Herbal Medicine is still widely used by Indonesian people for treatment. BPOM prohibits the use of Medicinal Chemicals in herbal preparations. This study aims to method validation analyze the content of allopurinol and dexamethasone in the stiff rheumatic herbal medicine circulating in Semarang Regency. The methods used were TLC for qualitative analysis and UV-Vis spectrophotometry for quantitative analysis. The samples used consisted of 8 samples of rheumatic and tamarind aching herbs. urates circulating in Ungaran, which are not registered with BPOM. The stationary phase used was Silica Gel GF 254 Plate. The mobile phase used chloroform:ethyl acetate 1:4. Of the 8 samples analyzed, there were 2 samples containing allopurinol, namely samples E and F which had an Rf value of 0.2 equal to the Rf value of the standard allopurinol used and none of the samples contained dexamethasone. The allopurinol content in sample E was 2.17% and in sample F was 1.14%. The results of the method validation carried out obtained RSD values = 0.75%, % recovery 98,2-101,6, LOD = 1.09 ppm and LOQ = 3.66 ppm. There are 2 samples of herbal medicine containing allopurinol with levels of 2.17% and 1.14%. The UV-Vis spectrophotometric method used met the validation. BPOM melarang penggunaan Bahan Kimia Obat di dalam sediaan jamu. Penelitian ini bertujuan untuk melakukan validasi metode serta analisis kandungan allopurinol dan deksamethason di dalam jamu pegal Linu yang beredar di Kabupaten Semarang. Metode yang digunakan adalah KLT untuk analisis kualitatif dan Spektrofotometri UV-Vis untuk analisis kuantitatif. Sampel yang digunakan terdiri dari 8 sampel jamu pegal linu dan asam urat yang beredar di Ungaran, yang tidak terdaftar pada BPOM. Fase Diam yang digunakan adalah Lempeng Silika Gel GF 254 Fase Gerak yang digunakan kloroform:etil asetat 1:4. Dari 8 sampel yang dianalisis terdapat 2 sampel yang mengandung Allopurinol, yaitu sampel E dan F yang mempunyai harga Rf sebesar 0,2 sama dengan harga Rf baku allopurinol yang digunakan dan tidak ada sampel yang mengandung deksametason. Kadar allopurinol pada sampel E sebesar 2,1% dan dalam sampel F sebesar 1,14%. Hasil validasi metode yang dilakukan diperoleh nilai RSD = 0,75%, perolehan kembali 98,2- 101,6 %, batas deteksi = 1,09 ppm dan batas kuantifikasi = 3,66 ppm. Terdapat 2 sampel jamu yang mengandung allopurinol dengan kadar 2,17% dan 1,14%. Metode spektrofotometri UV-Vis yang digunakan memenuhi persyaratan validasi.
