Abstract

Fetal bovine serum (FBS) is a gold standard as a supplement to cell and tissue culture media. This is due to a large number of Growth Factor (GF) contained in FBS. However, the use of FBS is at risk of transferring endotoxins, prions, bacteria and viruses from animals to humans, so it is risky to be used on cell therapy. Human Platelet Lysate (HPL) is a medium that can be developed as an alternative cell growth medium. The advantage of HPL is that it does not contain aggregate platelets so it does not cause the cells to clot. This condition causes HPL to be used as a substitute medium replacing FBS for cell propagation. The use of HPL for cell propagation has been widely reported. However, the use of HPL in cancer cells has not been found. Thus, this study aims to see the effectiveness of HPL as a T47D cell culture medium. The study began with donor selection with criteria for the male sex, the blood type O, the age ≤35 years. Furthermore, the Platelet Concentrate (PC) was processed into HPL then measured pH, total protein and albumin levels. The cell viability was measured using the MTT assay to determine the ability of cell proliferation when propagation using HPL. The doubling time test was carried out as in the cell proliferation test. However, the incubation was carried out for 24 h, 48 h and 72 h and the HPL concentration used was 5%. The result shows that HPL 10% and 20% ability to increase proliferation better than the FBS 10%. HPL with a 5% concentration ability to shortens the doubling time than FBS 10% (doubling time is less than 19.94 h). It this study, cell proliferation is influenced by the pH of HPL and total protein but not by the amount albumin.Keywords: Human Platelet Lysate, Proliferation, T47D cell line, total protein, albumin.

Abstract

Background: Blood donation is a process when people have their blood drawn and must fulfill the donation criteria, includes having minimum Hemoglobin levels of 12 g/dL. Low levels of hemoglobin often make unsuccessful blood donation selection for a blood donor. The levels of hemoglobin could be affected by some factors, such as tea-drinking habits from blood donors due to tannin content in the tea can inhibit the iron absorption to the body. Purpose: The objective of this study was to find the effect of tea-drinking habits among inhabitants of Dusun Kopeng, Desa Kepuharjo as a prospective blood donor.Methods: This research was conducted by cross-sectional design to discover the effect of tea-drinking habits on hemoglobin levels. As many as 46 respondents had observed this research with the inclusion criteria, such as tea-drinking habit, productive age (17-45 years old), body weight (45-65 kgs), while the exclusion criteria including older adults, menstruation and pregnant women.Results: Based on a simple linear regression test, the result showed that there was an effect of tea-drinking habit on hemoglobin levels among prospective donors in Dusun Kopeng, Desa Kepuharjo with the significance value of 0,000 0,0005. Conclusion: The tea-drinking habit among inhabitants of Dusun Kopeng affected hemoglobin levels and also impacted the low levels of hemoglobin, which can lead to unsuccessful donor criteria. People could improve the levels of hemoglobin by consuming foods with balanced nutrition, resting well, exchanging the tea-drinking habits with adequate water consumption.Latar Belakang: Donor darah merupakan proses pengambilan darah dari seseorang yang disebut dengan pendonor yang telah memenuhi kriteria donor, salah satu dari kriteria donor yaitu memiliki kadar hemoglobin minimal 12 g/dL. Calon pendonor seringkali gagal dalam seleksi donor disebabkan karena kadar hemoglobin yang rendah. Kadar hemoglobin dipengaruhi oleh beberapa factor, salah satunya adalah kebiasaan konsumsi teh karena dalam teh terkandung senyawa tannin yang dapat menghambat penyerapan zat besi. Tujuan: Penelitian ini bertujuan untuk mengetahui pengaruh kebiasaan minum teh pada masyarakat di Dusun Kopeng, Desa Kepuharjo terhadap Kadar Hemoglobin masysarakat yang merupakan calon pendonor. Metode: Penelitian dilakukan dengan desain cross sectional untuk mencari pengaruh kebiasaan minum teh terhadap kadar hemoglobin. Responden dalam penelitian sebanyak 46 orang dengan kriteria inklusi adalah warga yang memiliki kebiasaan minum teh, dalam usia produktif (17-45 tahun), berat badan 45-65 kg sedangkan untuk kriteria eksklusi yaitu warga yang sedang menstruasi, hamil, dan usia lanjut. Hasil: Berdasarkan hasil uji regresi linier sederhana dapat disimpulkan bahwa terdapat pengaruh kebiasaan minum teh terhadap kadar hemoglobin calon pendonor di Dusun Kopeng, Desa Kepuharjo dengan nilai signifikansi 0,000 0,005. Simpulan: Kebiasaan minum teh pada warga Dusun Kopeng berpengaruh terhadap kadar hemoglobin yang berdampak pada rendahnya kadar hemoglobin, sehingga tidak memenuhi kriteria pendonor. Kadar hemoglobin dapat ditingkatkan dengan mengurangi konsumsi kebiasaan konsumsi teh dengan mengganti teh dengan air putih.
