Abstract

Melaleuca alternifolia (tea tree) is one of Australian species for oil production purpose. Breeding strategies to improve the oil quality has been established since 1998, amongs them is artificial hybridisation between M. alternifolia with its close relatives of M. linariifiolia and M. dissitiflora. This study therefore aimed to produce hybrids of M. alternifolia x M. linariifolia and M. alternifolia x M. dissitiflora. These hybrids are expected to have oil with chemotypes meeting the standard requirements and to improve adaptability to drought environment. Two methods being used in this study were: conventional controlled pollination and one stop pollination. The results showed that both pollination methods successfully produce hybrid of M. alternifolia x M. dissitiflora. The number of capsule produced from conventional controlled pollination was much higher than from one stop pollination. Hybrids seedlings at 3 months old showed intermediate morphological characteristics of the parental seedlings.

Abstract

Pollen movement is a critical determinant in population genetic, plays important role in forest tree improvement and conservation genetic. This study was conducted to understand physical pollen movement and to determine seasonal genetic diversity of seed crops in a seedling seed orchard (SSO) of Pinus merkusii, located at Jember, East Java. Physical pollen movements were measured using 25 pollen traps conducted at three flowering seasons i.e. August 2005, November 2005 and March 2006. For offspring populations, seeds were collected at June, September and December that represented mating in each pollen observation season. Then, genetic diversity of the seed crops were analyzed using five microsatellites markers for P. merkusii. Results showed that number of pollen grains was significantly difference among seasons, but insignificant to both trap locations and interaction season-trap locations. The trapped pollen number was low in both August and March, but it was abundant in November. Value of gene diversity of offspring was highest in August (HE = 0.402) compared to other seasons. This study showed that physical pollen movement in the SSO might synchronize, however level of the HE varied among seasons. Seasonal variation in mating status was briefly discussed.

Abstract

Flowering stimulation were investigated to accelerate deployment of E. occidentalis. It was found that extended daylength (16-h) in juvenile plants stimulate flowers of very limited individuals/family with less number of buds than normal inflorescences. Extended daylength (16-h) promote flowering of most families from 2 year-old E. occidentalis plants. In combination with paclobutrazol (1 mg. a.i/mm), number of buds were doubled. Under 16-h daylength, juvenil plants were taller up to 25 weeks than under natural light but shorter for family 96-LM that produced more flowers than others. There were no height differences between 16-h daylength and in combination with paclobutrazol in 2-year old plants.

Abstract

The objectives of this study were to investigate the level of genetic variation and to describe its distribution within and among populations of merbau. Leaves of wildling were collected from four populations distributed in Papua and Moluccas islands. Genetic variation was detected by isoenzyme markers using four enzyme system i.e. Peraxidae (POD), Esterase (EST), Diaphorase (DIA) and Glutamate Oxaloacetate Transaminase (GOT). The result showed that 17 alleles were identified and distributed on 6 polymorphic loci. The mean percentage of polymorphic loci was 95.8 %. The average genetic diversity within population was 0.049. The UPGMA cluster analysis based on Nei’s standard genetic distance reflected three clusters, i.e. Haltim and Waigo formed first cluster, Nabire formed second cluster, and Seram formed third cluster separated from two other clusters.

Abstract

Falcataria moluccana seedling seed orchards was established in 2006 located in Kediri East Java in collaboration between the Center for Forest Biotechnology and Tree Improvement Research and Development and Perum Perhutani. 77 open-pollinated families from four seed sources were progeny tested in Pare Kediri, East Java. The trial was arranged in a randomized completely block design, 4 tree line plot and replicated 6 times. At 8 months of age there were significant differences among seed sources for diameter. The survival rate of family varied from 54.2 % to 86.5%. Difference betwen families within seed sours were significant only for diameter growth. Heritability estimanes for diameter were moderate (h2i= 0.16; h2f = 0.42).).Genetic correlation beetwen diameter and height was strong (rg = 0.89) and higher than  phenotipic correlatione (rp = 0.86).
