Abstract

Study on cryopreservation of Indonesian local cultivars and improved  varieties of maize and soybean has never been done. This method may be used for long-term preservation of seeds of maize and soybean. In this study, the method was applied to maize and soybean, Arjuna and Wilis respectively, as a model for preserving germplasm of ortodox seeds. Whole seeds and excised embryos of both varieties were subjected to twomethods of cryopreservation, i.e., two-stage cooling and rapid freezing with or without 15% dimethyl sulfoxide (DMSO) as cryoprotectant solution prior to immersion in liquid nitrogen (-196oC). Results indicated that there was no significant difference between the use of DMSO for both species in terms of viability, although pretreatment in DMSO was slightly reducedthe percentage of viability of both species. Slow freezing to -30oC prior to immersion in the liquid nitrogen could give as high as 76.67% and 51.67% surviving whole seeds of maize and soybean, respectively. Preserving excised embryos of maize in the liquid nitrogen using either slow or rapid freezing significantly reduced the percentage of viability from 20-76.67% to 5-18.33% (four folds) depending on treatments applied. Results also showed that one day or 15 minutes of immersion of samples in the liquid nitrogen gave rise to similar values of viability of maize and soybean, i.e., 20-60% and 20-51.67%, respectively depending on  treatments applied. These results implied that for long-term storage of maize and soybean seeds as they could survive at the rate of 76.67% and 51.67% respectively, the seed can be treated by prefreezing to -30oCwithout the presence of DMSO prior to immersion in liquid nitrogen.

Abstract

Ultisols are a major group of marginal soils extensively found in the upland area of Indonesia. To better understand the potential of the Ultisols developed from claystone and sandstone in the Sasamba Integrated Economical Development Area in East Kalimantan, chemical and mineralogical characteristics of 27 Ultisols pedons consisting of 76 topsoil and 118 subsoil samples were investigated. Besides analysis and interpretation of data, relationships of several soil characteristics wereconstructed using simple regression. The results indicated that Ultisols showed acid to very acid reaction, had low content of organic matter and low base saturation. Soils generally exhibited net negative charge, and the point of zero charge was reached at pH 3.6. Both potential and available phosphates were low, and there was a trend that amorphous aluminum was responsible for phosphate fixation. The low content of exchangeable potassium in topsoil and subsoil indicated a positive correlation with potential potassium. Clay mineral was composed chiefly of kaolinite, with small amounts of illite, vermiculite, and quartz. The domination of kaolinite and low organic matter content causes the soils to have low cation exchange capacity. Soil management in this area should befocused on building up and maintaining soil fertility, and applying appropriate soil conservation techniques to minimize erosion. To obtain sustained productivity, various soil amendments including the use of farm and/or green manure, liming with agricultural lime, and application of rock phosphate and K fertilizers were highly recommended.

Abstract

Farmers in the villages always face the same problem of how to reduce the moisture content of paddy harvest during harvest and postharvest time, especially in wet season. At the farm level, the grain quality is poor and alternative drying using kerosene fuel is very costly. The objective of this research was to design and evaluate the technical and economical  performance of drying paddy using an equipment known as "ABC" dryer.  The dryer uses paddy husk as fuel. The research was conducted at Research Institute for Rice over three years (1993/94-1995/96). The result showed that the "ABC" type dryer could reduce the moisture content of 5 tons wet paddy from 22.25% to 15.03% in 9 hours or an average drying rate of 1.05% moisture content per hour, with drying cost of Rp 18kg-1. Reducing the moisture content further to 11.83% needed 16 hours or average drying rate 0.82% hour-1, with drying cost almost doubled (Rp 32 kg-1). These costs are far below that of kerosene drying, i.e., Rp 30 kg-1 and Rp 60 kg-1 to reach moisture content of 15.03% and 11.83%, respectively. Budget analysis showed that the B/C ratio were 1.57 and 1.84 and the IRR are above 41.26% and 47.42% to reach moisture contentsof 15.03% and 11.83%, respectively. The break even points of "ABC" dryer were 130 and 60 tons, respectively. The milling test showed that the milling rice from the "ABC" dryer had better recovery (milling rendement) and rice quality (head rice) was higher than that of from sun-drying.

Abstract

Mosaic disease of maize and grasses is commonly found in Sulawesi. The symptoms resemble the common mosaic symptoms of virus infection, but the pathogen has not been identified. The objective of this study was to identify the causal agent of the mosaic disease of maize and grasses in Sulawesi. Transmissions of the virus were studied by mechanical inoculation and the insect vector aphid. Serological study was done by using enzyme linked immunosorbent assay (ELISA). Results of mechanical inoculation showed that the disease was caused by a virus which was transmitted from diseased maize and grasses to healthy sweet corn seedlings. The disease was also transmitted by aphid (Rhopalosiphum maidis). Serological study indicated that the virus was closely related to the sugarcane mosaic virus (SCMV). Based on these results, it can be concluded that the maize and grass mosaic disease was caused by SCMV.
