Abstract

Fasciola gigantica diagnosis usually performed by detection of worm eggs presence in the feces,but this conventional method has many disadvantages. Early diagnosis (early detection) cannot be performed in conventional methods because the worms in the host's body began to lay eggs at the age of 8–12 weeks of patency. The current detection method that is based on antibody­antigen reactions using excreted/secreted (ES) liquid by adult F. gigantica, is believed to be used for the early detection of fasciolosis. This study aimed to characterize the antigenic components of F. gigantica extretory/secretory products that could be used as a vaccine candidate development for early fasciolosis diagnostics. ES products were separated by PEG4000 at various concentrations (8%, 16%, 24%), then precipitates (pellets) obtained were dialyzed and characterized using SDS­PAGE and Western blotting. Results from SDSPAGE showed that there were 18 proteins bands with 7–70 kDa molecular weights. Western blotting on pellets derived from PEG separation at various concentrations affirmed that the proteins of 50, 25 and 20 kDa were antigenic at 8% PEG concentration, the 25 kDa and 50 kDa were antigenic at 16% PEG concentrations and the 25 kDa was antigenic at 25% PEG concentration

Abstract

Parasitic worms (Helminths) are the one of health problems related to the parasites investment that is often found in native chickens. This study aimed to identify the diversity of worm parasites found in native chickens in Jember Regency. The sampling procedure for native chicken excreta was carried out randomly. The excreta samples taken were 150 samples around Jember Regency. Identification of the diversity of worm species was carried out by examining worm eggs in excreta samples using the sedimentation method at BBVet (Balai Besar Veterinary) Wates, Yogyakarta. The parasitic worms identified in the excreta samples of native chickens were worms from the Nematode class consisting of Capillaria sp. (24.67%); Heterakis sp. (10.67%); Strongyloides sp. (3.33%); and Ascaridia sp. (2.67%).

Abstract

ABSTRAK Penggunaan energi oleh sektor transportasi yang mencapai lebih dari 30 persen dari total penggunaan energi nasional yang hampir seluruhnya (92%) bersumber dari bahan bakar minyak (BBM) selain masalah pada pasokan BBM juga berdampak buruk bagi lingkungan, sehingga harus ada upaya dalam mencari energi alternatif baru terbarukan, salah satunya adalahbiogas. Penelitian ini bertujuan untuk melihat kualitas biogas dari bebegai feses ternak seperti feses, sapi perah, sapi potong, kambing, domba, ayam broiler, ayam petelur dan feses keclinci. Pengamatan meliputi nilai pH dan temperatur harian, lama waktu pembentukan gas dalam masa fermentasi, lama produktivitas gas dari pembentukan gas pertama kali serta lama nyala api dalam dari hasi pembentukan gas harian. Biogas di produksi menggunakan digester portable dengan total waktu pengamatan selama 30 hari masa fermentasi dan produksi gas. Hasil Penetlitian menunjukan rata-rata nilai pH yang diapatkan dalam waktu 30 hari dari pH 6,2- pH 8. Temperatur rata-rata hasil pengamatan berada di angka 23-36 OC. Hasil uji ANOVA pada pada masing masing pengamatan yaitu pada lama pembentukan gas, lama produktifitas gas dan lama nyala api di dapatkan hasil yang signifikan yaitu f hitung <0,05. Hasil uji lanjut Tukey’S B pada masing-masing pengamatan di dapatkan hasil terbaik pada limbah feses sapi perah dengan data lama pembentukan gas (11,2 ± 1,3) hari, lama produktifitas gas (10,8± 1,9) dan lama nyala api adalah (20,6± 1,9) menit. This study aims to determine the quality of biogas from various animal feces such as feces, dairy cows, beef cattle, goats, sheep, broilers, laying hens and small animal feces. The observations included daily pH and temperature values, the length of time for gas formation during the fermentation period, the length of gas productivity from the first gas formation and the length of the internal flame from the daily gas formation. Biogas is produced using a portable digester with a total observation time of 30 days during fermentation and gas production. The results showed that the average pH value obtained within 30 days was from pH 6.2 to pH 8. The average temperature observed was 23-36 OC. The results of the ANOVA test on each observation, namely the duration of gas formation, the duration of gas productivity and the duration of the flame, obtained significant results, namely f count <0.05. Tukey'S B continued test results for each observation obtained the best results on dairy cow feces with data on the length of gas formation (11.2 ± 1.3) days, long gas productivity (10.8 ± 1.9) and duration of flame fire was (20.6 ± 1.9) minutes.
