Abstract

Striped catfish (Pangasianodon hypophthalmus) spawning success is strongly influenced by environment.Striped catfish spawning generally occurs during the rainy season while in the dry season is difficult to find mature female. This research was conducted to improve reproduction performance of female striped catfish during dry season through combination injection of hormone pregnant mare serum gonadotrophin (PMSG) and human chorionic gonadotrophin (HCG).The treatments were A : control (no hormone injections), B : injection of PMSG 10 IU/kg + HCG 10 IU/kg and C : injection of PMSG 20 IU/kg + HCG 10 IU/kg.Injections were done at interval of two weeks as many as six times. The research was conducted during dry season in June-October 2012 with rainfall 0.20- 14.40 mm/years. The results showed combination injection of PMSG + HCG could improve female reproduction performance. The best treatment was shown on C treatment with values of OSI (ovi somatic index), fecundity, fertilization rate, hatching rate and larva production was 12.18 %, 523 000 eggs/female, 79.65 %, 69.93 % and 290 000 larvae/female respectively. As for control treatment (A) the value of OSI, fecundity, fertilization rate, hatching rate and larval production, was 6.45 %, 197 000 eggs/female, 48.55 %, 35.59 % and 34 000 larvae/female, respectively

Abstract

Patin nasutus (Pangasius nasutus Bleeker, 1863) (Pangasiidae; Pisces) is a potential pangasiid catfish species to be cultured in Indonesia.Many efforts has been done to explore the potency of P.nasutus for aquaculture purposes.However, basic knowledge is still very rare such as its embryonic and larval development.The present study aimed to understand the embryonic and larval development of P. nasutus resulted from induced spawning of P.nasutus brooders at Research Institute for Fish Breeding, Sukamandi. Those embryonic and larval development were observed microscopically.The results showed that diameter of the ovulated oocytes ranged 1.40-1.65 mm and fertilized eggs ranged 1.90-2.15 mm.The embryogenesis consisted of several stages as follow: morula, blastula, gastrula, organ developments and newly hatched larvae occured within periods of (minutes) 20-200, 190-220, 210-660, 600-1,140 and 1,800-2,160 after fertilization at water temperature of 28-29oC. Total length of the newly hatched larvae ranged 4.80-5.10 mm with anterior, posterior and dorsal parts of the yolk sac coloured black pigmentation. On caudal peduncle, the black spot existed since 36 hours after hatching was a strong character for P.nasutus larvae.Volume of the yolk sac was 2.65±0.14 mm3, for which 50% absorbed at 36 hours after hatching and relative fully absorbed at 78 hours after hatching. The larvae have complete morphology as adult after seven days. During early developmental stage, larvae showed cannibalism since the third day, corresponding to the first exogenous feeding.

Abstract

The increasing demand for striped catfish and pasupati seed will be met through an intensive hatchery with a recirculation system and increased stock density. The purpose of this research was to figure out the effect of stocking density on the performance of growth and survival rate of striped catfish and pasupati in the nursery recirculation system. Rearing was carried out on the fiber with a capacity of 30 liters and equipped with aeration installation and recirculation systems. The study was conducted with three stocking density treatments, namely A) 1 fish L-1 , B) 5 fish L-1 , and C) 10 fish L-1 . We used juveniles of striped catfish and pasupati aged 28 days with an average standard length of 2.14 ± 0.31 cm and 2.22 ± 0.25 cm, and total length of 2.63 ± 0.39 cm and 2.75 ± 0.29 cm, and weight of 0.14 ± 0.06 g and 0.16 ± 0.04 g, respectively. The fish were fed thrice a day at satiation level with floating pellet with 30% crude protein content. The result showed that the optimum stocking density for striped catfish was 5 fish L-1 . During this treatment, the survival rate of of striped catfish was 92.33% with specific growth rate of the weight, standard length and total length of fish for 45 days of rearing were 7.85%, 2.42% and 2.40% , respectively. However, an optimum stocking density for pasupati was 1 fish L-1 . The survival rate of pasupati was 71.67% with specific growth rate of weight, standard length and the total length for 45 days of rearing were 10.78%, 3.35% and 3.32%, respectively. Abstrak Permintaan benih ikan patin siam dan pasupati yang terus meningkat akan dapat terpenuhi melalui pembenihan secara intensif dengan sistem resirkulasi dan peningkatan padat penebaran. Tujuan penelitian ini adalah mempelajari pengaruh padat penebaran terhadap performa pertumbuhan dan tingkat sintasan benih ikan patin siam dan pasupati pada pende- deran sistem resirkulasi. Kegiatan pendederan dilakukan pada bak fiber kapasitas 30 liter yang dilengkapi dengan insta- lasi aerasi dan sistem resirkulasi. Penelitian dilakukan dengan tiga perlakuan padat penebaran, yaitu A) 1 ekor L-1 , B) 5 ekor L-1 , dan C) 10 ekor L-1 . Ikan uji yang digunakan adalah benih ikan patin siam dan pasupati berumur 28 hari dengan rata-rata panjang baku 2,14±0,31 cm dan 2,22±0,25 cm, panjang total 2,63±0,39 cm dan 2,75±0,29 cm, serta bobot 0,14±0,06 g dan 0,16±0,04 g. Pakan diberikan secara at satiation berupa pelet terapung dengan kandungan protein ka- sar 30% dan frekuensi pemberian pakan sebanyak tiga kali sehari. Hasil penelitian pada ikan patin siam menunjukkan bahwa padat penebaran optimum ditunjukkan oleh perlakuan 5 ekor L-1 dengan laju pertumbuhan spesifik terhadap bo- bot, panjang baku, panjang total selama 45 hari pemeliharaan sebesar 7,85%, 2,42% dan 2,40%, dengan tingkat sintasan sebesar 92,33%. Hasil penelitian pada ikan patin pasupati menunjukkan bahwa padat penebaran optimum ditunjukkan oleh perlakuan 1 ekor L-1 dengan laju pertumbuhan spesifik terhadap bobot, panjang baku dan panjang total selama 45 hari pemeliharaan sebesar 10,78%, 3,35% dan 3,32%, dengan tingkat sintasan sebesar 71,67%.

Abstract

Bacterial disease in catfish has many detrimental to farmers, especially in the larvae stadium in the hatchery up to ready size of stocking seed. Therefore we need technology that can produce disease resistant of catfish. In that regard, one of the antimicrobial enzyme namely lysozyme which plays an important role in the maternal immunity can be introduced into the catfish genome. This study aimed to evaluate the success of the lysozyme gene insertion into the genome of striped catfish to generate maternal disease resistance. Gene transfer method was done by using electroporation on striped catfish spermatozoa. Lysozyme plasmid electroporation performed with a dose 100^g ml-1 and 125V.cm-1, pulse length 30 milliseconds with a pulse interval 0.1 milliseconds and pulse number 5 times. Electroporated spermatozoa were used to fertilize the egg. Lysozyme gene insertion success was indicated by tests performed on embryogenesis, larval, and seed stage. The test results at both DNA and RNA level showed a positive result. Individual that brings the lysozyme gene will be used as a candidate for breed line maternal immune as other ways to formation of resistant disease varieties. Abstrak Serangan penyakit bakterial pada ikan patin telah banyak merugikan para pembudi daya ikan patin terutama pada seg-men perbenihan hingga ukuran siap tebar. Oleh karena itu diperlukan teknologi yang mampu menghasilkan ikan patin yang tahan penyakit. Berkaitan dengan hal itu, salah satu enzim antimikroba yaitu lisozim yang memainkan peranan penting dalam imunitas bawaan dapat diintroduksikan ke dalam genom ikan. Penelitian ini bertujuan untuk mengevalu-asi keberhasilan insersi gen lisozim ke dalam genom ikan patin siam sebagai galur ikan patin tahan penyakit. Transfer gen dilakukan dengan menggunakan teknik elektroporasi pada spermatozoa ikan patin siam. Elektroporasi dilakukan dengan konstruksi gen lisozim berupa plasmid DNA dosis 100 ^g.ml-1 dengan voltase 125 V. cm-1, panjang kejutan 30 milidetik dengan interval kejutan 0.1 milidetik dan jumlah kejutan lima kali. Spermatozoa hasil elektroporasi digunakan untuk membuahi telur. Pengujian keberhasilan insersi gen lisozim dilakukan pada tahap embrio larva, dan pada benih. Hasil pengujian, baik pada tingkat DNA maupun pada tingkat RNA dari sampel spermatozoa dan larva (whole cell), memperlihatkan hasil yang positif. Individu ikan patin yang membawa gen lisozim dan telah terintegrasi ke dalam ge-nomnya akan digunakan sebagai kandidat dalam pembentukan galur ikan patin tahan penyakit.

Abstract

Pada segmen pembenihan, ikan patin Pasupati II hasil hibridisasi antara ikan patin Siam betina dan ikan patin Jambal jantan menunjukkan performa terbaik dibandingkan ikan patin Pasupati l, dan patin Siam F-1.Penelitian ini bertujuan untuk menguji performa ikan patin Pasupati II pada segmen pembesaran yang dilakukan di kolam air tenang (KAT) berukuran 50 m2, dan di jaring (berukuran 5 m x 3 m x 1,5 m) yang dipasang di tambak air payau (TAP, salinitas < 10 ppt). Ikan uji yang digunakan adalah ikan patin Pasupati II, Pasupati I, dan patin Siam F-1 dengan bobot awal di KAT 11,1-16,1 g/ekor, dan di TAP 21,3-32,5 g/ekor. Sebanyak dua KAT, dan dua jaring di TAP digunakan untuk setiap kelompok ikan. Parameter yang diamati meliputi: pertambahan bobot dan panjang harian, konversi pakan, sintasan, kualitas air pemeliharaan, dan konsentrasi hormon Insuline-like Growth Factor (IGF-I) pada plasma darah. Hasil penelitian menunjukkan bahwa performa ikan patin Siam F-1 pada parameter pertambahan bobot memberikan yang terbaik (P<0,05) dibandingkan patin Pasupati I dan II yang dipelihara di KAT dan di TAP. Kemudian performa pertambahan bobot ikan patin Pasupati II lebih baik (P<0,05) daripada patin Pasupati I yang dipelihara di TAP. Hasil analisis ELISA pada beberapa ikan uji yang dipelihara di KAT menunjukkan bahwa konsentrasi hormon IGF- 1 tertinggi terdapat pada ikan patin siam F-1 (4,48 ± 0,81 ng/mL), kemudian diikuti oleh patin Pasupati II (3,96 ± 0,51 ng/mL); dan terendah pada ikan patin Pasupati I (3,93 ± 0,54 ng/mL). Jika dicermati dari data pertumbuhan dan konsentrasi hormon IGF-1 ikan uji ternyata terdapat korelasi yang positif antara pertumbuhan ikan dengan konsentrasi hormon IGF-1, semakin tinggi tingkat pertumbuhan ikan maka semakin tinggi konsentrasi hormon IGF-1 pada ikan uji.In thenursery, Pasupati II catfish hybrid of female Siamese catfish and male Jambal catfish showed the best performance compared to Pasupati I catfish and Siam catfish F-1 generation. The aim of this study was to test performance of Pasupati II on grow out segment in freshwater pond (KAT) measure 50 m2 and in net cage (measure 3 m x 5 m x 1.5 m) which settled in brackishwater pond (TAP, salinity < 10 ppt). The fish that used were Pasupati II, Pasupati I, and Siam catfish F-1 with body weight of 11.1-16.1 g/fish in KAT, and 21.3-32.5 g/fish in TAP. A total of two KAT, and two TAP were used for each group of fish. Parameters observed were included daily body weight and length, feed conversion ratio (FCR), survival rate, water quality, and concentration of insulin-like growth factor (IGF-1) hormone on plasma. The results showed that the performance of Siamese catfish F-1 growth parameters give the best weight gain (P<0.05) than catfish Pasupati I and II were maintained at KAT and TAP. Then the weight gain performance catfish Pasupati II was better (P<0.05) than that of catfish Pasupati I reared in TAP. Results of ELISA analysis on some of the fish that are reared in the KAT were showed that the concentration of the hormone IGF-1 was highest in F-1 Siamese catfish (4.48 ± 0.81 ng/mL), followed by Pasupati catfish II (3.96 ± 0.51 ng/mL) and the lowest in the Pasupati catfish I (3.93 ± 0.54 ng/mL). When the data of growth and IGF-1 hormone concentrations in the tested fish was examined there was a positive correlation, the higher the growth rate of the fish followed the higher concentration of the IGF-1 hormone in the test fish.
