Abstract

ABSTRACT           Introduction: Dengue is the most important mosquito-borne flavivirus disease. The number of Dengue cases in Indonesia in 2010 range from 150,000 cases with the deaths of around 1,317 people. Huge number of cases have made Indonesia was the first ranked as the state with the highest Dengue cases in the ASEAN region and the world's second ranking after Brazil. The drugs  or antibiotics that can be administered effectively to cure this disease has not been found yet. Many study have been done and some that have been reported include viral RNA synthesis inhibitors, protein inhibitors of NS3 helicase and protease and inhibitors that inhibit Dengue virus maturation. Curcumin have preventive activity against several viruses: vasicular stomatis ( VSV ), HSV 1 and 2, parainfluenza - 3, reovirus - 1, feline corona virus, feline herpes virus . Curcumin also known have ubiquitin proteasome inhibition system was able to decrease the production of Japanese ensefalitis virus.Objectives: This study aims to determine safe concentrations of curcumin against vero cells (cytotoxic test results) and know the Dengue-2 antiviral potency of curcumin.Methods: Including quasi-experimental study. The anti viral potency of curcumin seen from the result of immunocytochemistry Streptavidin Biotin Peroxidase Complex (SBPC).  Data were analyzed by ANOVA.Results: The results showed that secure concentrations from cytotoxic of curcumin against vero cells is 6.25 ppm. The calculation of positive rate from immunocytochemistry in vero cells infected by Dengue - 2 incubation 1 and 3 days were the result is significantly different than the control.Conclusion: The secure concentration of curcumin against vero cells was 6.25 ppm and curcumin was able to lower the positive rate due to Dengue-2 infection.Key Words : Dengue virus, Curcumin, Immunocytochemistry.

Abstract

The improvement and development of science and technologycal in health area must be followed with improvement of requirement of test animal would especially white mouse (Mus musculus albino/mencit). Usage of white mouse (mencit) because of relatively easy in handling, the size relatively small, the price relatively cheap, once birth can reach 16 - 18, have similiarity in circulation system of human and doesn't have ability to vomit because having spillway in bouncing up, that is why white mouse used for drug trial research. This research expected to obtain stable white mouse colony in laboratory to fulfill requirement of white mouse as subject for research.Research was started in March-November 2008 in Loka Litbang P2B2 Banjarnegara. This research was elementary research with observational design. Data were collected from white mouse growth observation.Result of observation showed reproduction of white mouse after 5 month, from 6 white mouse increased become 17 ( increased 2 times more ), with number of deaths 11,76 %. This white mouse survive until 12 months, but the average was 6 month. Oldest mencit until research ends ( November 2008) ranges from 8 months. Daily observation data showed that white mouse can bear children until 6/pregnance. According to literature study mouse can bear children average 6 8/pregnance. Generaly baby mouse was around 1 gram, heavily borns depend on type ( strain) of white mouse. Result of white mouse weight showed increase of white mouse's body weight every 2 weeks was 8,6 grams. Observation data of development white mouse showed after 4 days the white mouse hair was seen, specially moustache that was clearly seen, on 5 day whole body was white. At the age of 10 days ear opened, part of other body like external mamilla and genitals become explains looked to be. At the age of 12 days eye started opens and active walking.

Abstract

Treatment of Dengue patients today are to relieve symptoms because specific therapies and effective anti-viral drugs have not been found. Curcumin is known has anti-viral activity, anti-inflammatory, anti-oxidant, anti-parasitic and anti-cancer. Curcumin has a weakness that is sensitive to acidity (pH) and light, so curcumin analogues that ispentagamavunon-0 (PGV-0) was made in order to obtain better anti viral activity. The purpose of this study was to determine the effect of curcumin and PGV-0 on vero cells infected by Dengue-2. This research was conducted in the laboratory of parasitology Faculty of Medicine of Gadjah Mada University in January-September 2013. Experimental studies using RT-PCR test to determine the presence of Dengue-2 antigen. Dengue-2 virus propagated in C636 cellsand then infected in vero cells to further treated with curcumin and PGV-0. The incubation period of Dengue-2 infections performed for 1 and 3 days. The results of RT-PCR showed Dengue-2 antigen was seen in the 3-day infection period in the treatment of curcumin. This result indicates PGV-0 has potential antiviral better than curcumin.

Abstract

ABSTRAKPenyakit demam Dengue endemik di lebih dari 100 negara di dunia. Obat anti virus Dengue efektif belum ditemukan danpengendalian vektor dinilai kurang efektif, sehingga diperlukan upaya pencegahan dengan vaksinasi. Vaksin Dengue yangideal adalah murah, mencakup 4 serotipe, efektif dalam memberikan kekebalan, cukup diberikan sekali seumur hidup, aman,memberi kekebalan jangka panjang, stabil dalam penyimpanan dan stabil secara genetis (tidak bermutasi). Beberapakandidat vaksin yang telah dan sedang dikembangkan oleh para peneliti di seluruh dunia adalah tetravalent live attenuatedvaccine, vaksin Chimera (ChimeriVax), vaksin subunit dan vaksin DNA. Vaksin Dengue dipandang sebagai pendekatan yangefektif dan berkesinambungan dalam mengendalikan penyakit Dengue. Tahun 2003 telah terbentuk Pediatric DengueVaccine Initiative (PDVI), yaitu sebuah konsorsium internasional yang bergerak dalam advokasi untuk meyakinkanmasyarakat internasional akan penting dan mendesaknya vaksin Dengue. Konsorsium vaksin Dengue Indonesia saat iniberupaya mengembangkan vaksin Dengue dengan menggunakan strain virus lokal.

Abstract

ABSTRAKToksoplasmosis merupakan zoonosis yang disebabkan oleh Toxoplasma gondii, mempunyai penyebaran yang luas padamanusia dan hewan piaraan maupun satwa liar. Penularan secara horisontal pada manusia terutama disebabkan karenadaging hewan/ternak yang terinfeksi T. gondii atau ookista pada makanan atau minuman yang terkontaminasi feses kucing.Oleh karena itu, perlu diketahui kucing liar dengan seropositif T. gondii. Jenis penelitian ini adalah potong lintang danlaboratorik serologi pada bulan Mei–Oktober 2013. Sampel sebanyak 22 ekor kucing liar yang berasal dari pasar induk,rumah sakit dan kompleks pertokoan kelurahan Semampir, Banjarnegara. Pemeriksaan serologis menggunakan FELISAimunostik. Hasil penelitian menunjukkan kucing liar dengan seropositif IgG T. gondii sebanyak 40,9% (9 dari 22 ekor).Kucing liar di kompleks pertokoan Kelurahan Semampir berpeluang lebih besar memaparkan T. gondii.

Abstract

A study on Bionomic of Anopheles mosquitoes as an approach to control the population of malaria (case studyat Kalibening Village, Sukoharjo Subdistrict, Wonosobo Regency, Central Java) had been conducted in July-October 2006. The objective of the study is to identify the bionomic of malaria vector. The result of this study revealed five species of Anopheles such as An. maculatus, An barbirostris, An. balabacensis, An. aconitus dan An. vagus. Three of them (An. aconitus, An. maculatus, An. balabacensis) represent as malarias vector in Java. Unfortunately these results were not supported by the ELISA test, since it proved that they are proven to be negative malaria vector. An. maculatus was found predominantly. The parous rate of An. maculatus was 39,89% (77 mortalities out of 193). Mosquoito age of An. maculatus ranged from 0.25-2.35 days. Thus, they were hardly to be a vectors. However, the situation might be different, under some circumstances, e.g. seasonal change. On the other way around, An. balabacensis and An. maculatus had a high density. An. Maculatus were mostly found in the ground pool around Salak plantation. Their bitting graphics (located at in door and out door) always occured at the same time. High in July then slowing down in Agust up to September and increased again in October whether those which were located in the cage were low in July, increasing in August up to September and slowing down in October. The peak density of An. maculatus which was biting on man occured at mid night (00. 00- 01. 00) and early in the morning (04. 00-05. 00) outdoors and indoors respectively.

Abstract

Some kind of mosquitoes can transmit desease through their biting. Some of them are Anopheles that transmit malaria, Aedes aegypti and Aedes albopictus transmit Dengue Haemorrhagic Fever (DHF) and chikungunya, some of mosquitoes from Culex, Aedes, Anopheles and Mansonia genera that transmit filaria, etc. To study the mosquitoes diversity , the collection of mosquitoes in different species was needed.In order to increased the mosquitoes collection in LokaLitbang P2B2 Banjarnegara, collection in endemic area were conducted. One of selected area as the location of mosquitoes collection was Blora districtThis survey was observational. Samples were taken with purposive sampling method that was all mosquitoes found during the study. Secondary data collected from health officer and public health center (puskesmas), observing the environment and social activities and also entomological survey. Result of entomological survey were various kind of mosquitoes, in Central Java Province, Blora district i.e.: Culex quinquefasciatus, Culex vishnui, Aedes aegypti, Anopheles vagus and Anopheles indefinitus.
