Abstract

Screening ISSR primer preliminary study of the relationship between small, big and red gingerScreening ISSR Primer as preliminary study the relationships of small, big and red ginger. A PCR- based method for genetic diversity analysis such as ISSR has been well developed recently. In this research, 28 ISSR primer were screened aiming to study the relationships of red, small and big ginger. Genomic DNA was isolated from rhizome using CTAB based method. The DNA were then amplified in a thermalcycler wite a total volume of 20 µl consisted of 30 – 100 ng genomic DNA, 10 pmol primer, 1x PCR buffer, 0,2 mM dNTPs, 1,5 mM MgCl2 dan 0,5 rTaq polymerase. Electroforesis of the amplification product was carried out on 1,5% agarose gel, stained with ethidium bromide and viewing on UV transiluminator light to see the banding pattern. Results showed that amplified banding pattern of ISSR range from 3 – 14. Among the 28 primers screened, 10 primers showed polymorphic banding pattern but only 3 primers showed polymorphic banas more than 25%, i. e. ISSR 18, ISSR 19, and ISSR 15 respectively. Based on the ISSR data, small and big ginger has closer relationships compared to the red ginger. To find the suitable primers that can be used to distinguished the ginger cultivar and find suitable molecular markers, more primers need to be screened.

Abstract

Cultivation, processing and farming system feasibility of gambier in 50 Kota district, West SumateraGambier (Uncaria gambir Roxb) is one of specific export commodities from west Sumatera. However, this propective cash crop has not followed by the increase of its productivity, quality and farmer income. This conditions, may due to the lack of farmers, know ledge in processing and limited capital for developing gambier farming system. The objective of this research was to evaluate the cultivation and processing techniques, feasibility of farming system, income of gambier farmers and the constrains for developing gambier farming system in this region. The research was conducted in Solok Bio-bio village, Harau sub district, district of 50 Kota, West Sumatera on Januari 2000. The rapid rural iralic (RRI) and survey methodes, by directly dialoque and filling of quistioner were used. Thity respondens were determined by simple random. Farmer income was analyzed 50 by analysis of income, while feasibility of farming system was evaluated by benefit cost ratio (B/C Ratio), net present value (NPV) and internal rate of return (IRR). The results indicated that the cultivation and processing of gambier in this region were still coducted by traditional methods, using simple equipments. The feasibility of farming system showed that at level interest of 15%, the B/C Ratio = 1,61, VPV = Rp. 9.763.532,- and IRR = 57%. Farmer income was Rp. 20.186.250,- or average Rp. 1.682.180,-/year. Based on this results is can be concluded that gambier farming system in Solok Bio-bio was feasible to develop. The main constraints for developing gambier were less of human resources, such as low education, high cost of post harvest, and limited of capital. In order to eleminate these problems emprowering of gambier farmers and its organization, and increasing farmers skill are very urgent.

Abstract

Limbah dari tanaman cengkeh yang berupa gagang dan daunnya belum sepenuhnya dimanfaatkan di daerah Sulawesi Utara, padahal dari limbah ini masih dapat diperoleh minyak dengan cara penyulingan. Penelitian ini bertujuan untuk menganalisis sistem perencanaan awal sebuah agroindustri penyulingan minyak atsiri dari daun cengkeh di wilayah Sulawesi Utara, serta menganalisis kelayakannya baik dari aspek teknik, manajemen maupun aspek finansialnya. Hasil penelitian ini diharapkan dapat membuka wawasan serta peluang pengembangan teknologi penyulingan minyak atsiri dari daun cengkeh khususnya di wilayah Sulawesi Utara dalam skala usaha yang lebih besar. Diharapkan teknologi ini mampu meningkatkan nilai tambah komersial yang akan meningkatkan kesejahteraan petani. Metode yang digunakan adalah rancangan tata letak pabrik (plant layout design) dan metode ekonometrik. Berdasarkan kriteria pemilihan lokasi pabrik di Sulawesi Utara, maka Kabupaten Minahasa merupakan lokasi yang tepat dilihat dari segi ketersediaan bahan baku, kemudahan pemasaran, kemudahan transportasi, ketersediaan tenaga kerja, sarana listrik, air, kemudahan investasi, iklim, unsur penunjang dan prospek jangka panjang. Secara teknis pabrik yang dirancang memiliki kapasitas penyulingan 18 ton daun cengkeh kering per harinya dengan prediksi perolehan minyak 504 kg/ hari pada rendemen penyulingan 2,8%. Secara finansial prediksi investasi yang dibutuhkan untuk membangun pabrik tersebut di atas adalah Rp. 863.132.800,-.Modal investasi ini diperkirakan akan kembali selama 0,63 tahun atau 7,56 bulan dengan titik pulang pokok 10.515,2 kg/ tahun. Kekayaan perusahaan pada akhir proyek sebesar Rp. 13.181.990.610,-. Dari hasil analisis kelayakannya diperoleh NPV Rp. 5.353.342.926,- (lebih besar dari nol), nilai IRR lebih besar dari tingkat suku bunga yang berlaku (18%) yaitu 49,2 % dan B/C rasionya 1,66 (lebih besar dari 1), sehingga dapat disimpulkan bahwa pengembangan teknologi penyulingan minyak di Sulawesi Utara layak untuk dilaksanakan.

Abstract

The effect of shade and phosphor on the growth and production of PogostemonA pot experiment aiming at the optimal effect of shade and phosphor fertilizer on the growth and yield of Pogostemon plant of the Sidikalang type was carried out at Cimanggu experimental garden of Indonesian Spice and Medicinal Crops Research Institute, Bogor, from September 1998 to January 1999. The research was carried out factorially with a randomized block design with four replications. The treatments consisted of two factors, which are shade (without shade and 50% shade), and phosphor fertilizer (without phosphor, 0,69 g P/polybag, 1,38 g P/polybag). The result of the research showed that 50% shade treatment significantly increased the plant height, number of leaf, number of branch, wide of crown, stem diameter, fresh and dry leaf weight, fresh and dry stem weight. The P treatment (0,69 g P/polybag) significantly increased the number of branch, number of leaf and stem diameter. Significant on Interaction between shade and P Occuronly on stem diameter. The interaction of 50% shade with 0 g P/polybag showed the biggest dramecer treatment, followed by 50% shade with 1,38 g P/polybag and 50% shade with 0,69 g P/polybag

Abstract

The effect of micorrhiza abuscular on the growth of vanilla cuttings (Vanilla flanifoliaAndrews)Mycorrhiza inoculation at nursery represent an effort to gain better quality and growth of vanilla,s cutting. A research aiming at the effect of arbuscular mycorrhiza (AM) inoculation on the growth of vanilla,s cutting was conducted in the green house of the Sukamulya ExperimentalGarden-Sukabumi, Indonesian Spices and Medicinal Crops Research Institute, from April 2002 to August 2002. A complete randomized design was used, arranged factorially with 2 factors and 3 replications. The first factor was vanilla types, consisted of Gisting and Anggerek, and the second factor was AM inoculation, such as without AM, 500 spores ofAM-p/plant, and 500 spores of Mycofer/plant. AM-p was a mixture of Acaulospora spp. and Glomus sp., while Mycofer was a mixture of Glomus sp., Glomus etunicatum, Gigaspora margarita, and Acaulospora sp. The result indicated that there was no significant interaction between vanilla type and AM inoculation on the plant growth. AM inoculation significantly affected the plant height, number of leaves, stem diameter, leaf area index, and dry weight of biomass. The difference of vanilla type significantly affected the root dry weight and precentage of AM root infection, but the interaction 19between AM and vanilla type did not significantly affect the plant growth. AM-pinoculation increased the plant height, number of leaves, stem diameter, leaf area index, and dry weight of biomass equal to 31,62%, 16,94%, 6%, 25,45% and 47,05%, while Mycofer increased those parameter 49,64%, 32,01%, 6%, 32,41% and 54,85%, respectively. The Anggerek type showed better responses to the AM inoculation than the Gisting type.

Abstract

Potency, opportunity and constraints in development of purwoceng (Pimpinella pruatjan Molkenb)Purwoceng (Pimpinella alpina) farming system, its potency and constraint in development was studied at Dieng area, Wonosobo regency, at September 2003. The location was chosen purposively, considering that the area is the center of origin and the development of purwoceng succeed only at this area. The study was conducted with a survey method, and the samples of farmers were taken by semi- census. The income of purwoceng farming was studied by analysis of income, the potency, prospect and constraint in developing purwoceng were studied byquantitative-descriptive analysis. The results of analysis showed that a net income of Rp. 34.900.000,- for 7 – 8 months yield after harvest was achieved with a 1.000 m square of land. This indicate that developing purwoceng farming system has a good prospect. Until now the demand for purwoceng at Wonosobo regency can not be full filled due to constrain in land that meet the technical requirements for this commodity.
