Abstract

Nowadays some efforts are being made to reduce obesity by using drugs that aim to reduce appetite of the drug users. However, the use of drugs still has harmful side effects. Endophytic fungi originating from the Dayak onion plant are thought to have secondary metabolites that have potential as anti-obesity. The aims of this study were to determine the compounds produced by endophytic fungi of Dayak onion and to determine the potential components as inhibitors of pancreatic lipase enzymes. Samples of Dayak onions were collected from Palangka Raya using purposive sampling method. Isolation of endophytic fungi was conducted using surface sterilization method and the isolates were grown on PDA. Isolates were characterized and fermented on PDB for 14 days. The selected isolate was analyzed by using GCMS then molecular docking was performed using Autodock Vina and PyRx. The research results showed that there were 7 isolates of endophytic fungi isolated from leaves and roots tissues of Dayak onion. GCMS analysis showed that EBD1 contains 38 compounds. Molecular docking analysis showed that 1,3-Di-O-Acetylpentopyranose, a test ligand, has the closest binding affinity value (-6.6 kcal/mol) to orlistat (control) (-6.8 kcal/mol). Thus, it has the potential as an inhibitor of pancreatic lipase enzyme protein.

Abstract

Antibiotic resistance is one of the main challenges faced by the health world today. Therefore, various efforts are being made to find new alternative antibiotics from various sources. Endophytic bacteria from medicinal plants are thought to have an antibacterial activity that can potentially inhibit pathogenic bacteria growth. This study aimed to isolate and characterize endophytic bacteria from the Dayak onion plant and determine their potential to inhibit the growth of two pathogenic bacteria Escherichia coli and Staphylococcus aureus. Bacterial isolation was carried out using the surface sterilization method and grown on nutrient agar media. The endophytic bacteria isolates obtained were then characterized macroscopically and microscopically. The agar plug diffusion method carried out the antagonism test against pathogenic bacteria. The result showed that 40 isolates of endophytic bacteria were successfully isolated from the Dayak onion plant's leaves, bulbs, and roots. Based on the antagonism test, 3 isolates were able to inhibit E.coli while 8 isolates were able to inhibit S. aureus. Three isolates (CED3, CED4, and CEU6) were able to inhibit both pathogenic bacteria, and the highest activity was shown by CED3 isolates.

Abstract

Perairan rawa gambut Kalimantan merupakan salah satu habitat bagi keragaman ikan air tawar, dimana keragaman ikan air tawar ini saat ini mengalami berbagai ancaman kepunahan yang disebabkan oleh penyakit dan patogen, polusi perairan, perubahan suhu dan iklim, laju deforestasi, dan degradasi habitat. Inventarisasi dan pengenalan jenis merupakan langkah awal dalam melestarikan keragaman jenis ikan air tawar. Tujuan dari penelitian ini adalah untuk menginventarisasi jenis ikan air tawar yang terdapat di kawasan Universitas Palangka Raya. Penelitian dilakukan selama 1 Februari - 30 April 2021 pada tiga titik pengamatan di Kawasan Sungai Universitas Palangka Raya dengan menggunakan metode penangkapan secara langsung pada lokasi pengamatan. Hasil pengamatan menunjukkan 2574  individu yang terdiri dari 8 famili, 11 genus, dan 15 spesies. Kelimpahan jenis ikan air tawar ditunjukkan pada jenis Pectenocypris korthausae dengan 1114 individu.
