Abstract

Vibriosis is a disease that often attack shrimp culture, some of the most dangerous types of Vibrio bacteria are V. harveyi. The use of natural materials is an alternative that can be applied to prevent bacterial disease. One of the mangrove species that has an antibacterial compound is A. alba. A. alba has alkaloid, saponins, and flavonoids compounds that can inhibit the function of cytoplasmic membranes and energy metabolism in bacteria. This study was conducted to determine the effectiveness of A. alba leaf extract A. alba in inhibiting V. harveyi disease in vaname shrimp (Litopenaeus vannamei). This research was conducted in several stages, namely extraction of mangrove leaves, both in vivo and in vitro. The highest SR, RPS and MTD values were obtamed at a concentration of 250 mg/l with each value of 80 %, 60 %, 75th respectively hour. Mangrove leaf extract A. alba was able to prevent diseases caused by the bacteria V. harveyi, with the best results at an extract concentration of 250 mg/l where the concentration can significantly higher SR, RPS, and MTD compared to other treatments.

Abstract

Enhalus sp. adalah salah satu jenis lamun dan merupakan tanaman yang sangat penting bagi organisme dan mikroorganisme laut diperairan, salah satunya adalah bakteri karena lamun menyediakan tempat hidup serta penyedia nutrisi pada bakteri. Beberapa hasil penelitian, menyatakan bahwa ekstrak lamun memiliki aktivitas terhadap bakteri yang membentuk biofilm. Terjadinya proses Biofouling diawali dengan terbentuknya film oleh bakteri yang menempel pada permukaan benda yang terendam di dalam laut yang disebut sebagai biofilm. Salah satu cara meminimalisasi biofouling pada umumnya dengan menggunakan bahan beracun yang tidak ramah lingkungan, sehingga perlu dilakukan pencarian alternatif baru untuk menghambat bakteri biofilm. Upaya untuk mengatasi permasalahan dengan menggunakan isolat bakteri endofit lamun merupakan salah satu alternatif ramah lingkungan. Penelitian ini bertujuan untuk mendapatkan bakteri endofit lamun Enhalus sp. yang mampu menghambat pertumbuhan bakteri biofouling. Pada penelitian ini, sampel didapatkan dari pantai Ketapang Pesawaran dan pantai Biha, Pesisir Barat Lampung yang diambil dari bagian akar, batang dan daun lamun. Tahapan penelitian yang dilakukan yaitu pengambilan sampel, isolasi dan purifikasi bakteri endofit lamun dengan metode gores diatas agar, pembentukan bakteri biofilm sebagai bakteri uji pada uji antagonisme, isolasi bakteri biofilm, dan uji antagonis bakteri. Hasil dari isolasi dan purifikasi didapatkan 53 isolat bakteri endofit lamun Enhalus sp. yang akan diuji pada 25 isolat bakteri biofilm. Uji antagonis bakteri endofit lamun dengan kode LEDB2.4 dapat menghambat 4 jenis bakteri biofilm. Besar zona hambat terbesar yaitu terhadap bakteri biofilm dengan kode 3.1 sebesar 18,73 mm. Seagrass Enhalus sp. is a plant that very important for marine organisms and microorganisms in waters, one of which is bacteria because seagrass provides a place to live and nutrients for bacteria. Some research results state that seagrass extracts have activity against bacteria that form biofilms. The occurrence of the biofouling process begins with the formation of a film by bacteria attached to the surface of objects submerged in the sea which is called a biofilm. One way to minimize biofouling in general is to use toxic materials that are not environmentally friendly, so it is necessary to search for new alternatives to inhibit biofilm bacteria. Efforts to overcome the problem by using endophytic bacteria isolates from seagrass are an environmentally friendly alternative. This study aims to obtain the endophytic bacteria of seagrass Enhalus sp. which can inhibit the growth of biofouling bacteria. In this study, samples were obtained from Ketapang beach Pesawaran and Biha beach, West Coastal of Lampung that’s taken from the roots, stems and leaves of seagrass. The stages of the research carried out were sampling, isolation and purification of endophytic bacteria from seagrass, formation of biofilm bacteria as test bacteria in the antagonism test, and isolation of bacterial biofilm, and bacterial antagonist test. The results of isolation and purification obtained 53 isolates of seagrass endophytic bacteria which will be tested on 25 isolates of biofilm bacteria. Antagonistic test of seagrass endophytic bacteria with code LEDB2.4 can inhibit 4 types of biofilm bacteria. The largest inhibition zone was against biofilm bacteria with code 3.1 of 18.73 mm.Â
