Abstract

Penelitian ini bertujuan untuk menentukan kondisi optimum produksi pigmen warna merah dari jamur Penicillium purpurogenum yang diisolasi dari tanah tercemar limbah susu kambing. Produksi pigmen menggunakan metode submerged fermentation dengan media PD Broth. Kondisi lingkungan yang diidentifikasi dalam produksi pigmen merah yaitu solid support (ampas kelapa dan rumput laut), suhu (30oC, 35oC, 40oC, dan 45oC), pH (4-10), waktu inkubasi (1-12 hari), sumber karbon (glukosa, sukrosa, dan pati) dan sumber nitrogen (ekstrak ragi, pepton, dan NaNO3). Pigmen merah yang diekstrak menggunakan akuades dan diukur nilai absorbansinya menggunakan spektronik 20+ pada panjang gelombang 490 nm. Jumlah pigmen direpresentasikan oleh nilai absorbansi pigmen. Hasil penelitian menunjukkan bahwa pigmen warna merah dari jamur Penicillium purpurogenum optimum dengan solid support ampas kelapa, suhu 30oC, pH 5, waktu inkubasi 11 hari, sumber karbon sukrosa dan sumber nitrogen ektrak ragi.Kata-kata kunci: kondisi optimum, produksi pigmen, Penicillium purpurogenum.ABSTRACTThe aim is to determine optimum condition of red pigmen production by Penicillium puurpurogenum that is isolated from from soil contaminated by waste of milk. The method is submerged fermentation with PD broth as media. Environmental conditions in the production of the red pigment are solid support (coconut pulp and seaweed), temperature (30oC, 35oC, 40oC and 45oC), pH (4- 10), the incubation time (1-12 days), carbon source (glucose, sucrose, and starch) and nitrogen source (yeast extract, peptone, and NaNO3). Red pigment is extracted using distilled water and absorbance value is measured using spektronik 20+ at wavelength of 490 nm. The amount of pigment is represented by absorbance value. The results showed that the red pigment of the fungus Penicillium purpurogenum optimum with coconut pulp as solid support, 30°C, pH 5, 11-day incubation period, sucrose as carbon source, and yeast extract as nitrogen source.Key words: Pigment production, Penicillium purpurogenum, optimum conditions.
