Abstract

In Vitro Test on the Effect of Some Herbicides Application towards Trichoderma sp. This research’sobjective is to study effect of herbicide ingredient with different concentration to growth of myceliumcolony and spore density of Trichoderma sp. Trichoderma sp was inoculated in PDA which havebeen added with six different herbicide ingredients, there are paraquat, glyphosate, fenoxaprop-ethyl,triclopyr, 2,4D, and fluroxypyr at three different concentrations, there are 100%, 75%, and 50% fromrecommended concentration. This research was conducted from May until October 2013 atPhytopathology Laboratory, Agriculture Faculty, University of Sumatera Utara, by using factorialcompletely randomized design with three replications. The result have showed that highest extensiveof mycelium colonywas observed at fluroxypyr 50% concentration, meanwhile the lowest extensive ofmycelium colony was observed at glyphosate 50% concentration. Highest spore density wasobservedat fluroxypyr 50% concentration , meanwhile the lowest spore density was observed at paraquat 100%concentration. Herbicide ingredient interaction with concentration of herbicide very significantlyaffected growth of mycelium colony and spore density of Trichoderma sp.Keywords: Trichodermasp, herbicide, biological agent

Abstract

Elaedobius kamerunicus adalah serangga penyerbuk kelapa sawit (SPKS) yang efektif untuk meningatkan fruit set kelapa sawit.Penurunan agresivitas kumbang E. kamerunicus diduga karena telah terjadi penurunan kemampuandalam beberapa tahun ini sehingga terjadi penurunan fruit set kelapa sawit. Penelitian ini bertujuan untuk mengetahui hubungan kekerabatan E.kamerunicusdi Sumatera Utara dengan menggunakan 2 primer yaitu LCO1490/HCO2198 dan r1138/f1094. Analisis keragaman menggunakan program MEGA Versi 7. Pohon filogenetik dikontruksi dengan cara ‘Neighbor-Joining Tree’. Hasil amplifikasi sekuen DNA terhadap 35 sampel genom E. kamerunicus dan 3 sampel genom S.oryza sebagai kontrol menunjukkan adanya keragaman yang tinggi. Hasil analisis menunjukkan bahwa E. kamerunicus dengan menggunakan Primer LCO1490/HCO2198 terdiri dari 2 kelompok besar yaitu kelompok 1 E. kamerunicus di Ajamu, Binanga, Bukit Sentang, Padang Madarsah dan PT TPS Sibolga kelompok 2 Marihat dan Bah Birong Ulu. Pada primer r1138/f1094 terdiri dari 2 kelompok besar yaitu kelompok 1 E. kamerunicus di Bah Birong Ulu dan PT TPS Sibolgadan kelompok 2 E.kamerunicus di Marihat, Ajamu, Binangan, Padang Madarsah dan Bukit Sentang.

Abstract

Effectiveness test entomopathogenic nematodes Steinernema sp. at the coffee berry borer pest (Hypothenemus hampei Ferr.) (Coleoptera: Scolytidae) in the laboratory. The aimed of the research was to determine the effect of application of Steinernema sp. to control H. hampei. This research was conducted at the Laboratory of Plant Disease, Faculty of Agriculture, University of Sumatera Utara, Medan at an altitude of ± 25 meters above sea level, from November 2014 to  April 2015. The research used Complete Randomized Design (Non Factorial) with various nematodes level. They were N0 (No nematode/control), N1 (68 JI / ml), N2 (138 JI / ml), N3 (208 JI / ml). The parameters observed were the symptoms of an attack and the percentage of mortality H. hampei. The results showed that the best result to control H. hampei was showed on treatment N3 (208 JI / ml) with a percentage of mortality by 82 % .
