Abstract

Abstract:Gastric ulcer is focal lesions in the gastric mucosa caused by the influence of gastric acid and pepsin. Seeds of avocado (Persea americana Mill) has a starch that has not been widely used. The study was conducted to determine the antiulcer activity of starch avocado seed in rats induced by acetosal. Antiulcer activity test performed on male Wistar rats were induced by acetosal 450 mg/kgBB for 7 days. Test animals were randomly divided into 6 groups: normal control, negative control (Na-CMC 0.5%), positive control (sucralfate 360 mg/kgBB), starch of avocados seed dose of 0.5 g/kgBB, 1 g/kgBB and 2 g/kgBB. Each group was treated orally for 7 days. On the 8th day of test animals were sacrificed and examined macroscopically hull according to Ashok et al., (2006) and histopathological test. Data in the form of a score of gastric damage further non-parametric statistical analysis. The results showed that administration of the starch avocado seed dose of 2 g/kgBB  have antiulcer activity comparable to the positive control with a score of 0.4 ± 0.42 damage. Hystopathology observations showed that the starchof avocado seed dose of 2 g/kgBB provided protective effects against gastric damage induced by acetosal.Keywords: Persea americana, starch, avocado seed, antiulcer, acetosal  Abstrak: Ulser lambung adalah lesi lokal pada mukosa lambung yang timbul akibat pengaruh asam lambung dan pepsin. Biji avokad (Persea americana  Mill) mempunyai kandungan pati yang belum banyak dimanfaatkan. Penelitian dilakukan untuk mengetahui aktivitas antiulser pati biji avokad pada lambung tikus yang diinduksi asetosal. Pengujian aktivitas antiulser dilakukan pada tikus jantan galur Wistar yang diinduksi asetosal 450 mg/kgBB selama 7 hari. Hewan uji dikelompokkan secara acak menjadi 6 kelompok, yaitu kontrol normal, kontrol negatif (Na-CMC  0,5%), kontrol positif (sukralfat 360 mg/kgBB), pati biji avokad dosis 0,5 g/kgBB, 1 g/kgBB dan 2 g/kgBB. Masing-masing kelompok diberi perlakuan peroral selama 7 hari. Pada hari ke delapan hewan uji dikorbankan dan dilakukan pemeriksaan lambung secara makroskopik menurut Ashok et al., (2006) dan uji histopatologi.  Data berupa skor kerusakan lambung selanjutnya dianalisis statistik non parametrik. Hasil penelitian menunjukkan bahwa pemberian pati biji avokad dosis 2 g/kgBB memiliki aktivitas antiulser sebanding dengan kontrol positif dengan skor kerusakan 0,4±0,42. Hasil pengamatan histopatogi menunjukkan bahwa pemberian pati biji avokad dosis 2 g/kg BB memberikan efek proteksi terhadap kerusakan lambung tikus yang diinduksi asetosal.Kata kunci: Persea maericana, pati, biji avokad, antiulser, asetosal

Abstract

Abstract: Mussels blood (Anadara granosa) is a fishery product that has high economic value. The way of life of the mussels blood settling causes high levels of heavy metals one of which was lead (Pb) and cadmium (Cd) in the body mussels shells. However, there is still lack of Informastion and knowledge in societies about the processing of blood clams result in high levels of heavy metals Pb and Cd. The reduction of heavy metals can be done  using  sequestrant substance that is citric acid which is contained in tomatoes. This reseacrh due to analyze the effect of immersion solution of tomato (Solanum lycopersicum L.) towards decreased levels of the heavy metal Pb and Cd in mussles blood (Anadara granosa). This is an  experimental research which designed by  Complete Random Design with tomatoes filtrate comparation treatment (15%, 25%, and 50%) and soaking time (15 and 30 minutes). Samples were analyzed using atomic absorption spectrophotometer (AAS). The results showed that levels of cadmium in the blood before soaking clams with tomato filtrate was 1,27 mg / kg while for Pb was 0,9603 mg / kg. The levels exceed the TLV established by BPOM (2009). After soaking with tomato filtrate results showed that the highest decreased levels of the immersion of blood clams with tomato filtrate 50% for 30 minutes is equal to 0,31 mg / kg for the concentration of Cd and for the concentration of Pb is equal to 0,51 mg / kg. Data was analyzed with  Anova. The results showed that the soaking solution of tomato affect to decreased levels of heavy metals Pb and Cd in mussels blood  with a value of significance (p <0.05).Keywords: Mussles blood, Anadara granosa, Cadmium, Lead, Atomic Absorption Spectrophotometer  Abstrak: Kerang darah (Anadara granosa) merupakan produk perikanan yang memiliki nilai ekonomis yang cukup tinggi. Cara hidup dari kerang darah yang menetap menyebabkan tingginya kadar logam berat salah satunya adalah Timbal (Pb) dan Kadmium (Cd) di dalam tubuh kerang darah. Informasi dan pengetahuan masyarakat yang masih kurang mengenai pengolahan kerang darah menyebabkan masih tingginya kadar logam berat Pb dan Cd. Penurunan kadar logam berat  dapat dilakukan dengan menggunakan zat sekuestran yaitu asam sitrat yang terkandung dalam tomat. Penelitian ini bertujuan untuk menganalisa pengaruh  perendaman larutan tomat (Solanum lycopersicum L.) terhadap penurunan kadar logam berat Pb dan Cd pada kerang darah (Anadara granosa). Jenis penelitian yang digunakan yaitu penelitian eksperimental dengan rancangan penelitian yang digunakan yaitu Rancangan Acak Lengkap dengan perlakuan perbandingan filtrat tomat (15%, 25%, dan 50%) dan lama perendaman (15 dan 30 menit). Sampel dianalisis menggunakan Spektofotometer Serapan Atom (SSA). Hasil menunjukkan bahwa kadar Cd pada kerang darah sebelum perendaman dengan filtrat tomat yaitu 1,27 mg/kg sedangkan untuk kadar Pb yaitu 0,9603 mg/kg . Kadar tersebut melebihi NAB yang ditetapkan oleh BPOM (2009). Sesudah perendaman dengan filtrat tomat hasil penurunan kadar yang paling signifikan yaitu pada perendaman kerang darah dengan filtrat tomat 50% selama 30 menit yaitu sebesar 0,31 mg/kg untuk kadar Cd dan sebesar 0,51 mg/kg untuk kadar Pb. Data yang diperoleh dianalisis dengan Anova. Hasil penelitian menunjukkan bahwa perendaman larutan tomat berpengaruh terhadap penurunan kadar logam berat Pb dan Cd pada kerang darah dengan nilai signifikasi (p < 0,05).Kata kunci : Kerang darah, Anadara granosa, Kadmium, Timbal, Spektrofotometer  Serapan Atom

Abstract

Singkil leaves (Premna corymbose Rottl et Wild) contain secondary metabolites in the form of flavonoids, saponins, triterpenoid/steroid tannins and phenolic compounds. Phenolic compounds have antioxidant, antitumor, antiviral and antibiotic activity. The drying process of simplicia will have an impact on the content of phenolic compounds. This study aims to determine the effect of the drying method on the total phenolic compounds in singkil leaf extract. This type of research is experimental. Singkil leaf simplicia drying method was carried out using an oven at a temperature of 50°C and air-dried. The extraction method uses maceration with 70% ethanol as solvent. Determination of total phenolic content was carried out by colorimetric method using UV-Vis spectrophotometry with Follin Chiocalteau reagent and gallic acid as standard. The calibration curve was made in 5 concentration series, namely 6,8,10,12, and 14 ppm. The absorbance was calculated at a wavelength of 662.85nm and the readings were used as a determinant of the total phenolic content of the singkil leaf extract. The results showed that the drying method could significantly affect the total phenolic content of singkil leaf extract. The total phenolic content of singkil leaf extract using the wind dry method was 13.37±0.00% while the 50oC oven drying method was 10.64±0.00057%.

Abstract

Singkil leaves (Premna corymbosa) contain secondary metabolites in the form of flavonoids, saponins, tannins and triterpenoids/steroids that can inhibit the growth of Salmonella typhi bacteria. The purpose of this study was to determine the antibacterial activity of the ethanolic extract of singkil leaves in inhibiting Salmonella typhi bacteria in vitro. Extraction was carried out using maceration method with 70% ethanol solvent, then continued with extract quality control and phytochemical screening of flavonoid compounds, saponins, and tannins. Antibacterial activity using disc diffusion method with Mueller Hilton Agar (MHA) media. The antibacterial concentration of the extract was 50%, 60%, 70%, 80%, and 90%; positive control (chloramphenicol 30µg) and negative control (DMSO 10%) with 3 replications. Incubation was carried out at 37°C for 24 hours. The parameter observed was a clear zone formed around the paper disc. The data obtained were then analyzed statistically using Kruskal Wallis followed by Man Whitney with a 95% confidence level. The results showed that variations in the concentration of the ethanolic extract of singkil leaves had a significant effect (p<0.15). The largest inhibition zone was seen at a concentration of 90% at 20.1 ± 1.05 mm with a very strong category. The minimum inhibitory power produced at a concentration of 70% was 17.7 ± 0.88 mm with a strong inhibition category.

Abstract

Yellow passion fruit (Passiflora edulis var. flavicarpa) is a perennial plant in the form of a shrub that can spread or spread. Yellow passion fruit peel contains many flavonoid compounds that have potential for antioxidants, anti-inflammatories and antibacterials. The aim of this study was to analyze the effect of the type of solvent on the amount of yield and total flavonoid content of yellow passion fruit peel extract. The extraction method uses remaceration with 96% ethanol and ethyl acetate as solvent. Extract quality control using organoleptic, drying shrinkage, yield calculation and qualitative test of flavonoid compounds. Testing for total flavonoid levels was carried out using the colorimetric method with AlCl3 reagent by UV-Vis spectrophotometry and the wavelength used was 436.4 nm. The standard uses quercetin. The results showed that the yield of yellow passion fruit peel extract with 96% ethanol solvent was 4.43%w/w while ethyl acetate solvent was 2.57%w/w. The total flavonoid content of yellow passion fruit peel extract showed a significant difference (p <0.05) which was analyzed by independent sample t-test, namely with 96% ethanol of 4.23 ± 0.02 mgQE/gram and 13.02 mgQE/gram of ethyl acetate. 89±0.01 mgQE/gram.
