Abstract

Gyrinops verstegii is one of the species which produce high economic value of gaharu (agarwood). The objectives of this research were to estimate genetic diversity within and among populations, investigate distribution of genetic diversity, and genetic relationship among five populations of G. Verstegii. Leaf samples were taken from five populations of G. Vertegii in Lombok, NTB,. Genetic diversity was analyzed using 21 selective primers out 200 primers. A total of 42 polymorphic loci were detected. Mean of genetig diversity was 0.288, and genetic distance betwen populations was 0.148. The Proportion of 85.24 % of genetic diversity was distributed within population, and the remaining 14.76 % was distributed between populations. Based on cluster analysis, the five populations were separated into two major group consisted of Pusuk Pohon Induk, Pusuk Sumber Benih, and Mantang I. The second group consisted of Mantang Pohon Induk and Mantang II. Collecting genetic materials for conservation and tree improvement program should be focused on the materials within population and each group (regent) should be a representative of at least one population.

Abstract

Preliminary shoot groeth on acacia hybrids (Acacia mangium x A. Auriculiformis) was observed to assess their in- vitro shoot initiation. Explants were taken from 4 months old seedlings of A. Mangium, A. Auriculiformis and their hybrid. Murashige & Skoog’s (MS) medium was used supplemented with various concentration of BAP, IBA,NAA, and GA4. Parameters measured were bud breaking, number average number of leaves of acacia hybrid was 7 and 1.9 cm and 5.2 respectively, found in medium Ak2 culture. Bud breaking of acacia hybrids on that medium happened 15 days earlier than pure Acacia. This finding showed that acacia hybrid could be propagated by  micro propagation succesfully.

Abstract

The purpose of this research is to examine the effect of auxin (NAA) and cytokinin (BAP) on shoot initiation in tissue culture of sengon (Falcataria moluccana)using cotyledon segment. Cotyledon segments were colleect from aseptic seedlings. Cotyledon segments were aseptically cultured on a murashige and  Shoog’s (MS) medium containing different concentrations of Benzyl-amino-purine (BAP) ranging from 1 to 3 mg/l and Naphtalene-acetic-acid (NAA) ranging from0,01 to 0,03 mg/l. Result of analysis of variance inicates that plant groeth regulator of BAP and NAA had significant effect on shoot formation up to 4 months age. The subsequent Duncan Multiple Range Test indicates that concentration of 0,001, 0,02 and 0,03 mg/l gave no significant effect on shoot formation and shoot growth

Abstract

In-Vitro contamination of plant tissue cultures by bacteria is one of the most serious problem in plant micropropagation. Ulin (Eusideroxylon zwageri) bacterial contamination in vitro may survive surface sterization of the explant because they are in interior tissue and some Bacterial spores can also survive the sterilization procedure even if they are un the tissue surface. Bacteria  secrete some sort of glycocalyx, an auter viscous covering of fibers extending from the Bacterium. This Study was conducted to examine the colony thickness and to observe the bacteria glycocalyx contaminant of ulin in vitro folowing three surface explant sterization treatments with fungisida, detergent, alkhohol,HgCl2  and Na2CLO3 as antimicrobial agents. The longer the sterilization time the lesser the bacterial colony thickness farmed. Transmission binocular microscopy showed that distribution of glycocalyx material surrounding coccus ulin bacterial contaminant cells were capsulated like Steptococcus lactis or Enterobacter aerogenes.

Abstract

Melaleuca alternifolia is the main source for Australian tea tree production. Improving oil quality has been attempted by controlled cross pollination between plus trees. The availability of flowers during flowering season is important for control pollination between , therefore the work to  stimulate flowering should be done. The objective of this study was to increase the flora bud production of 24 month cuttings of M. alternifolia by application of growth retardant (paclobutrasol) and exposure to low  temperatures. Results showed that the application of paclobutrazol was not effective to increase the number of floral buds production. Paclobutrazol application effected only some morphological characteristics. Such as reduced height, smaller leaves, and shorter internodes. On the other hand, exposure of shoot cutting to low temperature for 4 months period increased the number of floral buds and paclobutrazol application following exposure to low temperature was then assumed to have been responsible for the floral response..
