Abstract

Materials used in the research are Ocimum sanctum L, a herbaceous shrub used in traditional medicine displaying as immunomodulator, anti-stress, hepatoprotective, kemopreventif, and anti-inflammatory. A research to identify its ethanol extract of Ocimum sanctum has been conducted, showing that it possesses cytotoxic activities to HeLa cells with IC50 value of 209µg/ml. The aim of this research is to find out activities cytotoxic and apoptotic propagation chloroform fraction of Ocimum sanctum towards HeLa cells line. The extraction was done using maceration using ethanol 96% solvent and fractionation was done using chloroform. Effects of chloroform fraction on HeLa cells were tested using MTT at various dosages (500, 250, 125, 62.5, and 31.25) µg/ml. Cisplatin (Kalbe) was applied as positive control. The effect apoptotic propagation chloroform fraction of Ocimum sanctum on HeLa cells was observed by DNA painting method using orange ethridium bromide-acridine. The test results showed that the chloroform fraction of Ocimum sanctum extract on HeLa cells had an IC50 value of 155.67 µg / ml ± 46.99 and Cisplatin had an IC50 value of 16.81 µg / ml ± 20.37. Double staining test shows the existence of cells undergoing apoptosis. Based on these results, the chloroform fraction Ocimum sanctum has been shown to be cytotoxic and has the possibility of stimulating apoptosis of cervical cancer cells (HeLa). This can be used as a basis for the development of these plants as anticancer agents with specific action targets.

Abstract

Di Indonesia kanker serviks merupakan salah satu kanker penyebab kematian wanita setelah kanker payudara. Herba kemangi (Ocimum basilicum L) banyak digunakan sebagai alternatif pengobatan kanker. Tujuan: mengetahui efek sitotoksik dari herba kemangi terhadap sel HeLa dan sel Vero. Metode: ekstraksi herba kemangi dilakukan dengan maserasi kemudian difraksinasi menggunakan pelarut n-heksan, etil asetat dan air. Ekstrak dan fraksi diuji efek sitotoksiknya menggunakan metode MTT dengan seri konsentrasi 500; 250; 125; 62,5; 31,25; 15,625 µg/mL. Cisplastin digunakan sebagai kontrol positif dan kontrol sel sebagai kontrol negatif. Hasil: aktivitas sitotoksik ekstrak, fraksi n-heksan, etil asetat dan air herba kemangi terhadap sel HeLa menunjukkan nilai IC50 berturut-turut yaitu (156,27; 120,55; 170,17; 186,62) µg/ml dengan indeks selektivitas lebih dari 3 untuk ekstrak dan ketiga fraksi. Kesimpulan: ekstrak dan fraksi herba kemangi memiliki efek sebagai agen kemopreventif terhadap sel kanker serviks.

Abstract

Daun rambusa (Passiflora foetida L.) mempunyai khasiat sebagai antioksidan, antidiabetes dan antikolesterol, akan tetapi masih belum banyak informasi ilmiah tentang potensi daun rambusa sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui golongan senyawa aktif dan aktivitas antibakteri dari ekstrak daun rambusa secara difusi. Daun rambusa diekstraksi secara maserasi menggunakan etanol 96%. Ekstrak yang diperoleh kemudian diidentifikasi kandungan golongan senyawa aktif menggunakan KLT-Bioautografi dan diuji aktivitas antibakteri dengan konsentrasi 20%, 40% dan 60% menggunakan metode difusi agar. Kontrol positif yang digunakan adalah kloramfenikol 0,1% dan kontrol negatif menggunakan DMSO 5%. Hasil penelitian menunjukkan bahwa ekstrak etanol daun rambusa efektif menghambat aktivitas bakteri Klebsiella pneumonia dan Pseudomonas aeruginosa. Konsentrasi ekstrak yang paling besar zona hambatnya yaitu konsentrasi 60% yang dikategorikan kuat. Pengujian aktivitas antibakteri dilakukan dengan metode bioautografi kontak hasilnya menunjukkan terdapat bercak pada kromatogram KLT yang menghasilkan zona hambat. Karakterisasi bercak dilakukan dengan penampak bercak AlCl3 dan diduga bahwa bercak tersebut flavonoid.

Abstract

TOGA (Tanaman Obat Keluarga) merupakan jenis tanaman pilihan yang berkhasiat sebagai obat dengan perawatan yang mudah dan biaya relatif murah. TOGA menjadi alternatif obat keluarga yang aman karena jarang menimbulkan efek samping, mudah diolah dan dikonsumsi untuk pertolongan pertama pada kasus penyakit ringan seperti demam, batuk, atau membantu menjaga stamina. Keberadaan TOGA di lingkungan rumah menjadi sangat penting. Tujuan dari pengabdian ini yaitu memberikan penyuluhan mengenai pemanfaatan TOGA sebagai alternatif pengobatan mandiri dan untuk memberikan informasi mengenai penyakit artritis yang dapat diobati menggunakan TOGA kepada masyarakat RW 21 Kelurahan Nusukan. Metode yang digunakan dalam pengabdian ini adalah dengan memberikan penyuluhan, pelatihan, serta pemberian bibit pohon TOGA untuk ditanam. Target mitra yaitu kelompok ibu-ibu rumah tangga di Wilayah RW 21 Kelurahan Nusukan, Banjarsari, Surakarta. Hasil pengembangan kegiatan di RW 21 dapat meningkatkan motivasi ibu-ibu rumah tangga yang datang untuk lebih memanfaatkan TOGA sebagai pengobatan, meningkatkan pengetahuan masyarakat mengenai artritis dan bahanbahan alamiah yang dapat digunakan untuk mengurangi nyeri artritis.

Abstract

Jengkol leaves ethyl acetate extract shows to have antibacterial activity because it contains saponins, flavonoids and tannins. The use of jengkol leaves ethyl acetateextract directly assessed less well, so that it made cream preparations to make it more convenient to use. This study aimed to obtain the optimum formula of cream of jengkol leavesethyl acetate extract using additional materials polysorbate 80 and sorbitan 80 by Factorial Design method. The Jengkol leaves extract obtained by soxhletation using n-hexane solvent followed by ethyl acetate was then evaporated to obtain a viscous extract. Jengkol leaves extract ethyl asetat cream was made four made in four formulas based on factorial design method. The resulting cream conducted stability tests including organoleptic test, viscosity, adhesiveness, and dispersive power. Optimum formulation parameters based on physical properties, namely: viscosity, viscosity shift, and dispersive power, using Design Ease® Software version 7.1.6. Optimum formula was determined the physical properties and the data were analyzed using t-test. Antibacterial activity was tested by diffusion method. Optimum formula cream of jengkol leaves ethyl acetate extract obtained from proportion polysorbate 80 of 0.44 g and sorbitan 80 of 1.15 g with the physical properties of the response of the optimum formula predictions and experimental result showed no significant difference. Optimum cream showed antibacterial activity presented as inhibition diameter of 14 mm after one day incubation, more potential than formulation 1 (12.8 mm), formulation A (13,8 mm), formulation B (13,2 mm), and formulation AB (13 mm) against Staphylococcus aureus bacteria.

Abstract

Antioxidant is very important to keep human’s health because it can scavenge free radical. Garcinia mangostanaL. which is used by people as traditional herbal medicine and it has traditional name mangosteen is known contain of flavonoid, tannin and saponin. The aim of the experiment was to find out the antioxidant activity of extracts and fractions of mangosteen folium against the DPPH radicals (1,1-diphenyl-2-picrylhydrazyl) to determine the IC50 and classes of compounds that play a role in antioxidant activity assay.Mangosteen leaves were extracted by maceration method using 96% ethanol. Extract was partitionedwith water and n-hexane, and then with ethyl acetate. The antioxidant activity of fraction was tested against DPPH radicals. The test was conducted in 5 series of concentrations by adding 4.0 ml test solutions with 1.0 mlDPPH 0.45 mM. The radicals scavenging activity was measured with spectrophotometer at 517 nm wavelength anddetermined the IC50 value. The experiment used Rutin as positive control.The identification of the class of compounds can be carried out using the Thin Layer Chromatography (TLC) method whose results can be identified by spray reagents or compared with the standard standard. The result of the experiment showed that Mangosteen folium had antioxidant activity with IC50 of ethanol extract, n-hexane fraction, ethyl acetate fraction and water fraction: 17.64 μg / ml, 45.36 μg / ml, 13.71 μg / ml, 12.10 μg / ml respectively. The water fraction has the strongest activity compare to the other with the IC50 is almost near often Rutin’s IC50 is 15.24 μg as the positive control. Abstract

Infectious diseases are still one of the important public health problems to be considered especially in developing countries. Mangosteen leaf (Garcinia mangostana L.) is a well-known plant that is useful as an antioxidant, antibacterial and anticancer. This study aims to determine the chemical content and antibacterial activity of ethanol extract, n-hexane fraction, ethyl acetate and water of mangosteen leaves (Garcinia mangostana L.) on Staphylococcus aureus by diffusion and dilution Mangosteen leaves were extracted by maceration using 96% ethanol then continued by fractionation using a liquid-liquid extraction method. Antibacterial activity was then tested using disc diffusion method with a concentration of 12.5%, 25% and 50%. Positive control used was ciprofloxacin 5μg / disk and negative control of 5% Tween 80. Dilution test was carried out on the most active extract by looking at the MIC and MBC. Identification of compound groups can be done using Thin Layer Chromatography method. The results of antibacterial activity testing showed that the diameter of the largest inhibitory zone in Staphylococcus aureus bacteria was found in the water fraction of mangosteen leaves at 50% concentration with inhibition zone diameter (10 mm). The results of the minimum MIC inhibitory level and the minimum suicide rate of MBC at a concentration of 12.5%. The TLC test results showed that in the extracts and fractions of mangosteen leaves contained flavonoids, saponins and steroids

Abstract

Infectious disease is still one of the public health problems, especially in developing countries. Etiology is generally caused by Staphylococcus aureus and Pseudomonas aeruginosa. Basil (Ocimum sanctum L) is a plant known as lalapan which has antioxidant, anticancer and antimicrobial properties but there is not much scientific information as an antibacterial. This study aims to determine the chemical content and antibacterial activity of ethanol extract, n-hexane fraction, ethyl acetate and basil herb water on the growth of Staphylococcus aureus and Pseudomonas aeruginosa by diffusion and dilution. Herbs are macerated using 96% ethanol. The extract obtained was then suspended with water and then fractionated with n-hexane and ethyl acetate. Basil extract and the fractions obtained were then tested for antibacterial activity at concentrations of 12.5%, 25% and 50% using the diffusion method. The MIC and MBC of the most active fractions can be known using dilution tests with concentrations of 0.78, 1.56, 3.125, 6.25%, 12.5%, 25% and 50%. Ciprofloxacin positive control 5μg/15μl and negative control using sterile 80% 5% tween. Identification of groups of compounds is done by the test tube method. Antibacterial activity test results showed that the zone of inhibition zone in gram-negative bacteria in general tends to be larger than gram-positive bacteria. The largest inhibition zone diameter in the bacterium Pseudomonas aeruginosa in the ethyl acetate fraction basil concentration 50% with a diameter of inhibition zone (10 mm), in the Staphylococcus aureus bacteria found in the ethyl acetate fraction at a concentration of 50% has inhibitory power of (8.6 mm). Abstract

In Indonesia cervical cancer ranks second after breast cancer experienced by women. Stevia is a shrub that is used as a low-calorie sweetener. Stevia also has bioactive compounds such as steviosida, alkaloids, tanin, steroid and flavonoid which are useful as a treatment. Stevia leaf research as an anticancer has not been done much. Therefore, research on the cytotoxic effects of stevia leaves on HeLa cells. The purpose of this study was to determine the IC50 value of stevia leaves on HeLa cells. Stevia leaf extraction was carried out by maceration method using 96% ethanol solvent. The ethanol extract of stevia leaves was tested for its effect on HeLa cells using the MTT method with a concentration series of 500; 250; 125; 62.5; 31.25; 15,625 µg / mL. Cisplastin (Kalbe) is used as a positive control and cell control as a negative control. The results of testing the cytotoxic activity of stevia leaf ethanol extract on HeLa cells showed an IC50 value of 512.94 ± 1.62 µg / ml and IC50 value of Cisplatin 16.16 ± 1.75 µg / ml. This shows that ethanol extract of stevia leaves can be used as a cancer chemo preventive agent that acts to prevent the development of cancer cells

Abstract

Corona Virus Disease-19 (COVID-19) has been declared a pandemic by the World Health Organization (WHO) on March 11, 2020, previously on March 2, 2020 Indonesia has reported 2 confirmed cases of COVID-19. Immunomodulators that can increase the body's resistance to the COVID-19 virus by stimulating the immune system. The purpose of this study was to determine the effect of COVID-19 on the sale of health supplement products in a pharmacy in Surakarta.This research is an analytical observational research. Data was collected from sales records at pharmacies in the 3 months before the COVID-19 pandemic and 3 months at the beginning of the COVID-19 pandemic. Data analysis using statistical tests.Sales of health supplement products for 6 months were 17843 pcs. There were 4773 health supplement products sold before the pandemic, while 13070 pcs were sold during the pandemic. The results of the normality test showed that the data were not normally distributed. The Wilcoxon test results show that the significant value is 0.028 (<0.05), meaning that there is a difference in sales before and after the determination of the pandemic. The conclusion of this study is that COVID-19 has an effect on the sale of health supplement products in a pharmacy in Surakarta.
