Abstract

This study aims to reveal the phenomenon of presence / absence of anomalies in the early development of Mimi-giant mintuno (Tachypleus gigas) during artificial incubation in the vial bottles. Samples 5 eggs are fertilized incorporated into transparent 50 bottles and vials each filled with clear sea water medium. Embryonic stages (instars) hatch, bottles marked, then dumped seawater medium was replaced with 4% formalin solution and glycerin amount of 5% by volume, up to ¾ of the total volume of the vial bottles. Standard stages of giant Mimi-mintuno embryonic normal development Mimi-mintuno according to Itow (1988). The description type of anomalies contained in the post-hatching embryo development. The observations are documented in the form of stereo-microphotograph. The results showed that there are forms anomalies: (a). Delayed development, the structure of the body is not perfect; (b). In observation of the embryo hatches, open shell and egg perivitelline membrane has been opened but delayed development, the structure of the body is not perfect; (c). Embryos after hatched perfectly, abnormalities of morphologic structure such as abnormal protrusion on the dorsal carapace part found. Keywords: Tachypleus gigas, artificially, incubation

Abstract

Abstrak Penelitian ini bertujuan untuk mengetahui hubungan antara kandungan Hg dan kualitas air kolam fakultatif 1 dengan bioakumulasi Hg pada ginjal ikan tombro yang dipelihara di kolam fakultatif 1 IPAL Sewon, serta hubungan bioakumulasi Hg terhadap struktur mikroanatomik ginjal ikan tombro tersebut. Populasi penelitian adalah ikan tombro yang dipelihara di kolam fakultatif 1 IPAL Sewon dan kolam kontrol di animal house FMIPA UNY. Sampel ikan tombro dari masing-masing kolam sebanyak 40 ekor untuk analisis kandungan Hg ginjal dan pembuatan preparat mikroanatomiknya. Penelitian ini dilakukan pada bulan Mei sampai Juni 2012. Variabel bebasnya adalah kandungan Hg, DO, COD, turbiditas, suhu dan pH air. Variabel tergayut adalah kandungan Hg pada ginjal dan struktur mikroanatomik ginjal ikan tombro uji. Data dianalisis dengan regresi ganda. Hasil pengukuran kandungan Hg pada air kolam fakultatif 1 IPAL Sewon adalah 0,457 ppb, pada ginjal 50,273 ppb, sedangkan pada kolam kontrol dan ikan kontrol tidak terdeteksi. Hasil analisis regresi menunjukan bahwa konsentrasi logam berat Hg pada ginjal ikan tombro (Y) dipengaruhi oleh kandungan Hg, pH, BOD, COD, DO, temperatur, dan turbiditas dengan persamaan garis regresi: Y = 0.001 + 87.530X₁ + 8.339X₂ + 1.592X₃ - 2.299X₄ + 0.000X₅ + 0.002X₆. Pengamatan struktur mikroanatomik ginjal ikan tombro menunjukkan adanya kerusakan sel pada kapiler glomerolus, piknosis, sel tubulus mengalami lisis. Kata kunci: Bioakumulasi, Hg, mikroanatomi, ginjal, ikan tombro, fakultatif 1 IPAL.

Abstract

Abstract Meniran (Phyllanthus niruri, L.) known as a beneficial rich plant. This research aimed to see its influence to size measured and histological structure of male mouse prostate gland (Rattus norvegicus, L.). Adult male white rat, 24 heads, age ±2,5 months, divided to become six treatment groups covering: A (control), B (placebo), C (0,7 mg/200g bw), D (0,9 mg/200g bw), E (1,8 mg/200g bw) and F (2,7 mg/200g bw). Treatment of herb meniran extract per oral counted 2,0 ml/day during 49 days (one cycle of rat spermatogenesis). Research conducted in LP3HP UGM, animal house, and histology laboratory of Biology Education Department FMIPA UNY. On day-50, each treatment killed and cut open to be taken its prostate gland, then diameter of this organ measured, fixated, and processed for the making of microanatomical slide preparats with paraffin method. Data covered the prostate glands weight and diameter, tested with Analysis Variance (ANOVA = 0,01 or 0,05), then continued with Duncan’s Test (DMRT). Qualitative analysis conducted on histological structure of prostate glands and presented on microphotographs. Result of research showed that extract of meniran influenced the size of male white rat prostate gland. The higher concentration tend to degrade prostate gland diameter size. Extract of meniran have a qualitative effect on histological structure of male rat prostate gland. This showed from  the higher extract concentration give influence progressively thickness lost of lobular epitel wall and decreasing of mass secretions in the prostate gland luminal.   Keywords: rat, prostate gland, meniran, histology.
