Abstract

Abaca is a type a banana with high economic value with it stem fiber used in textile and paper industries. As a superior commodity, its number is relatively limited, with the need of a largeplanting area to meet the high market demand. The aim of the research was to observe the abaca banana explants response to various media and Thiamin. The experiment was done at Biotechnology laboratory, UPN “Veteran” Yogyakarta. Treatments were arranged in completely randomized design with 2 factor. The first factor is the growing media: Murashige & Skoog, a half Murashige & Skoog media, Vacint & Went Media and the second factor is the Thiamin concentration: 2 mg/L; 3 mg/L; 4 mg/mL.The results showed there is an interactions on the parameters of planlet height, number of lenghth of root in the combination of Murashige and Skoog and thiamin 3 mg/L medium. Murashige and Skoog medium produced the highest fresh weight and dry weight and Thiamin concentration 3 mg/L produced fresh and dry weight in the highestKey words: abaka, root induction, various media, thiamin

Abstract

The economic value of the Abaca banana is on the trunk which contains fiber for valuable textile and paper industry raw materials. The multipurpose fiber and its prospects are quite good, so the abaca plant gets a lot of attention from various community groups both private, state-owned, cooperative and farmers. Propagation through multiplication humps is very low, planting material is easily damaged in transportation, is not durable and requires large space so that transportation costs are high. One alternative to overcome the provision of healthy, large and fast seedlings is to use seedlings from tissue culture. The aim of the study is to get the right thiamin and leaf fertilizer concentrations at the acclimatization stage. The experimental design used was a 2-factor Complete Randomized Design, Thiamin concentration (1 ml / l, 2 ml / l, 3 ml / l) and leaf fertilizer (0.5 ml / l, 1 ml / l, 1.5 ml / l). The data obtained were analyzed for diversity at 5% level and further tests using Duncan Multiple Range Test (DMRT) level of 5%. The results showed no interaction between thiamin and leaf fertilizer, Thiamin 2 ml / l gave the best results on all parameters and 1.5 ml / l leaf fertilizer could increase plant height and the number of leaves of abaca banana plants.Keywords: Thiamin, leaf fertilizer, abaca banana, acclimatization

Abstract

Mas Kirana Banana (Musa acuminata C.) is one of the most popular tropical fruit plants in the community. The problem of banana tissue culture is browning on the media against phenolic substances that need to be done before the browning inhibitor. This study was conducted to determine the effect of browning inhibitor substance and sucrose on the growth of plantlet banana mas kirana. Research method with RAL (Completely Random Design) two factors + 1 control. The first ingredient consisted of 3 levels, namely polyvinylpyrrolidone 75 g / L, Vitamin C 0,50 ppm, Active Charcoal 1 g / L, while the second factor consisted of 3 levels, sucrose 15 g / L, 20 g / L and 25 g / L Each treatment was repeated 3 times. Data with anova and test with DMRT at 5% level. Browning inhibitors Vitamin C results in the number of shoots, wet weight,  dry weight  and  browning level compared with polyvinylpyrrolidone and activated charcoal. Abstract

Water apple citra is a local fruit that contains a complete mineral. Production of water apple citra can be increased through vegetative propagation, one of them is cuttings. The growth of cuttings can be stimulated using plant growth regulators and growing medium. This study aims to determine the effect of plant growth regulators type and growing medium as well as the interaction between these factors. The research was conducted on March until May 2016 at the garden experiment, Faculty of Agriculture, UPN “Veteran” Yogyakarta. This research used Randomize Completely Block Design (RCBD) factorial pattern with three replications. There were two factors studied, the first factor was the type of plant growth regulators consist of root-up, rootone–F, atonic and without plant growth regulatos, the second factor was growing medium consist of the river sand, floral foam, and river sand+compost. The observed data were analyzed by analysis of variance (ANOVA) and continued by DMRT at 5% real level. The results showed that rootone-F could increase percentage of cutting life and root length. Growing medium of river sand+compost could increase percentage of cutting life. There were an interaction due to the combination of treatment on the observation variable such as root length and root volume.was analyzed by using Duncan Multiple Range Test (DMRT) at 5% level. The result showed that there were an interaction between liquid organic fertilizer various and Trichoderma sp. on the plant height at 42 days (the days after planting), the leaves number at 42 days (the days after planting), the root lenght, the roots number, and the crop weight of red cabbage. The treatment combination of cow urine and Trichoderma sp. gave the best crop weight compared to other treatments. There was no significant effect on vitamin C and vitamin A content in the red cabbage.Keyword: water apple citra, plant growth regulators, growing medium

Abstract

The research was to determine the best combination of Chrysanthemum explant and concentrations of BAP (6 - benzyl amino purine), the effect of the explant material and the effect concentrations of BAP (6 - benzyl amino purine) in vitro culture by Murashige and Skoog medium. The experimental methode was Completely Randomizes Design (CRD) which consisted of two factors. The first factor is the explant material, consisting of three standards are: P1 = node, P2 = shoots, P3 = leaf. The second factor is the concentration of BAP which consists of four levels, namely: B1 = 0.5 ppm, 1.0 ppm = B2, B3 = 1.5 ppm, B4 = 2.0 ppm. The results showed there were significant interaction on fresh weight, dry weight, number of shoots and length of shoots. The treatment combination between bud explants with BAP 0.5 ppm, showed the best results on fresh weight, number of shoots and long shoots. Use of explant shoots, can accelerate the current growth of shoots, expanding the number and length of root buds. The addition of plant growth regulators BAP 0.5 ppm in the tissue culture medium, can spur the rapid growth of shoots, root number and length of root best.Key words : in vitro, chrysanthemum, kinds of eksplant, the concentration of BAP

Abstract

Gladiolus flower can be multiplied by using corm or cormel. The multiplication of this flower using plating material takes a long time, about 3,5-6 months. By using in vitro method, the seed of this flower can be produce in relatively short time. The aim of this research was to determine the interaction of gibberellins on any kind of media (MS, B5, and MS + vitamin B5), to determine the perfect combination of concentration of Gibberelin, and to determine the perfect type of media that can be use for induction of root and shoot of gladiolus cormel. The research design used was a Completely Randomized Design (CRD) of the factorial pattern with 3 replications. The first factors (concentration of Gibberellin) G1: 1 ppm, G2: 2 ppm, dan G3: 3 ppm, the second factors (type of media) M1: MS , M2: B5 (Gamborg) , M3: MS + B5 vitamin. The result showed that the addition of various concentration of Gibberelin (GA3) and any type of media had no significantly improve the growth of gladiol cormel explant, by addition concentration of Gibberelin 2 ppm has significantly improved fresh weight of planlet and percentage of life, by addition B5 media, has significantly improved length of root and growth of shoot, and by using MS media with B5 vitamin has significantly improved the growth of shoot, height and fresh weight of planlet.Keywords: Gladiol, gibberellin, plant media, in vitro
