Abstract

The addition of precursor into in vitro Murashige Skoog (MS) media culture expected to be able improve curcuminoid synthesis within the callus. Phenylalanine is precursor in curcuminoid synthesis. 2,4-D hormone is also added into media culture to induce callus formation. The concentration of 2,4-D hormone and phenylalanine is 0, 2 and 4 ppm, respectively. The qualitative analysis was done with color reaction and Thin Layer Chromatography (TLC) with gel silicon GF254 as stationary phase and chloroform : ethanol 96%: glacial acetic acid (94:5:1) as movement phase and detected by the visible ligh. The quantitative analysis was done by measuring the spot area in the TLC chromatogram with TLC scanner, and then was calculated using standart curcumin curve. Base on the experiment indicates 2,4-D Hormone with 4 ppm result the fastest time callus induction. Curcuminoid content is higher in rhizome compared in callus. The combination of 2,4-D hormone and phenylalanine yield the higher curcumin content than desmetoxi-curcumin for each treatment combination. The highest curcumin content is produced by 2,4-D hormone and phenylalanine combination with 4 ppm concentration that is 0,39 %. Meanwhile the highest desmetoxi-curcumin is produced by 2,4-D hormone and phenylalanine combination with 4 and 2 ppm concentration, respectively, that is 0,18 %. 2,4-D hormone and phenylalanine affects more significantly in curcumin synthesis rather than desmetoxi-curcumin.

Abstract

Interactive mixtures give advantages for the mixtures containing low dose drug such as prednisone as it guarantee homogeneity, stability and increase the dissolution of prednisone. Flowability and compactibility of the interactive mixtures are relatively good, it can be directly compressed to produce tablet. The addition of lubricant / glidant of Mg stearat , talc or their mixture as the cohesive materials to the prednisone-granule interactive mixtures produce prednisone- granule-lubricant/ glidant which affect the tablet properties such as disintegration time and release of the drug. Based on these matter, a study was done to examine effect of Mg stearat, talc or their mixtures to the disintegration time and dissolution of prednisone interactive mixtures tablet. For the investigation of these effects, experiments were done using optimization approach based on simplex lattice design with Mg stearate and talc as the independent variable. The experiments data were implemented into the equation relation to the simplex lattice design : Y= a(A) + b (B) + ab (A) (B). Based on the coefficient value of the disintegration time and drug dissolution equations, profiles are made, lubricant/glidant effects on the disintegration time and drug dissolution can be analyzed. Magnesium stearate cause prednisone interactive mixtures disintegration time tablet are more longer than talc, although the affect interaction is relativity more little. Magnesium stearate compared with talc gives C30 value and DE-60, although the interaction gives affect relativity more little.

Abstract

Avicel PH 101 is an excipient in tablet making that can be used as diluent, binder, lubricant, and disintegrator. Avicel is not economically an advantage so that it needs to be combined with cheaper materials. Combination of Avicel PH 101 with corn starch is able to accelerate the disintegration time of the tablet. Mimba (Azadirachta indica A. Juss) leave is a traditional medicine that is used as anti-inflammation. The study was to find out the optimum proportion of Avicel PH 101 and corn starch mixture in tablet making of mimba leaves extract by simplex lattice design method. The extract of mimba leaves was obtained by maceration of mimba leaves powder with ethanol 70%. The obtained extract was evaporated until a thick extract was obtained. Tablet formulation was based on SLD method with excipients of Avicel PH 101 (A) and corn starch (B) i.e. F1 (100% A), F2 ( 50% A : 50% B), F3 (100% B). The tablets of mimba leaves extract were made by wet granulation. The equation was used to make tablets with total respond of the most optimum granules physical properties. The data of the physical properties of granules from optimum formula as calculation result based on SLD equation with the experiment result was analyzed using T-test. Tablets from optimum formula were physically tested including weight uniformity, hardness, friability and disintegration time. The result of theexperiment indicated that the mixture of 90% Avicel PH 101 – 10% corn starch produced an optimum result in granules physical properties and the obtained tablets of mimba leaves extract fulfilled tablet physical property test.

Abstract

Iron Deficieny Anemia (IDA) is one of msjor micronutrient deficiency in Indonesia. IDA is that Indonesian people suffered from 100 million of population. One of the correct ways IDA problems is suplementation with ferrous sulfate. To can use raw material farmacy, it must fulfilquality standart in Indonesia Pharmacope Edition IV.The aims of research is producing, purification and analyses ferrous sulfate from iron waste. Producing of ferrous sulfate is reacting iron waste with acid sulfuric 25% during 2 days. Separated and purification crystal with recrystalyzation then determination with SEM-EDS analysis. The obtain of research shows ferrous sulfate content Fe 54,46%, O 33,55% and S 11,99%. These results show that ferrous sulfate from iron waste fulfil quality standart in Indonesia Pharmacope.

Abstract

Mondokaki plant (Tabernaemontana divaricata, R.Br.) is a traditional medicine used in medicating high blood pressure, pharyngitis, conjunctivitis, mastitis, mad dog bite, antitusissive, anti-inflammatoty and sprain. The aim of the research was to know the anti-inflammatory effect of mondokaki leaves ethanolic extract on male shite rat. Mondokaki leaves were extracted using ethanol 70% by maceration method. The extract was tested to each group of the rat with doses of 3.75 mg/200 g bw, 7.5 mg/200 g bw, 15 mg/200 g bw. The negative control group was given CMC 0.5% solution and the positive control group was given sodium diclofenac as antiinflammatory agent. The compounds were all given orally one hour before the rat plantar was injected with carragenin 1% to make edema. The anti-inflammatory effect was observed from the ability to reduce edema volume at one hour interval for five hours. One way analysis of varian at 95% confidence was done to know the difference in each treatment and LSD test was done to know the difference between the treatment groups. The result of the research showed that ethanolic extract of 15 mg/200 g bw dose did not significantly different compared with the positive control. It was indicated that ethanolic extract of 15% mg/200 g bw dose had anti-inflammatory effect by reducing edema volume of male white rat in proportion with the positive control.
