumption. Water user communities really feel the benefits of appropriate water purification technology developed in community service activities

Abstract

Meristematic shoot tip culture to obtain good quality seedling of citrus (Citrus nobilis Var. Brastepu) free from citrus vein phloem degeneration (CVPD) is explained in this research. The method is conducted by using a diseased plant and is then propagated in optimum conditions. The research is conduted in Tissue Culture Laboratory Department of Biology, University of North Sumatra, Indonesia. Optimum conditions for citrus propagation have been obtained to produce good quality of Citrus seedlings that is free from CVPD. The analysis of the DNA using PCR method has confirmed that the seedlings are healthy and free from CVPD. Genomic analysis using 10 RAPD markers show the planlets regenerated from the cultures are 100% similar with the mother plant.

Abstract

Bakteri pelarut fosfat merupakan bakteri yang mampu melarutkan fosfat yang tidak tersedia menjadi tersedia sehingga dapat diserap oleh tanaman. Studi tentang isolasi dan karakterisasi parsial bakteri pelarut fosfat dari guano gua kampret dan uji kemampuannya dalam meningkatkan pertumbuhan tanaman telah dilakukan. Dua isolat bakteri pelarut fosfat berhasil diisolasi dan keduanya berasal dari kelompok bakteri Gram positif, yaitu ST02 dan ST03. Isolat bakteri diuji pada benih tanaman cabai merah selama 30 hari. Rata-rata pertambahan tinggi tanaman tertinggi diperoleh dari perlakuan bakteri ST03 sebesar 5,96 cm. Rata-rata pertambahan jumlah daun terbanyak diperoleh dari perlakuan kontrol pupuk (TSP) sekitar 24 helai dan diikuti oleh kontrol guano sekitar 6 helai. Rata-rata berat basah tanaman tertinggi diperoleh dari perlakuan bakteri ST03 sebesar 4,71 gram. Rata-rata berat kering tertinggi juga diperoleh dari perlakuan bakteri ST03 sebesar 0,72 gram.   Kata kunci: bakteri pelarut fosfat, cabai, guano, gua kampret

Abstract

Permasalahan sampah pasar tradisional akibat kegiatan jual beli dipasar memberikan dampak negatif terhadap lingkungan. Sampah pasar yang berupa sisa sayur-sayuran, buah-buahan, dan makanan lainnya adalah  bahan yang mudah membusuk dan menghasilkan bau tidak sedap. Masalah ini juga yang dirasakan oleh masyarakat dan pedagang di Pasar Tradisional Setia Kongsi Kecamatan Medan Sunggal, Kota medan. Pemilahan sampah dari sayuran yang diterapkan melalui Kegiatan Pengabdian kepada Masyarakat di pasar ini berupa metode sayur petik yang dikemas menggunakan kotak plastik dan kantong plastik dengan berbagai ukuran. Selain mudah disimpan, sayuran yang dalam keadaan segar dan bersih akan mudah dimasak sehingga menghewat waktu dan tenaga dalam mengolah bahan makanan. Partisipasi aktif dari pedagang sayuran dalam kegiatan pengabdian ini diharapkan  dapat mewujudkan pasar menjadi bersih dan tertata dengan baik. Metode ini juga merupakan salah satu cara mengurangi jumlah sampah dari pasar tradisional ke rumah tangga yang sekaligus dapat mengefisiensikan distribusi sampah dalam hal waktu, tenaga dan biaya.Kata Kunci :Sampah, Pasar Tradisional, Sayuran,dan Pemilahan

Abstract

Kebutuhan akan ketersediaan air bersih menjadi tantangan di pedesaan, terutama desa yang belum dijangkau oleh Perusahaan Daerah Air Minum (PDAM). Salah satu desa yang mengalami masalah ketersediaan air bersih adalah Desa Tanjung Selamat Selamat, Kabupaten deli Serdang. Masyarakat di desa ini hanya mengandalkan air tanah yang berasal dari sumur bor pribadi. Permasalahan yang dihadapi adalah kekeruhan air relatif tinggi, dan tingkat sterilisasi air juga sangat rendah  karena mengandung mikro organisme penyebab penyakit seperti disentri, tipus diare dan sebagainya. Usaha untuk membersihkan air menggunakan asam sitrat sudah dilakukan tetapi tingkat kekeruhan air masih diatas ambang batas. Untuk mengatasi permasalahan ini diperlukan teknologi tepat guna pengolah air  mengahasilkan  air bersih. Tujuan kegiatan ini adalah merancang dan menerapkan teknologi untuk mengolah bahan baku air menjadi air besih untuk konsumsi masyarakat. Pengabdiankepada masyarakat berupa penerapan teknologi  penjernihan air dengan bantuan tenaga surya dan antimikroba dengan bantuan tenaga surya telah dilaksanakan di Perumahan Tanjung Selamat Deli Serdang Tahun 2019. Langkah yang dilakukan adalah survey keberadaan baku air, uji laboratorium mikroba dan uji derajat keasaman. Kualitas baku air sebelum dilaksanakan pengabdian kepada masyarakat tergolong rendah karena memiliki tingkat kekeruhan tinggi, warna kekuningan, derajat keasaman 6, 23 dan mengandung bakteri coliform E.coli, sehingga membutuhkan pengolahan sebelum digunakan menjadi bahan baku air untuk konsumsi domestik dan air minum. Penerapan teknologi tepat guna penjernih dengan bantuan panel surya mampu mengubah air baku yang  menjadi air yang jernih, memilki derajat keasaman 8,6 dan bebas bakteri E.coli. Air yang dihasilkan juga bersifat alkali sehingga baik untuk konsumsi domestik. Abstract

A study on about the utilization of chitinolytic bacterial isolates to inhibit growth of Curvularia sp. causal agent of leaf spot disease of cucumber was done in Laboratory of Pest and Disease, Medan Johor, UPT-Balai Protection Plant and Horticulture I and Laboratory Microbiology Departement of Biology, Faculty of Mathematics and Natural Sciences, University of Sumatera Utara, Medan. Six chitinolytic isolates were tested in vitro to inhibit growth of Curvularia sp.. The result showed that Bacillus sp. BK13 inhibited more with inhibition zone of 2,75 cm and Enterobacter sp. BK15 with inhibition zone of 2,55 cm, whereas Enterobacter sp. PB17 showed the lowest inhibition zone of 1,3 cm. Soaking seed treatment in chitinolytic bacterial suspension reduced the percentage of leaf spot. Enterobacter sp. BK15 showed more ability (50%) to inhibit leaf spot attack.   Keywords: Bacillus sp.
