Abstract

Currently, obstacle in malaria treatment is the emergence of resistance to antimalarials. Brotowali (Tinospora crispa (L.)) is one of the medicinal plant which efficacious as antimalarials. This study aimed to obtain information on the structure prediction of chemical compounds that potential as antimalarials from F.EtOH.4 brotowali stems with heme polymerization inhibition mechanism using Huy method. F.EtOH.4 was fractionated using column chromatography with silica gel 60 as a stationary phase and dichloromethane-methanol eluent. F.EtOH.4.2.1.a has the highest heme polymerization inhibitory activity and chemical structure identification with UV spectroscopy, FTIR and LCMS. The IC50 value of F.EtOH.4.2.1.a was 253.46 ppm and IC50 from chloroquine sulfate was 178.74 ppm as a positive control. Identification of F.EtOH.4.2.1.a isolates based on MS spectrum was the possibility of borapetoside D compounds with molecular formula C33H46O16 and molecular weight of 698. FTIR spectrum results indicated aromatic and alcohol groups and UV spectrum results indicated chromophore groups.Keywords: borapetoside D,  heme polymerization, malaria, Tinospora crispa (L.) ABSTRAKMalaria adalah penyakit yang disebabkan oleh parasit Plasmodium falcifarum dan ditularkan oleh nyamuk Anopheles betina. Saat ini, kendala yang dihadapi dalam pengobatan malaria adalah timbulnya resistensi terhadap antimalaria sehingga mendorong penelitian untuk mencari antimalaria baru. Salah satu tanaman obat yang berkhasiat sebagai antimalaria adalah brotowali (Tinospora crispa (L.)) dengan kandungan alkaloid, glikosida pikroretosid, pikroretin, berberin, palmatin dan kolumbin. Penelitian sebelumnya menunjukkan bahwa ekstrak etanol 70% dan F.EtOH.4 batang brotowali memiliki aktivitas penghambatan ß-hematin tertinggi dengan metode Basilico. Penelitian ini bertujuan untuk memperoleh informasi prediksi struktur senyawa kimia yang berpotensi sebagai antimalaria dari F.EtOH.4 batang brotowali dengan mekanisme penghambatan polimerisasi heme menggunakan metode Huy. F.EtOH.4 difraksinasi menggunakan kolom kromatografi dengan fasa diam silika gel 60 dan eluen diklorometana-metanol. Hasil fraksinasi F.EtOH.4.2, F.EtOH.4.2.1 dan F.EtOH.4.2.1.a masing-masing dilakukan pengujian aktivitas penghambatan pelimerisasi heme. F.EtOH.4.2.1.a adalah fraksi yang mempunyai aktivitas penghambatan polimerisasi heme tertinggi dan dilakukan identifikasi struktur kimia dengan Spektroskopi UV, FTIR dan LCMS. Nilai IC50 dari F.EtOH.4.2.1.a sebesar 253,46 ppm dan IC50 dari klorokuin sulfat sebesar 178,74 ppm sebagai kontrol positif. Identifikasi isolat F.EtOH.4.2.1.a berdasarkan spektrum MS adalah kemungkinan senyawa borapetoside D dengan rumus molekul C33H46O16 dan bobot molekul 698. Hasil spektrum FTIR menunjukkan adanya gugus aromatik dan alkohol dan hasil spektrum UV menunjukkan adanya gugus kromofor.Kata kunci:          borapetoside D, malaria, polimerisasi heme, Tinospora crispa (L.)

Abstract

This study deals with endophytic microbes, known as a potential source of secondary metabolites. The microbes (F) were isolated from Cinchona pubescens Vahl and fermented in Phoma media, observing the pH, biomass growth and secondary metabolites of alkaloids for 14 days. The result obtained showed that the pH was between 4 and 5, the biomass growth relative stable after 8 days and the production of alkaloids reached its maximum also after 8 days (32.81 mg/l).

Abstract

Four alkaloids, brucine, brucin N-oxide, colobrine N-oxide, and strychine N-oxide were isolated from the bark of Strychnos ligustrida Bl. (Loganiaceae). Their structures were determined by spectroscopic data analysis, in particular by the Nuclear Magnet Resonance ('H and 13C NMR) and 2D NMR ('H-'HCOSY, 13C-'H COSY) and mass spectroscopy (MS).

Abstract

It is well known, that temu putih (Curcuma zedoaria Berg.), a rhizome of the Zingiberaceae, is oftenly used as medicine. In this connection, the potential secondary metabolites as anticancer and antimicrobes are produced by endophytic microbes in the plant. In this research, five endophytic microbes from the Curcuma zedoaria’s rhizome were fermented to produce compounds as antimicrobes.

Abstract

Some endophytic microbes were isolated from medicinal plant Artemisia annua collected of Tawangmangu and Cibinong. Microbe isolate were made on corn meal malt agur supplemented with chloramphenicol and nutrient agar supplemented with nystatin from tissue subjected after the triple surface sterilization procedure. The potencies of these isolates to produce-terpene compounds were screenned by phytochemistry analysis, thin layer chromatography (tlc) and high performance liquid chromatography (hplc) after fermentation in four different medium. The results showed that 6 isolates from 17 microbe isolates can produce terpene compounds.

Abstract

Caesalpinia major Dandy ex Excell is one of the traditional medicinal plants from Nusa Tenggara Timur Province (Flores island, Ruteng) which the local people called as “Dekar". The root of this plant is used as anthelmintic, anti rheumatic, back-ache and also as tonic. Chemical structure elucidation of isolated compound from C. major caesaldekarin A has been done and it's based on ultraviolet, infrared, nuclear nagnetic resonance and mass spectrometry: To know the correlation between proton and proton, proton and carbon, carbon and carbon, NMR two dimensional (2D) have been also interpreted by the data of 'H-'H COSY, 13C-'H COSY, COLOC, HMBC and INADEQUATE. The stereochemistry of the relative structure of caesaldekarin A is determined by 2D NMR NOESY.

Abstract

The leaves of yacon (Smallanthussonchi folius [Poepp. &Endl.] H. Robinson) or in Indonesia known as “Daun insulin” (Insulin leaf), scientifically has various medicinal properties and one of them as antidiabetic. The aim of this research is to know α-glucosidase enzyme inhibitor compound in the ethanol 96% extract of yacon leaves. Fractionation of ethanol extract subjected to column chromatography (SiO2; dichloromethane-methanol-water = 5 : 5 : 1) gave 5 fractions (Fr -1 ~Fr – 5). The results of the -glucosidase enzyme inhibition activity test in vitro against the five fractions obtained the 3rd fraction as the most active of 87.84 %. Purification of the 3rd fraction with preparative thin layer chromatography (SiO2; n-BuOH-ethanol-water = 4 : 1 : 2.2) gave pure isolate as Fr 3-1. Identification of pure isolates Fr 3-1 was carried out by interpreted spectra of Ultraviolet, Fourier Transform-Infra Red, Mass Spectra (from LC-MS) and compared data on the chemical shift of the Nuclear Magnetic Resonance (proton and carbon) and determined as Nystose (GF3) compound. The results of the -glucosidase enzyme inhibition activity showed that Fr 3-1 has IC50 of 21.36 ppm

Abstract

Isolation and structure elucidation of an alkaloid, a bioactive compound from the roots of the Indonesian medicinal plant Kemuning, Murraya exotica (Rutaceae), including the brine shrimp lethality test (BSLT) have been carried out. The bioactivity test using Artemia salina, isolation and purification of the ethyl acetate extract performed by column chromatography gave yellow crystals. Based on ultraviolet, infrared, nuclear magnetic resonance (1H-&13C-NMR) and gas chromatographymass spectra the isolated bioactive compound is an indole alkaloid typical to yuehchukene.

Abstract

The leaves of tea benalu (Scurrula oortiana (Korth) Danser) were extracted with nhexane, ethyl acetate, methanol, and water respectively. The antioxidant activities were measured using 1,1-diphenyl-2-picrylhidrazyl (DPPH) methods. The result showed that the isolated compound has scavenging effect with IC50 92.22 ppm. Identification by interpretation of ultraviolet, infrared, and nuclear magnetic resonance spectra showed that the isolated compound was cathecin.
