Abstract

Oyster mushroom is one of the organisms that can be used as a chitosan source. Chitosan can be produced from chitin through deacetylation. As a chitin derivative compound, chitosan has immense application. In this study, chitin and chitosan were isolated from oyster mushrooms as much as 1.696% and 0.174% from 500 g of dried mushroom. The degree of deacetylation of chitosan is 80.70%. As a polycationic polymer, one of its applications is a metal adsorbent. Cr metal is a heavy metal that is mostly found in leather industrial waste. Chitosan is used as a Cr metal adsorbent to reduce Cr metal content in the solution. This study showed that there was a reduction of Cr metal content as much as 82.75%. Thus, chitosan isolated from oyster mushrooms in this study has the potential to be a bio-adsorbent in order to reduce Cr metal content in leather industrial waste.

Abstract

Cancer is a disease that is the leading cause of death worlwide. Exploration of a natural product containing anticancer agent provides a promising line for research on cancer. One of plant that used in traditional medicine for the cancer treatment is Moringa oleifera. The previous study has reported that the n-hexane fraction of M. oleifera leaves induce apoptosis and cell cycle arrest in T47D cells. Based on the preliminary result, ethyl acetate fraction of M. oleifera (EMO) leaves showed medium cytotoxic activity on T47D cells with IC50 values of 243.58 μg/mL and induced apoptosis cell death. This study was carried out to investigate the anticancer activity mechanism on the cellular and molecular basis of EMO against T47D breast cancer cell line by observing cell cycle progression and Cyclin D1 expression level. Analysis of the cell cycle was performed using flow cytometry and cyclin D1 expression was analyzed using the immunocytochemistry method. The result shows that EMO induced cell cycle arrest in G0/G1 phases. Immunocytochemistry assay showed that the EMO decreased expression of  cyclin D1. This result indicates that EMO has a potential compound to be explored as chemo preventive agent for breast cancer.

Abstract

Gout atau hiperurisemia adalah keadaan dimana terjadinya peningkatan kadar asam urat dalam darah. Xantin oksidase berperan dalam oksidasi xantin dan hipoxantin menjadi asam urat. Salah satu pengobatan kelebihan asam urat adalah menghambat aktivitas enzim xantin oksidase. Penelitian ini menguji aktivitas anti asam urat ekstrak etanol dari buah andaliman kemudian dan menentukan nilai persentase penghambatannya. Hasil pengujian menunjukkan bahwa ekstrak buah andaliman memiliki potensi yang baik untuk menjadi kandidat antihiperurisemia, dengan nilai persentase penghambatannya sebesar 70,67% pada konsentrasi ekstrak sebesar 100 µg/mL.

Abstract

ABSTRAK Pengetahuan akan pencegahan penyebaran penularan virus Covid-19 bagi anak-anak usia dini sangat penting untuk dilakukan karena seringkali anak susah untuk diminta memakai masker dan kebanyakan anak-anak tidak sadar akan pentingnya membersihkan tangan terlebih dahulu sebelum dan sesudah melakukan aktivitasnya baik didalam maupun diluar sekolah. Tujuan kegiatan pengabdian masyarakat ini adalah untuk memberikan pengetahuan kepada siswa SPS PAUD Darussalam Desa Tambakbaya dalam mengantisipasi pencegahan penyebaran penularan Covid-19 melalui gerakan menggunakan masker dan mencuci tangan yang baik dan benar. Metode penyuluhan terdiri dari tahap persiapan, pelaksanaan dan evaluasi. Dimulai dari kegiatan mempersiapkan lokasi, peserta yaitu siswa SPS PAUD Darussalam, Desa Tambakbaya, Kecamatan Cisurupan, Kabupaten Garut, Provinsi Jawa Barat. Pada tahap pelaksaan diberikan dengan beberapa tahapan seperti penjelasan, diskusi, dan praktek. Rincian kegiatan yaitu penjelasan leaflet mengenai cara menggunakan masker dan mencuci tangan dengan baik dan benar dan praktek 7 langkah mencuci tangan menggunakan sabun dengan baik dan benar. Tahap evaluasi kegiatan dilakukan dengan terus membangun komunikasi dan memberikan informasi-informasi yang penting dengan Guru SPS PAUD Darussalam dan juga kepala Desa Tambakbaya. Hasil pengabdian masyarakat yang telah dilakukan menunjukkan bahwa terjadi peningkatan pemahaman siswa terkait pencegahan penularan Covid-19 dengan pemberian edukasi penggunaan masker dan mencuci tangan dengan baik dan benar. Pengukuran kemampuan pemahaman siswa tidak dilakukan secara kuantitatif dengan menggunakan kuisioner melainkan hanya secara kualitatif dari proses diskusi dan tanya jawab yang berlangsung setelah pemaparan materi. Pemahaman siswa dapat dilihat secara langsung dengan meminta beberapa siswa untuk menjelaskan dan memperagakan kembali penjelasan materi yang telah diberikan. Kesimpulan dari penyuluhan ini bahwa terjadi perubahan perilaku lama menjadi perilaku baru dalam pencegahan penularan Covid-19 selama pandemi. Hal ini diharapkan menjadi perilaku baru yang membudaya untuk siswa dalam pencegahan penularan Covid-19. Penyuluhan ini memiliki manfaat besar dalam menimbulkan kesadaran bagi anak usia dini sehingga turut berkontribusi dalam mencegah penyebaran virus SARS-CoV-2 penyebab Covid-19. Kata Kunci : Covid-19, Mencuci Tangan, Penggunaan Masker  ABSTRACT Knowledge of preventing the spread of Covid-19 virus transmission for early childhood is very important because it is often difficult for children to be asked to wear masks and most children are not aware of the importance of cleaning their hands first before and after carrying out their activities both inside and outside school. . The purpose of this community service activity is to provide knowledge to SPS PAUD Darussalam students in Tambakbaya Village in anticipating the prevention of the spread of Covid-19 transmission through the movement of using masks and washing hands properly and correctly. The extension method consists of the stages of preparation, implementation, and evaluation. Starting from the activity of preparing the location, the participants were SPS PAUD Darussalam students, Tambakbaya Village, Cisurupan District, Garut Regency, West Java Province. At the implementation stage, it is provided with several stages such as explanation, discussion, and practice. The details of the activity are an explanation of leaflets on how to use masks and wash hands properly and correctly and practice 7 steps of washing hands using soap properly and correctly. The evaluation phase of the activity is carried out by continuing to build communication and provide important information with the Darussalam SPS PAUD teacher and also the head of Tambakbaya Village. The results of community service that have been carried out show that there is an increase in student understanding regarding the prevention of Covid-19 transmission by providing education on the use of masks and washing hands properly and correctly. The measurement of students' understanding ability is not done quantitatively using questionnaires but only qualitatively from the discussion and question and answer process that takes place after the presentation of the material. Students' understanding can be seen directly by asking several students to explain and demonstrate again the explanation of the material that has been given. The conclusion from this counseling is that there is a change in old behavior into new behavior in preventing Covid-19 transmission during the pandemic. This is expected to be a new, entrenched behavior for students in preventing the transmission of Covid-19. This counseling has great benefits in raising awareness for early childhood so that it contributes to preventing the spread of the SARS-CoV-2 virus that causes Covid-19. Keywords: Covid-19, Washing Hands, Wearing Masks

Abstract

ABSTRAKPandemi Covid-19 yang terjadi saat ini beberapa waktu yang lalu menyebabkan kelangkaan hand sanitizer dipasaran, hal ini membuat resah sebagian masyarakat. Pembuatan hand sanitizer yang dibuat dengan komposisi yang kurang tepat maka efektivitas hand sanitizer dalam membunuh bakteri dan virus tentunya akan berkurang. Pemanfaatan bahan alam sebagai bahan alternatif dalam pembuatan hand sanitizer. Diketahui  beberapa tanaman yang memiliki kemampuan daya hambat terhadap bakteri yang bisa digunakan sebagai zat aktif maupun zat tambahan di dalam produksi hand sanitizer salah satunya adalah lidah buaya. Tujuan dari kegiatan pengabdian ini adalah menghasilkan suatu produk kesehatan yang dapat dimanfaatkan baik untuk diri sendiri, keluarga dan masyarakat sekitar dalam rangka untuk mencegah penyebaran virus Covid-19. Metode yang digunakan dengan memberi penjelasan kegunaan dan keunggulan sediaan hand sanitizer yang terbuat dari bahan alami yaitu lidah buaya (Aloe vera) melalui video dan pamflet. Hasil dari kegiatan pengabdian kepada masyarakat ini dinilai dapat memberikan dampak positif kepada masyarakat Desa tambakbaya, Kecamatan Cisurupan, Kabupaten Garut dilihat dari antusiasme masyarakat Desa dalam menerima pamflet dan video sebagai sumber informasi yang sangat bermanfaat bagi mereka dalam membuat hand sanitizer berbahan dasar alami (Aloe vera) sehingga bisa dirasakan manfaatnya. Kerjasama antara kepala Desa dan aparatur Desa harus terus ditingkatkan agar dampak positif dapat menjadi dampak berkelanjutan. Kata Kunci: Hand sanitizer, antiseptik alami, aloe vera, pencegahan virus corona  ABSTRACTThe Covid-19 pandemic that occurred some time ago caused the scarcity of hand sanitizers in the market, this made some people nervous. Making hand sanitizers that are made with inappropriate compositions will reduce the effectiveness of hand sanitizers in killing bacteria and viruses. Utilization of natural materials as alternative materials in making hand sanitizers. It is known that several plants can inhibit bacteria which can be used as active substances or additives in the production of hand sanitizers, one of which is aloe vera. The purpose of this service activity is to produce a health product that can be used for both yourself, your family, and the surrounding community to prevent the spread of the Covid-19 virus. The method used is to explain the uses and advantages of hand sanitizer preparations made from natural ingredients, namely aloe vera (Aloe vera) through videos and pamphlets. The results of this community service activity are considered to have a positive impact on the people of Tambakbaya Village, Cisurupan District, Garut Regency, seen from the enthusiasm of the Village community in receiving pamphlets and videos as a source of information that is very useful for them in making hand sanitizers made from natural ingredients (Aloe vera) so that the benefits can be felt. Abstract

Tanaman kelor (Moringa oleifera) adalah salah satu tanaman yang telah banyak digunakan dalam industri pengobatan tradisional. Beberapa penelitian telah melaporkan bahwa ekstrak daun M. oleifera memiliki aktivitas antiproliferatif pada beberapa sel kanker, di antaranya terhadap sel kanker hati HepG2, sel kanker paru A549, sel kanker kolon Caco-2 dan sel kanker payudara MDA-MB-231, namun penelitian terhadap sel kanker payudara T47D belum dilakukan. Oleh karena itu, penelitian ini bertujuan untuk mengetahui aktivitas antikanker daun M. oleifera terhadap sel kanker payudara T47D. Ekstrak daun M. oleifera diperoleh dengan cara ekstraksi menggunakan pelarut etanol, kemudian difraksionasi dengan pelarut n-heksana dan etil asetat. Uji sitotoksik yang dilakukan pada penelitian ini menggunakan metode MTT assay. Pengujian terhadap induksi apoptosis dilakukan dengan metode pewarnaan Annexin V-PI dan dianalisis dengan flow cytometry. Kajian molekular juga dilakukan melalui pengamatan protein regulator apoptosis, yaitu Bcl-2 dengan metode imunositokimia. Berdasarkan hasil dari MTT assay, sel T47D yang diberi perlakuan dengan fraksi etil asetat daun M. oleifera menunjukkan aktivitas medium sebagai antikanker dengan nilai IC50 sebesar 243,58 μg/mL. Pengujian terhadap apoptosis menunjukkan bahwa fraksi etil asetat daun M. oleifera dapat menginduksi apoptosis. Hasil pengamatan dengan imunositokimia menunjukkan bahwa pemberian fraksi etil asetat daun M. oleifera mampu menurunkan ekspresi protein Bcl-2. Abstract

Kaempferia galanga L. belongs to the family of Zingiberaceae, an endangered medicinal plant with pharmacology activities. Ethyl-p-methoxycinnamate (EPMC) is an essential phytoconstituent of K. galanga rhizomes. Several studies have reported that EPMC has anticancer activities in several cancer cells, including CL-6 gallbladder cancer cells, HepG2liver cancer cells, MCF-7 breast cancer cells, and Raji lymphoma cancer cells. However, studies on A549 lung cancer and B16 melanoma cancer cells have not been reported. This study aimed to determine the anticancer activity of EPMC against A549 lung cancer and B16 melanoma cancer cells. EPMC was obtained by extraction using n-hexane, then recrystallized with chloroform. The isolate was then analyzed by thin-layer chromatography (TLC), and the structure was characterized by Fourier Transform Infrared (FTIR) and Nuclear Magnetic Resonance (NMR) spectroscopy. Cytotoxic activity was determined under Presto Blue assay. Based on the result, EPMC from K. galanga showed the cytotoxic effect on B16 cells with an IC50 value of 97.09 μg/mL, whereas EPMC showed no significant cytotoxic effect on A549 with an IC50 value of 1407.75 μg/mL. It was concluded that EPMC has potential cytotoxic on B16 melanoma cancer cells, but it showed inactive activity against A549 lung cancer cells. Further molecular mechanism underlying EPMC cytotoxic activity needs to be conducted.
