Abstract

The objective of this research was to investigate the capabilities of bacteria isolated from industrial tanning waste to degrade TCMTB. The bacteria was initialy screened, based on their tolerance to various concentration of TCMTB using paper disk method. Then, those strains were further analyzed in terms of their ability to produce ammonia (NH4+) and sulphate (SO42-). Degradation activity was measured based on remaining residue of TCMTB analyzed using HPLC. The superior strain that showed the highest activity in degradation of TCMTB then were characterized and identified based on phenotypic and 16S rDNA sequence analysis. The result of the experiments showed that four selected strains among seven were choosen based on their high tolerance to various concentration of TCMTB, namely PK1, PK2, PK4 and PK6. All four strains showed the ability to produce ammonia and sulphate but three of which, namely PK2, PK4 and PK6 showed the high capability to degrade TCMTB. One particular strain (PK2) was observed to degrade TCMTB 40.8% within 7 days, but the others were less than 30%. Based on the phenotypic characteristics and 16S rDNA sequence analysis, the best strains (PK2) was identified to be member of genus Pseudomonas.

Abstract

Tlte isolation and identification of sulfat reducing bacteria (SRB) had been performed at Kamojang Geothermal Power Plant. Isolation and identification of sulfat reducer bcrcteria from the soil and corroded steel  bolt obtained from the area of Kamojang Geothermal  power plant.  Four (4)  spesies  of bacterias were obtained from the soil among  them were Desulfotomaculum  spl, Desulfutomaculum  sp2,  Desulfotomaculum sp3, Desulfomonas  pigra,  and  ones  (l)  spesies from coruoded  steel  bolt  Desulfovibrio sp.

Abstract

Tanaman kepel (Stelechocarpus burahol, Hook F&Th.) dikenal oleh masyarakat untukmengobati banyak penyakit. Tujuan dari penelitian ini adalah mengetahui aktivitas antifungifraksi etanol infusa daun Kepel terhadap Candida albicans, dan mengetahui kandungan kimiayang terdapat dalam fraksi etanol infusa daun Kepel (Stelechocarpus burahol, Hook F&Th.).Simplisia daun Kepel diekstraksi dengan metode infundasi kemudian difraksinasi menggunakanpelarut etanol 70%. Uji antifungi dilakukan dengan metode dilusi cair dan zona hambat .Metode dilusi cair menggunakan uji pendahuluan menggunakan konsentrasi bertingkat 50%,25%, 12,5%, 6,25%. Hasil  Uji pendahulan kemudian diperkecil dengan skalaperbandingan 5%(25% b/v, 30% b/v, 35% b/v, 40% b/v, 45% b/v, 50% b/) untuk menentukan KBM. Zona hambatdengan menggunakan cawan petri yang dibuat dengan sumuran. Uji Kromatografi Lapis Tipisdilakukan untuk mengetahui senyawa yang terdapat dalam fraksi etanol infusa daun Kepel.Hasil dari penelitian ini menunjukkan bahwa KBM ekstrak etanol daun Kepel terhadap Candidaalbicans adalah 45% b/vv sedangkan KHM tidak dapat ditentukan karena sampel keruh danberwarna coklat pekat. Zona hambat fraksi etanol infusa daun kepel menunjukan luas zonahambat dengan diameter rata-rata 2cm.Berdasarkan kromatografi lapis tipis menunjukkanbahwa fraksi etanol infusa daun Kepel mengandung golongan senyawa flavonoid. Berdasarkanhasil penelitian ini menunjukan bahwa fraksi etanol infusa daun kepel memiliki aktivitassebagai antifungi terhadap Candida albicans dengan kandungan utama flavonoid.Kata Kunci : Fraksi etanol, daun kepel, Antifungi, Candida albicans
