Abstract

Daun kesum (Polygonum minus Huds.) merupakan salah satu sumber daya hayati yangsangat potensial yang telah lama dimanfaatkan oleh masyarakat. Daun kesum mengandungsenyawa golongan fenolik,alkaloid, flavonoid, steroid-terpenoid, tannin dan saponin yangberperan dalam aktivitas antibakteri. Tujuan dari penelitian ini adalah untuk mengetahuikomposisi optimum dari karbopol dan HPMC dalam memformulasi gel ekstrak daun kesummenggunakan metode Simplex Lattice Design. Ekstraksi daun kesum menggunakan metodemaserasi dengan pelarut metanol. Uji efektivitas antiseptik menggunakan metode discdifussion. Gel formula optimum diuji sifat fisik dan kimia. Hasil uji One Way ANOVA padakonsentrasi 5%, 10% dan 15%, menunjukan bahwa konsentrasi 10% memberikan dayahambat optimum dan diformulasikan dalam bentuk gel. Formula gel optimum berdasarkanSimplex Lattice Design adalah kombinasi Karbopol 80% - HPMC 20%. Hasil verifikasidengan uji T satu sampel bebas menunjukkan tidak ada perbedaan yang signifikan antaradaya hambat formula optimum hasil perhitungan (10,205mm) dengan hasil pengujiansesungguhnya (12,389mm). Pada perbandingan gel optimum dengan kontrol positif jugamenunjukkan tidak adanya perbedaan yang signifikan terhadap efektivitas daya hambat.Hasil gel optimum yang didapatkan memiliki warna hijau tua dengan rata-rata viskositassebesar 41,667 poise, daya lekat 50,06 detik, daya sebar 6,820 cm2dan pH 4,567.Kata kunci : kesum, Polygonum minus, gel, karbopol, HPMC

Abstract

ABSTRACTCiprofloxacin is an antibiotic that has low solubility in water and formed intosuspension in order to produce stable product in liquid form. This research aims tooptimize the proportion of a mixture suspending agent in order to produce optimumphysical properties. Optimize suspension physical properties used factorial designmethod with Design Expert software version 8.0.7.1 trial. Pulvis Gummi Arabici (PGA)and Carbopol 934 as independent variable meanwhile viscosity, sedimentation volumeand redispersibility as dependent variable. Based on that method, obtained four formula(formula A, B, C, and D) and physical properties were measured. Analytical of the resultsobtained from the equations and graphs for each response, by which optimum formulacan be determined. The optimum formula based on factorial design method was FormulaA (Carbopol 934 and PGA low concentration) with value of desirability 0,684 and showlow viscosity, particles rapidly settle but easy redispersibility. Analytical statistic ofphysical properties in optimum formula and prediction show insignificance difference (pvalue> 0,05). The antibacterial activity in optimum formula and positive control(ciprofloxacin) show insignificance difference (p-value > 0,05) which mean ciprofloxacinstill has antibacterial activity in suspension form. Abstract

Kratom (Mitragyna speciosa Korth.) is one of traditional medical plant that canproduces both stimulant effects (in low doses) and sedative effects (in high doses). Thisstudy aims to prove the sedative effect of ethanol fraction of kratom leaf, the effectivedose, and compared the effect ethanol fraction of kratom leaf with diazepam. Dry leavesof kratom was maceration in ethanol 70% for 3x24 hours. Ethanol extracts wasfractination in n-heksan, ethyl acetat, and ethanol 70% to produce fraction ethanol. Thisstudy was conducted by Traction test and Fireplace test method on male mice of BALB/cstrain. Observation as qualitative is observing the reflect act of turning the body and thecornea reflect, while quantitative is fell down time, turning the body, and turn out ofcylinder tube. Twenty five heads of mice were randomly in five groups : positive controlgroup (diazepam), negative control group (suspense CMC 1%), group of kratom leaffraction ethanol 27.20 mg/20gBW (Dose I), 54.39 mg/20gBW (Dose II), and 108.78mg/20gBW (Dose III). The results show, there are sedative effect at doses of 27.20mg/20gBW, 54.39 mg/20gBW, and 108.78 mg/20gBW, which the all dose are greaterpotential than positive control group (diazepam). Effective dose of fraction ethanol ofkratom leaf is 27.20 mg/20gBW.Keywords: sedative, kratom leaf, ethanol fraction.

Abstract

Insomnia is a sleep disorder that can cause a variety of serious health problemsrequiring treatment. The using of synthetic drugs have dangerous side effects than therapywhich is resulted. Therefore, alternative treatments which derived from natural such askratom leaves are needed. The purpose of this study is to know the sedative effect of nhexaneextract, the group of secondary metabolites, the effective dosage of n-hexaneextract of kratom leaves in producing sedative effect and thepotential of the extract nhexaneif it compares with diazepam. Extracts is obtained by maceration and somephytochemical screenings are done. The study used 30 mices were divided into 6 groups.Diazepam is used as a control (+), vegetable oil as a control (-) and n-hexane extract withdosage 1 (24 mg/kgBW), dosage 2 (48 mg/kgBW), dosage 3 (72 mg/kgBW) and dosage4 (96 mg/kgBW) which is given by oral administration. Sedative effects experiment wasconducted by using the traction test and fireplate test. The observation was done both ofqualitative and quantitative ways. Qualitative data includes turning the body and corneawhile quantitative data includes time falling, turning the body and jumping. Quantitativedata was tested by statistic using One-way ANOVA Post Hoc Test (Tukey HSD andLSD) Kruskal Wallis or Mann-Whitney. The results showed that n-hexane extract hadsedative effect contained the compound group of alkaloids, glycosides, steroids andflavonoids. The dosage 4 (96 mg/kgBW) of n-hexane extract of kratom leaves gavesedative effects better than diazepam.Keywords: Insomnia, Sedatives, Kratom

Abstract

Drug interaction is one of eight drug-related problems categories that may affectpatient clinical outcome. Increasing complexity of the drugs used in current treatment andthe likelihood of polypharmacy practices, make possible the incident of drug interactions.The research aims to know the number and mechanism of the drug-drug interactions inprescriptions contains oral antidibetic, identify drugs are often interact as well as analyzecorrelation between the number of medication in one prescription with drug interactionfound. The study was conducted in dr. Soedarso hospital, Pontianak from the January toMarch 2013. The research used non-experimental methodology by using descriptiveanalysis in a retrospective way. Data analysis using Chi-Square Test on SPSS 17.0. From1435 outpatient prescription sheets that meet the inclusion criteria, found that druginteractions occur 62.16% prescription which received medication oral antidibetic. Thepatterns of interaction mechanism are pharmacokinetic interaction 13.56%,pharmacodynamics 34.15%, and unknown 52.29%. The types of drugs that often interact,among metformin and gliclazide. Incidence of drug interactions occur 6 times higher insubjects who received ? 5 drugs compared with patients who received < 5. The result ofstatistical analysis showed that there is significant correlation between the number ofmedication in one prescription with incidence of drug interaction found drugs (p<0.05).Keyword : drug inteaction, oral antidiabetic, dr. Abstract

Chemical contents of Kesum (Polygonum minus Huds), such as phenolic, flavonoids, alkaloids, steroid-terpenoids and tannin, can act as antibacterial agent.The aims of this research was to determine whether methanol extract of kesumleaves have antibacterial effect and its effectivity when it was formulated in gelproduct. Extract activity test group being used are negative control, methanolextract 5%, 10% and 15%. Gel effectivity test group being used are negativecontrol, positive control, Gel A, (5% methanol extract), Gel B (10% methanolextract) and Gel C (15% methanol extract). Disc diffusion Kirby-Bauer methodwas used to determine the antibacterial effectivity against Escherichia coli. Gelstability test carried out for 31 days include organoleptic, viscosity, ability ofspreading and sticking and pH. The methanol extract 5%, 10% and 15% gaveantimicrobial effect. It was formulated in gel product with that concentrations.The result of the effectivity test and stability test were analyzed statistically bySPSS 17.0 using One-Way ANOVA and continued to Post Hoc Tukey Test.Diameter of the inhibition zone gel was Gel A (10.66 mm) Gel B (12.11 mm), GelC (12.33 mm). Gel C gave best antimicrobial effect between Gel A, Gel B andpositive control. So it has potensial to be antiseptic agent. In all variables of gelstability test, Gel C has a few changes, so Gel C stability is better than Gel A andGel B.Keywords: kesum leaves, gel, antiseptic, Escherichia coli

Abstract

Ascorbic acid is a compound that unstable, easily oxidized when exposedto aerial (oxygen) and this process can be accelerated by heat. The purpose of thisresearch is to see the influence of storage conditions on the quality of drugsphysical and chemical tablets of ascorbic acid at the district health centers whichincludes Kampung Bali, Alianyang, Pal tiga and Karya Mulia with differentstorage conditions in each health centers. The method used is non-experimental,and use probability sampling method with simple random sampling. Samplingwas conducted during two months. In this research, do measurement temperature,humidity and evaluation test tablets that include test physical appearance,uniformity of weight, uniformity of size, friability, hardness, disintegration time,dissolution and value, then the results were analyzed statistically using One WayANOVA and Kruskal Wallis. The results showed that ascorbic acid tablets in thefirst and second health center in Kampung Bali, Alianyang, and Pal tigaparameters qualify the uniformity of weight, disintegration time and dissolution,while the Karya Mulia of community health center tablets meets all requirementsof ascorbic acid tablets are good, except the value parameters and uniformity ofsize. Based on the results of this research concluded that the influence of theconditions and duration of storage of drugs affecting the quality of ascorbic acidtablets. Statistical analysis evaluating ascorbic acid tablets between each healthcenters in the first month and the second shows that the health center Karya Muliashowed a significant difference with health center Kampung Bali, Alianyang andPal tiga.Keyword: Ascorbic acid Tablets, Health Centers, Storage Conditions

Abstract

Mefenamic acid is a derivative of anthranilic acid is widely used by the public asan analgesic, antipyretic, and anti-inflammatory. Mefenamic acid is a drug which issensitive to light, temperature and humidity. This research aims to determine the qualityof mefenamic acid caplets that stored in Pontianak City community health centre as amatter of consideration in storage of both drugs in community health centre. Research hasbeen conducting by doing some observation on drug storage conditions such astemperature and humidity with non-experimental methods. Mefenamic acid samples weretaken twice during the research, mefenamic acid samples at the beginning and at the endof storage that had been stored for a month in storage room of each community healthcentre. The quality of mefenamic acid caplets evaluated, namely physical and chemicaltests. The results showed that the storage conditions at 4 community health centres do notall qualify, only Alianyang community health centre that qualified. Community healthcentres room temperature ranged from 25 to 33,3 C, humidity 52 to 88,3%. Temperatureand humidity affect the physical and chemical quality of mefenamic acid caplets in allexamination parameters except weight uniformity. Quality phsysical and chemical ofmefenamic acid caplet getting dropped after being stored for a month, but still meet therequirements of a good quality caplets.Keywords: mefenamic acid, temperature, humidity, health centre

Abstract

Drug interaction happen when drugs is given together with foods. This researchwas aimed to observe the effect of mangosteen juice to the paracetamol pharmacokineticprofile in Wistar male white rats. The study was using male Wistar rats weight 200-350 g.The animals were divided into three groups with the total of the male rats are 15. Group I(paracetamol) was given a single oral paracetamol 9 mg/200gBW. Group II (treatment 1)was given a single oral paracetamol 9 mg/200gBW together with mangosteen juice 2,88g/200gBW. Group III (treatment 2) was given a single oral paracetamol 9 mg/200gBWtogether with mangosteen juice 0,66 g/200gBW. The serial blood was collected from tailvein for 9 hours and were analyzed using spectrophotometer UV for unchangedparacetamol. The pharmacokinetic parameters of paracetamol were calculated byrecidual method and were analyzed by One Way ANOVA using 95 % confidenceinterval. The difference between groups were analyzed using Post Hoc LSD-test. Theresults showed that the group II (paracetamol-mangosteen 1) and group III (paracetamolmangosteen2)increased of ka, Cpmaks, t, tab, ke, tel, CL, AUC and decreased ofka, ke, and CL. The research showed that mangosteen juice doses 2 give higher decreasedmetabolism and decreased elimination of paracetamol in rat.maksKey words : Mangosteen fruit, Pharmacokinetics, Paracetamol, Drug interaction

Abstract

Insomnia  is a condition difficult start to sleep, can not sleep or inadequate sleepduration. One of plants which guess have sedative effect is kratom (Mitragyna speciosa). This research was conducted to determine the sedative effect of kratom leaves infusa to male BALB/cstrain mice. In addition, it aims to determine the effective dose and potential kratom leaves infusawhen compared with diazepam The mice divided into 5 groups of  positive control group (diazepam), negative control(aquadest), and infusa of kratom leaves groups dose 1,95; 3,9; and 7,8 g/KgBW. The test of sedativeeffect was conducted with Traction test and Fireplace test method on male BALB/c strain mice.Monitoring of quantitative data are duration time of mice to turning the body and duration time ofmice to fall down, as soon as duration time of mice to come out from cylinder tube. Qualitative data isobserved presence or absence of reflexes behind the body and corneal reflexes. Result of the quantitative data was analyzed with One Way ANOVA and supported by SPSS.Based on the result, all of kratom leaves infusa dosages has sedative effect, which is the mosteffective dose kratom leaves infusa at dose of 7.80 g / KgBW. But  the sedative effect still below ofdiazepam. Key Word : kratom leaves, sedatives, infusa.
