Abstract

Abstract: Examination of urine leukocyte count usually use urine samples during and urine examination is performed while still fresh because elements in the form of urine begins to break down within 2 hours. The purpose of this study was to determine the effect of storage time on the number of leukocytes urine checked immediately (0 hours) and were kept 30, 60, 90, 120, 150 and 180 minutes at room temperature. Types of Research true experiment to investigate the result of a treatment, research conducted on four samples of urine of patients with diabetes mellitus who meet several criteria, namely blood glucose levels> 200 mg / dl and urine amount leukositnya> 30 cells / mm3, calculated using the Improved Neubauer . The result showed a decrease in the number of stored urine leukocytes 30 minutes = 10%, 60 min = 25%, 90 min = 32%, 120 minutes = 39%, 150 minutes = 43%, and 180 minutes = 51%. Values obtained by linear regression test p = 0.034 (

Abstract

Alternative methods of controlling glucose levels in patients with diabetes is by type of food, either by utilizing yam flour. At the flour contained inulin and resistant starch that can activate the enzyme AMPK-α2. Activation of these enzymes will stimulate glucose transport in skeletal muscle and liver, thus causing a decrease in glucose production. Varying doses of flour is expected to affect the expression of AMPK-α2. This study aims to dosage relationship yam flour (Dioscorea esculenta) with tigkat-α2 AMPK enzyme expression in the nucleus skeletal muscle and liver in mouse models of diabetes mellitus. The study was a quasi-experimental design with Post Test Only Group Design. Rats were divided into 5 (five) groups, healthy mice, the mice with type 2 diabetes, and type 2 groups of diabetic rats with dietary intake yam flour addition of 1.25 g (TG-1:25),), 2.5 g (TG -2.5), 5.0 g (TG-5.0). Yam flour is mixed into the rat diet feed with varying doses. The results showed no significant correlation between the dose of yam flour with AMPK-α2 expression levels in skeletal muscle nuclei (p = 0.312) and liver (p = 0.474) in a mouse model of DM. The need for other studies using other types of food as an alternative arrangement of food for patients with diabetes

Abstract

Early detection of Soil-Transmitted Helminths (STHs) infection in laboratory medical facilities is one of the government's efforts in its elimination. Government regulations recommend Kato-Katz's quantitative method forthe diagnosis of STHs infection. Kato-Katz's domestic product is not availableyet. This research aims to create a laboratory-scale modified Kato-Katz kit for early detection of STHsinfection by fecal egg counts (FECs) in estimating the number of eggs per gram of stool (EPG).An experimental study was conducted to create a resin template, spatulas, and 3M-adhesive tape as a modified Kato-Katz Kit. 120 Kato-Katz thick smear preparations were done from 30 samples for modified and standard Kato-Katz with duplication. The amount of stool for each was weighed. All Kato-Katz thick smear microscopic examination was done at least 60 minutes by identifying the STHs eggs and counting them.The result for both methods showed no differences in EPG for Trichuris trichiura eggs p-value=0.012 (p<0.05), meanwhile the differences for Ascaris lumbricoides eggs p-value=0.156, and Hookworm eggs p-value=0.102 (p>0.05). Under microscopic examination, some A.lumbricoides eggs morphology was not visible because the adhesive in modified Kato-Katz blurry it, and the Hookworm fragile eggshells made it disappear within less than 60 minutes.The study concluded no difference in EPG for T.trichiura eggs by FECs betweenmodified Kato-Katzas this study product with the standard. It is recommended to improve modified Kato-Katz in further study, especially the adhesive tape, before using it as a quantitative toolforearly detection of STHs infection.

Abstract

Wright's stain is a stain for blood preparations consisting of methylene blue and eosin reagents. Strawberries contain anthocyanin dyes and tomatoes contain lycopene, both of which have the function of giving a red pigment, making it suitable for use as a substitute for eosin taking into account the acidity of the pH used by eosin solution with alternative dyes to be investigated. The pH of eosin is 5.2 - 6.0 in an acidic environment, while the pH of strawberries is 5.5 - 6.5 and the pH of tomatoes is 6.0-6.5. The purpose of this study was to determine the morphology of blood cells in the peripheral blood smear (SADT) and the effectiveness of natural alternative dyes used. The type of method in this research is Quasi Experiment. The research material used was normal control blood. The data collection technique was in tabular form and analyzed using a descriptive test. The results of microscopic observations of the wright-stain dye as a control with the morphology of erythrocytes, leukocytes, and platelets, produced good staining with a percentage of 96%, while strawberry and tomato juice as alternative colors obtained poor staining results with a percentage of the same amounting to 12.5%. It can be concluded that the natural alternative dyes of strawberries and tomatoes are able to color some blood cells well and have quite good staining quality in some stained blood cells.
