Abstract

Malariais one of theinfectious diseaseis stillaproblem ofthe world withhigh mortality. Therapeutic purpose of uncomplicated malariais to eliminateplasmodiumcause infection to preventinfectionseverity, complications andbreak the chain oftransmission. While the purpose therapyof severemalaria is to prevent mortality.Recommendedtherapy of malaria isa combination of twoormore antimalarial drugsthat mechanisms actionkillsmalarialparasitesin thebloodand theamount of each drugworks ondifferent receptors. The use ofa combination ofseveralantimalarial drugshas becomea necessityforprevention ofmalariaparasitestrainsthat are resistantto certain drugs. Abstract

S. oleosa belongs to the Sapindaceae family and has phytochemicals phenolic acid so that it has enormous benefits in the process antioxidant. The presence of antioxidant activity in cells can affect the defense of the cell protein membrane, so that the ability of cells to perform division is optimal. In vitro this is mostly done by adding hormones to cell culture media or natural compounds to increase cell proliferation. Every population of cell that has undergone division during the proliferation process to confluent is an indication that the cell is aging. This study was an experimental study was designed using a completely randomized design (CRD) with 5 treatments and 5 replications as follows: K0: granulose cells and culture media without treatment (0 µl), P1: granulose cells and culture media with 1% kesambi leaf extract. (30 µl), P2: granulose cells and culture media given kesambi leaf extract 1.5% (45 µl), P3: granulose cells and culture media given kesambi leaf extract 2% (60 µl) and P4: granulosa cells and The culture media was given 2.5% (75 µl) to the leaves extract., the characteristics of goat ovarian granulosa cells by giving kesambi (Scheichera oleosa) leaf extract at different passages were at the level of confluence showing the highest value in passage I, namely treatment P1. Meanwhile, the level of viability shows the highest value in section I, namely the P4 treatment

Abstract

Kurkumin (diferuloylmethane (C21H20O6) merupakan komponen berwarna kuning–orange yang terkandung dalam tanaman Curcuma longa.  Secara tradisional masyarakat Indonesia  memanfatkan bagian rimpang  tanaman tersebut sebagai salah satu komponen bumbu masakkan sehari-hari dan sebagai komponen jamu dari berbagai kondisi dan penyakit seperti  panas, hepatitis, jerawat, infekasi bakteri, inflamasi, scabies dll. Studi in vivo dan in vitro menunjukkan bahwa kurkumin mempunyai berbagai aktivitas farmakologis  antara lain antiinflamasi, antiviral, antibakteri, antifungal,antikanker, antioksidan, antidiabetes mellitus, antirheumatoid arthritis. Sedangkan studi klinik juga telah banyak dilakukan diantaranya pada kanker pancreas, kanker kolon, kanker payudara, dan multiple myeloma. Pada makalah ini penulis akan membahas tentang etnomedisin kunyit (Curcuma longa), studi pre-klinik baik secara in vitro maupun in vivo, studi klinik pada pasien dan mekanisme kerja senyawa kurkumin sebagai agen kemopreventif dan kemoterapi penyakit kanker. Kata Kunci :Kurkumin. Abstract

Calotropis gigantea radix is one of plant which has bioactive component as antimalarial.The purpose of this research are to know antimalarial activity from combination of Calotropis gigantea radix extract and artemisin. The research consist of extraction of Calotropis gigantea radix was done with extraction maseration method. Extraction was performed by maseration with  80% ethanol solvent. Concentrated extract was in vivo antimalarial tested to animal model. Mice were peritoneal infected with 106Plasmodium berghei ANKA and divided into 5 treatment groups: negative control; positive control (artemisin of dose 4 mg/kgBW); combination of artemisin(1/4 dose of artemisin standar) and Calotropis giganteain 3 doses: 0,1 mg/KgBW; 1 mg/KgBWand 10 mg/KgBW. Data of inhibition was analyzed using SPSS program with Two Way ANOVA Test dan continued with Tukey Test.The result shows that combination of Calotropis gigantea radix extract and artemisin with dose 10 mg/KgBB has higher antimalarial activity than control positive (artemisin). The value of parasite inhibition is 55,2% for dose 0.1 mg/KgBW ; 72,8% for dose 1 mg/KgBW ; 87,3% for dose 10 mg/KgBW and 56,06 mg/KgBW for control positive (artemisin). The phytochemical compounds in 80%  ethanol solvent extract are tannins and steroids. Keywords: Calotropis gigantea,antimalaria, combination, artemisin, Plasmodium berghei

Abstract

Tanaman Widuri mengandung beberapa golongan senyawa aktif, diantaranya flavonoid, alkaloid, terpenoid dan steroid.Senyawa-senyawa tersebut telah terbukti bermanfaat sebagai antimalaria.Selain itu, daun Calotropis proceraberpotensi antimalaria, sehingga diduga akar Calotropis gigantea juga berpotensi sebagai antimalaria karena mempunyai kekerabatan dalam satu genus.Penelitian ini bertujuan untuk mengetahui  efektivitas antimalaria ekstrak etanol 80 % akar Widuri (Calotropis gigantea) pada mencit terinfeksi Plasmodium berghei dan identifikasi golongan senyawa aktifnya.Tahapan penelitian ini meliputi analisis kadar air sampel basah, preparasi sampel, analisis kadar air sampel kering, kemudian ekstraksi senyawa aktif dengan maserasi. Setelah itu, dilakukan uji antimalaria dan dilanjutkan dengan uji fitokimia.Senyawa aktif yang positif terhadap uji fitokimia diidentifikasi dengan metode Kromatografi Lapis Tipis.Hasil kadar air sampel basah sebesar 51,17 %, sedangkan kadar air sampel keringnya sebesar 4,96 %. Isolasi senyawa aktif dilakukan dengan metode maserasi menggunakan pelarut etanol 80 %. Efektivitas antimalaria ekstrak etanol 80 % akar tanaman Widuri (Calotropis gigantea) dalam menghambat pertumbuhan Plasmodium berghei pada mencit adalah sangat baik dengan nilai ED50 sebesar 4,26 mg/Kg BB. Golongan senyawa aktif  yang terkandung dalam ekstrak etanol 80 % akar Widuri (Calotropis gigantea) berdasarkan uji reagen adalah golongan senyawa terpenoid dan saponin. Hasil identifikasi menggunakan Kromatografi Lapis Tipis menunjukkan bahwa eluen terbaik untuk golongan senyawa terpenoid adalah n-heksana : etil asetat (2 : 8) yang menghasilkan penampakan warna ungu pada panjang gelombang 366 nm dengan pereaksi Lieberman-Burchard, sedangkan eluen terbaik untuk saponin adalah kloroform : aseton (4 : 1) dengan penampakan warna ungu pada panjang gelombang 366 nm dengan pereaksi H2SO4 0,1 M. Kata kunci :Calotropis gigantea, antimalarial, Fitokimia, Kromatografi Lapis Tipis
