Abstract

Cephalopods ink has shown potential antiretroviral activity. The ink extracts of cuttlefish showed an antibacterial effect. This study aims to investigate the antibacterial activity of the methanolic extract of the ink of cuttlefish Sepia sp. against Aeromonas hydrophila. A. hydrophila are opportunists and associated with aquatic fish and shrimp disease. The shade-dried ink sample from approximately 30g ink sacs obtained from 15 animals was immersed separately in methanol (1:3 w/v) solvents for overnight. The experiment in this study used the dried extract of cuttlefish ink. The isolate of A. hydrophila was originated from Jepara Brackishwater Aquaculture Center. The average yield percentage of cuttlefish ink extract obtained was 4.86%. The results of the MIC test in Table V show that the highest average absorbance value obtained was at a concentration of 50 ppm, which was equal to 1.716 nm, and the lowest absorbance was obtained at a treatment dose of 300 ppm at 0.841 nm, while the Mc Farland tube was 0.933 nm. The results of antibacterial test on Table II showed antibacterial activity of cuttlefish ink extract at negative concentration control showed diameter zone of 5 ± 1.2 mm, at positive control showed diameter zone of 31 ± 1.2 mm, at 250 ppm result 19 ± 0.9 mm, at 300 ppm result 22 ± 1.4 mm, at 350 ppm result 31 ± 1.2 mm.

Abstract

Lemna sp. is known to have several bioactive compounds and polysaccharide macromolecules that can function as immunomodulators to affect non-specific immune responses to increase the body's resistance to pathogens. This study aims to determine the potential of catfish eye extract as an immunostimulant by observing non-specific tilapia immune parameters. The extraction method used was 96% ethanol maceration for 2 days with a ratio of 1: 4. The experimental design used a Completely Randomized Design with 5 treatments (doses 0.2, 0.4, 0.6 mg/kg, control + and control -) and 3 replications. The non-specific parameters of immunity observed included total plasma protein (Bradford method), superoxide dismutase and lysozyme activity. The data obtained will be analyzed using ANOVA, if there is a significant difference, it will be further tested with Duncan Multiple. Range Test (DMRT). The results showed that the highest total plasma protein was found in treatment C (giving an extract of 0.3 mg/kg body weight) with an average total plasma protein after 12 days of maintenance of 4.99 g / dL. The extract dose of 0.3 mg/body weight showed a rapid decrease in SOD and increase Lysozyme activity.

Abstract

Mystacoleucus marginatus is a type of freshwater fish that is spread in various countries with tropical climates, one of which is Indonesia. This fish can be found in the Brantas River Basin (DAS), East Java. In its development, identification of fish species needs to be done in detail and more deeply, so that the correct identification results can be obtained. This study aims to identify genetic and phylogenetic characteristics of Mystacoleucus marginatus in the Brantas River Basin based on the Cytochrome Oxidase C Subunit I (COI) gene. Test samples were taken from 11 individuals of Mystacoleucus marginatus in the Brantas watershed, from June to August 2018. The results showed that Mystacoleucus marginatus in the Brantas watershed had 2 Haplotypes with Haplotype (Hd)=0.96 diversity values and nucleotides diversity (Pi)=0.00357. These results indicate that the population in the Brantas watershed is interconnected between one location and another sampling location, this is supported by a phylogenetic topology of DNA barcodes showing a monophyletic clade, as well as a haplotype distribution indicating that the sample area is genetics are not isolated from one another. So that the type of Mystacoleucus marginatus scattered along the Brantas watershed is still in one species, which is indicated by the low level of genetic variation. Genetic identification efforts must be carried out continuously so that appropriate information can be obtained for the diversity of fish species, especially endemic fish.

Abstract

Komunitas perifiton sungai telah merespons kondisi lingkungan. Mereka berguna dalam menggambarkan keadaan ekologis, kinerja, dan keberlanjutan ekosistem karena kemampuan untuk mengukur berbagai parameter lingkungan dan menghubungkannya dengan keanekaragaman, kemerataan, dan kekayaan. Studi ini menyelidiki keanekaragaman perifiton Sungai Bango di Malang dan karenanya menghasilkan daftar periksa kekayaan perifiton sungai, terutama perifiton epilitik. Pengambilan sampel perifiton dan identifikasi dilakukan di Laboratorium Hidrobiologi Fakultas Perikanan dan Ilmu Kelautan Universitas Brawijaya Malang. Penelitian ini dilakukan seminggu sekali dalam sebulan (Mei 2020) antara jam 9 dan 13 (GMT + 7). Identifikasi dan sampel Perifiton dikumpulkan dengan cara mengikis substrat (batuan/batuan) di dalam dan di sekitar titik pengambilan sampel air masing-masing. Hasil penelitian menunjukkan kelimpahan total perifiton berkisar antara 938.900 Ind/mm2 sampai dengan 1.598.125 Ind/mm2 dengan 30 genera yang ditemukan. Kami berasumsi bahwa titik sampling 1 sampai 3 memiliki indeks keanekaragaman sedang; Sebaran jumlah individu tiap spesies tergolong sedang; stabilitas komunitas sedang dan organisme akuatik dapat mentolerir kondisi lingkungan. Namun, titik pengambilan sampel 4 memiliki keragaman yang rendah; sebaran jumlah masing-masing spesies rendah; stabilitas komunitasnya rendah dan hanya dapat ditoleransi oleh organisme akuatik tertentu.

Abstract

The pathogenic bacterial infection is one of the problems in the cultivation of vannamei shrimp (Litopenaeus vannamei), causing a high mortality rate of cultured shrimp. The use of antibiotics or chemicals with inappropriate concentrations can harm the aquatic environment, cause resistance, and endanger consumer health because the residues from the chemicals used will periodically accumulate in the body of shrimp. One way to control and prevent shrimp disease is to increase the shrimp immune system by using immunostimulants from seaweed. This study aims to analyze the immunostimulant activity of seaweed extract (Gracilaria sp. and Padina sp.) against vannamei shrimp (L. vannamei) infected with Vibrio harveyi by observing the nonspecific immune system based on its hematological features, namely by counting the number of hemocytes and phagocytic activity. The research was conducted at the Hatchery Unit, Brackish Water Cultivation Development Center (BPAP) Situbondo, East Java. Seaweed sample Gracilaria sp. and Padina sp. purchased from seaweed farmers in Jepara, Central Java. The result of this study shows that supplementation of Gracilaria sp. and Padina sp. at a dose of 10 g/kg of feed can increase the total number of hemocytes and phagocytosis activity of L. vannamei shrimp. The best treatment is Gracilaria sp.

Abstract

Clarias sp. is one of major genera of freshwater catfish and also common commodity in tropical aquaculture. Excessive light intensity is expected to be responded to by fish as environmental stressors. Fish that experience stress will increase the secretion of catecholamine, cortisol, blood glucose levels and p38 MAPKs in catfish. The different intensity of lights will affect the fish physiological processes. The purpose of this study was to analyze physiological responses (Level of blood glucose, primary stress hormone (Cortisol) and p38 MAPKs) catfish Clarias sp. which were incubated at different lights intensity of catfish. 300 fish were collected from Kepanjen Regency, East Java. Blood and meat sampling are carried out to test the profile of glucose, cortisol, and p38 MAPKs. The experimental research at this stage were carried out by transferred the catfish from Kepanjen to experimental aquarium (50 x 30 x 30 cm) that setted the different lights intensity (0 lux (control), 400 lux, 800 lux and 1200 lux). Each experimental aquarium is filled with 15 catfish and will be kept it for 21 days. Quantificationof the parameter using ELISA method. The study result showed highest peak of primary stress hormone (cortisol) levels are 34±1,50 ng/mL, blood glucose levels are 120±3,4 mg/dL, and p38 MAPK are 97±1,1 %. All of it were achieved in the treatment of 1200 lux on the 2nd week.

Abstract

Pesticides are widely applied in the agriculture sector to protect crops and pest control. The poisonous substance of pesticides will affect all of the organisms, either target and non-target organisms. Fish can play the role of an indicator of genotoxic presence in aquatic environments. Polysaccharide extracts from sargassum have promising anti-genotoxic potential. This study aimed to analyze the anti-genotoxic activity of brown seaweed (Sargassum polycystum) methanol extract against erythrocyte and micronuclei of tilapia exposed by methomyl-base pesticide. Brown seaweed (Sargassum sp.) purchased from farmers in Sumenep Regency, Madura, East Java, then macerated using methanol 1: 3 (w / v) for 3x24 hours at room temperature. The phytochemical screening was including flavonoids, alkaloids, triterpenes, saponins, and tannins. Tilapia fish (TL ± 9-12cm) purchased from the Technical Application Unit of Freshwater Fish (UPT Perikanan Air Tawar), Sumberpasir, Malang, East Java. The result of this study showed that exposure of methomyl-based pesticides in the concentration of 4.015 ppm indicates the formation of micronuclei of 318.33 ‰. The increased concentration of extract treatment is directly proportional to the decrease of micronuclei. It means that sargassum extract can reduce the genotoxic effect on exposed tilapia by methomyl-based pesticides. The best concentration of Sargassum sp. extract that can reduce genotoxic was D (200 ppm). Keywords: Antigenotoxic, Extract, Methomyl, Pesticides, Sargassum sp., Tilapia.

Abstract

Study aims to analyze the genotoxic potential of glyphosate-based herbicide on common carp (Cyprinus carpio L) using Micronucleus and hematological assay. The concentration of Glyphosate-based herbicide in this study was 0 ppm, 1.35 ppm, 1.8 ppm, 2.4 ppm, 3.2 ppm, 4.2 ppm, 6.5 ppm, and 8.7 ppm. Administration of herbicide base on modification of published methods with 96 hours of exposure. This research result has shown that the average number of micronuclei was increased simultaneously with increasing the concentration of herbicide exposure. There are also other types of cell nucleus abnormalities, namely: blebbed, lobed, notched, and binuclear. In the treatment of 0 ppm of herbicide shown blebbed nuclei are 8 ‰, lobed nuclei are 6.6 ‰, notched nuclei are 10 ‰, binuclei is 4 ‰. From the research results, it can be concluded that the LC50-96 hours exposure of the herbicide isopropylamine glyphosate in carp (Cyprinus carpio L.) was obtained at a concentration of 8.57 ppm. Based on the evaluation on hematology, it was found that there was a decrease in the number of erythrocytes, hematocrit, and hemoglobin of fish blood, whereas the number of leukocytes, micronuclei, and other abnormal micronuclei showed an increase along with the increase in the dose of exposure to the herbicide isopropylamine glyphosate, which indicates a genotoxic effect.

Abstract

Nuggets are one of the results of fishery diversification as a practical and fast food. The aim of this study to determine the effect of adding carrot flour (Daucus carota L) on the nutritional value and organoleptic of snakehead fish nuggets (Channa striata). The research method used a completely randomized design (CRD) with 4 treatments and 3 replications. The best test results for the nutritional value of snakehead fish nuggets were the Addition of 75 grams of carrot flour to a weight of 500 grams of snakehead fish meat which had a water content of 33.36%, protein content of 25.30%, ash content of 2.45%, fat content of 19.09% and carbohydrate content of 19.79%. The best organoleptic test results were the Addition of 75 grams of carrot flour to a weight of 500 grams of snakehead fish meat based on preference and willingness on the average nugget taste of 5.8, average nugget color 5.8, average nugget aroma 5.9 and average nugget texture 5.9 . The best result of the nugget effectiveness index test was the Addition of 75% carrot flour to the weight of 500 grams of snakehead fish meat with an effectiveness index value of 0.57.
