Abstract

Arbuscular mycorrhyza fungi (AMF) constitutes one of biological agent which is able to enhance the growth and productivity of plant. AMF reproduction has been frequenly done as well as inoculum formulation improvement. It is done by adding other subtances like organic fertilizer which is aimed at speeding up plant growth response prior to associating with AMF and increasing more growth after associating with AMF. Thus, This research is aimed at studying the response of AMF selected types G.etunicatum and Glomus sp. Toward  vermicompost and test vermicompost addition in increasing AMF inoculum quality. The research is factorial experiment with RAL using two treatment factors. First factor is AMF inoculum factor and the second factor is medium formulation. The research revealed that G.etunicatum is more tolerant on vermicompost addition compared to Glomus sp. (indegenous), the use of vermicompost, G.etunicatum type with 30% vermicompost  namely 0,12 x 104 propagul and Glomus sp. Type with 40% vermicompost is 0,19 x 104. Abstract

Standard method in physical and physiological quality testing is very imprortant to support a controlling system of distributed seed quality by certification that will provide a guarantee to prosedure; distributor, and user. Five seed lot of sawo kecil (Manilkara kauki) from Mokmer (Bali), Benoa (Bali), Lombar (Lombok), Kaliurang (Yogyakarta) and Alas Purwo (Banyuwangi) were used to identify the best method for moisture content and germination testing. Completely randomized design was applied to determine the best method for moisture content testing. The treatments in selection of the appropriateMethod were oven method on the temperature 130 – 133 oC (1-,2-,3-, and 4 hours) and temperature 103 ± 20C (16-,18-,20-,22-, and    haours). The selection of germination method used completely randomized design with 3 factors of treatment, i.e. sowing media, pretreatment, and germination condition. The result showed that oven method at temperature 103 +_20C for 24 hours and at temperature 130-1330 C 4 hours provided the best result for seed moisture content testing. For seed germination testing, seeds sowed on the sand media with drying-wetting pretreatmen for 3 days on the germination box covered by transparent plastic provided the best germination capacity and speed toward 5 seed lots. The methods can be considered to be appied in the testing of moisture conten and germination of sawo kecik seeds.

Abstract

When genetic base in a seed orchard is narrow, a startegy of genetic infusion should be applied to broaden and maintain its ganetic variation over several successive generations. For this purpose, genetic-infusion populations were established in tree improvement programs of Pinus merkusii located in jember. To be successful genetic variation of the infusion populations should be assessed. This study was aimed to determine genetic diversity of three infusion populations of P merkusii i.e. Takengon, janto and Blankejeren population using five microsatellite markersi. The results  showed that gene diversity of plus trees in the seed orchard was  at a moderate level (HE=0.508).Significant inbreeding deviation from the Hardy-Weinberg equilibrium (HWE) was detected (FIS = 0.073). Gene diversity of the infusion populations was comparatively high. It ranged between 0.498 (Janto) and 0.524 (Blankejeren). The FIS was low and insiqnificant deviation from HWE at any genetic infusion populations. The genetic differentiation among the populations (in both seed orchard and genetic infusion) was very low (FST=0.003) indicating that they were genetically related. It is recommended that the infusion populations have a potential to maintain to genetic diversity in the seed orchard.However, Since they are related, it is necessary to find other potential genetic infusion populations that could broaden genetic diversity more effectively in the seed ochard.

Abstract

The incidence of rot disese has inflicted substantial damage to some plantation of Acacia mangium in Sumatera and Kalimantan. As soil-borne pathogen, Ganoderma is commonly found in the tropic and in Indonesia the disease also couse significacant losses to rubber and soil palm plantation. To help control the disease, appropriate identification of the fungi cousing the disease and other field characteritics are critical. Morphological characteistics applied in this study has proved to be effective to identify the pathogen. The disease is predominantly caused by G. philippii. The result of morphological identicication was verified with the DNA sequence of the same fungus and confirm the accuracy of the morphological characteristics. Other fungi that attack root of Acacia mangium are G. australe and Phellinus spp.

Abstract

In Indonesia the demand of genetically improvement seed of teak has been increasing due to the expansion of teak  plantation, while the availability of high quality seed from seed orchard is very limited. A porgeny test of teak at Ciamis West Java that will be converted into a seedling seed orchard is planned to produce improved seed. The trial is laid out in Randomized Complete Block Design (RCBD), consisting of 160 families, 4 treeplots and 10 blocks. The objective of this research is to assess the growth and its variation among the families tested at 5 years old. The best tree height (12 m) was achieved by several seed sources, while the best diameter (14.6 cm) and volume increament (27.21 M3/ha/years) was from Ciamis local seed source. Family affected diameter and volume growth significantly, but not for tree height. Individual tree heriability for stem diameter was low (0.09), while that for stem volume was moderate ().10). The genetic correlation between height and diameter was strong (rg=0.84).
