Pemberian Air Kelapa Muda untuk Menurunkan Kadar Glukosa Darah Puasa pada Tikus dengan Sindroma Metabolik

Abstract

Metabolic syndrome can be associated with increased lipid peroxidation processes which are characterized by increased levels of MDA, associated with inflammation which is characterized by abnormal production of proinflammatory cytokines, associated with oxidative stress, decreased antioxidants and increased ROS. The purpose of this study was to prove the effect of young coconut water on fasting blood glucose, HbA1C, IL-6 and MDA levels in rats with metabolic syndrome. This experimental research design was posttest only with control group. Twenty four male Wistar rats were used in this study, which were randomly divided into 4 groups, namely group K1 (control); K2 (MS); K3 (MS+ young coconut water 8 mL/200 gBW); K4 (MS + metformin 45 mg/kg BW). Induction of metabolic syndrome rats were given food high in fat and fructose, folic acid for 14 days and Streptozotocin (STZ) 65 mg/kgBB and Nicotinamide 230 mg/kgBB for 3 days. Data on fasting blood glucose, IL-6 and MDA levels were analyzed using the One Way Anova test, while HbA1C levels were using the Kruskal Wallis test. The results of the analysis showed that the average fasting blood glucose, HbA1C, IL-6 and MDA levels in K2 increased compared to K1, decreased in K3 compared to K2 as well as in K4. P value = 0.000, it was concluded that young coconut water has proven to have an effect on fasting blood glucose, HbA1C, IL-6 and MDA levels in rats with metabolic syndrome.Keywords: young coconut water; metabolic syndrome; fasting glucose levels; IL-6 level; MDA levels ABSTRAK Sindroma metabolik dapat dikaitkan dengan peningkatan proses peroksidasi lipid yang ditandai dengan meningkatnya kadar MDA, berhubungan dengan inflamasi yang ditandai dengan produksi sitokin proinflamasi yang abnormal, berhubungan dengan stress oksidatif, penurunan antioksidan dan peningkatan ROS. Tujuan penelitian ini adalah membuktikan pengaruh air kelapa muda terhadap kadar glukosa darah puasa, HbA1C, IL-6 dan MDA pada tikus dengan sindroma metabolik. Desain penelitian eksperimental ini adalah posttest only with control group. Dua puluh empat ekor tikus jantan galur wistar digunakan dalam penelitian ini, yang dibagi menjadi 4 kelompok secara random yaitu kelompok K1 (kontrol); K2 (SM); K3 (SM+air kelapa muda 8 mL/200 gBB); K4 (SM+metformin 45 mg/kgBB). Induksi sindroma metabolik tikus diberikan makanan tinggi lemak dan fruktosa, asam folat selama 14 hari dan Streptozotocin (STZ) 65 mg/kgBB dan Nicotinamid 230 mg/kgBB selama 3 hari. Data kadar glukosa darah puasa, IL-6 dan MDA dianalisis dengan uji One Way Anova sedangkan kadar HbA1C dengan uji Kruskal Wallis. Hasil analisis menunjukkan bahwa rerata kadar glukosa darah puasa, HbA1C, IL-6 dan MDA pada K2 meningkat jika dibandingkan dengan K1, pada K3 terjadi penurunan dibandingkan dengan K2 begitu juga pada K4. Nilai p = 0,000, maka disimpulkan air kelapa muda terbukti berpengaruh terhadap kadar glukosa darah puasa, HbA1C, IL-6 dan MDA pada tikus dengan sindroma metabolik.Kata kunci: air kelapa muda; sindrom metabolik; kadar glukosa puasa; kadar IL-6; kadar MDA
