Abstract

This research was conducted to determine the impact of work motivation and work environment on employee job satisfaction at PT Indomarco Prismatama (a case study of Indomaret in Depok City), both partially and simultaneously. The research method used was quantitative associative. The sample comprised 67 respondents obtained through a saturated sampling technique. The types of data used included primary and secondary data, collected through observation, interviews, questionnaires, and literature studies. Data analysis techniques included data instrument tests, classical assumption tests, simple and multiple linear regression tests, correlation and determination coefficient tests, and partial and simultaneous hypothesis tests. The results showed that work motivation has a positive and significant effect on employee job satisfaction, with a significance value of 0.000<0.05 and a t-value of 4.145>1.997. The work environment also has a positive and significant effect on employee job satisfaction, with a significance value of 0.000<0.05 and a t-value of 5.987>1.997. Simultaneously, work motivation and work environment have a positive and significant effect on employee job satisfaction, with a significance value of 0.000<0.05 and an F-value of 267.424>3.14. Work motivation and work environment simultaneously also have a determination level of 89.3%.

Abstract

Bacillus subtilis, a causative agent of foodborne illness in bread, cakes, cereals, and cheese leading to symptoms like diarrhea and nausea, was investigated for its role in nosocomial infections, including endocarditis, bacteremia, septicemia, and meningitis. The research focused on three genes: codY, narH, and ureC, which respectively contribute to bacterial survival through biofilm and spore formation, acid resistance, and adaptation to anaerobic conditions. The objective of this study was to evaluate the codY, narH, and ureC genes using the Gradient PCR method for Bacillus subtilis detection in bread and cheese through Real-Time PCR. During the research, bacterial growth was observed with an OD600 of 1.438 in Tryptic Soy Broth (TSB), and colonies of medium size, smooth, cream-coloured, and round shape were successfully isolated on Tryptic Soy Agar (TSA). The DNA template used had a 71.5 ng/µL concentration with an A260/280 ratio of approximately 1.830. The annealing temperature for Gradient PCR used was 53-62°C. The primers successfully amplified codY (175 bp), narH (222 bp), and ureC (153 bp) gene amplicons. The optimal annealing temperature for the primers used was 60°C, as indicated by the presence of a single bright band in electrophoresis. Using these three different genes, testing can also be conducted with the Multiplex PCR method. The next step involves developing a detection kit using optimized primers and annealing temperatures for the identification of Bacillus subtilis in bread and cheese samples using Real-Time PCR.

Abstract

The existence of political parties is an indicator for improving the quality of a democratic country. The Indonesian People's Wave Party (Gelora) is one of the new parties, which is participating in the realm of politics in Indonesia. The institutionalization of a party can be seen through four dimensions as the process of institutionalizing party: the systemic dimension, value identity, autonomy and the public image. Researchers used descriptive qualitative research methods to describe research results in general. The research informants were determined by means of purposive sampling consisting of political party elements and academics or political observers. The results of the study show that the institutionalization process of the regional leadership council Gelora party of West Sumatra has not run optimally. This can be seen from the work program in the form of political activities. The surge party is in a defensive position and indeed does not want to directly engage in political activities in society. Of the four dimensions for measuring political party institutions, the Gelora party is still improving on the systemic dimension and value identity.

Abstract

Abstract

Construction supervision consultants are essential for infrastructure and construction projects that involve public funds. Construction supervision consultants are responsible for ensuring that projects run according to standards, comply with regulations, and reduce the risk of failure or accidents. Negligence committed by construction supervision consultants includes lack of supervision of project implementation to the point of not complying with established quality and safety standards. This study utilizes normative research methods, also known as legal research methods, positive legal research methods, doctrinal legal research methods, and pure legal research methods. During a construction project, the supervising consultant ensures that all work is carried out in accordance with the established technical specifications and quality standards. Ensuring that all projects are executed with professionalism and regulatory compliance, the consultant is responsible for reporting and documenting all important aspects of the project as well as managing changes that may occur during the construction process. The concept of negligence includes various types of non-compliance with technical standards, inaccurate planning, negligence in supervision and monitoring, corruption, and opacity in the construction process. The negligence of construction supervision consultants can negatively impact the success of the construction project itself.
