Abstract

Durian seeds are one type of seed that contains glucomannan. The aim is to determine glucomannan levels in the durian seed. The study was designed using a completely randomized design (CRD). The extracted glucomannan was determined by the spectrophotometry method at various temperature and hydrolysis time. The crude extract of glucomannan was hydrolyzed using 3 M HCl for 70, 80, 90, 100, and 110 minutes at 70, 80, 90, 100, and 110°C. The highest glucomannan concentration of 39.60% was obtained at a hydrolysis temperature of 100°C for 100 minutes. The analysis of the FTIR spectrophotometer shown that the vibration of the C-O-C functional group (glycoside bond) was obtained at the wavelength number of 1020.34 cm-1 which is a characteristic of glucomannan. Abstract

Cork Fish (Channa striata) is a freshwater fish that was spread throughout most of Indonesia and it contains albumin that was beneficial to the human body. Albumin was easily dissolved in water during the cooking process both boiled and steamed. This study aims to determine the influence of boiling and steaming times at 100oC against albumin content of cork fish that lost and left in fish meat. The times of boiling and steaming that was used to processing the fish were 10, 15, and 20 minutes. The albumin concentration that lost and left in for the boiling and the steaming process was determined by the biuret test that used a UV-Vis spectrophotometer at albumin maximum wavelength of 530 nm. The results showed that albumin content that lost for 10 minutes boiling process (62 mg/10g) was smaller than the other boiling times and albumin left in fish meat was 164 mg/10g. The same result was also obtained at 10 minutes steaming process which results left in albumin content of 167 mg/10g and lost albumin of 62 mg/10g. The albumin content that lost or dissolved in water for the steaming process was smaller than the boiling process, therefore the steaming process is more efficient than the boiling process at cork fish processing. Abstract

The research about the fermentation of protein-free grated coconut with Aspergillus niger to produce of crude lipase enzyme has been carried out. This study aims to determine the effect of water content on fermentation medium and the incubation time which results lipase enzyme extract with the highest activity. This study used a Completely Randomized Design (CRD) consisting of 2 factors, i.e. variations of water content in medium of 40%, 50%, 60%, and 70% and incubation times of 48, 96, 144, and 192 hours. The parameters observed were lipase enzyme activity produced in each treatment. The results showed that the highest medium water content was 70% with the lipase enzyme activity 4.33 µmol/mL.minute and the best incubation time was 96 hours with the lipase enzyme activity of 0.83 µmol/mL.minute. Abstract

Research on the antioxidant activity of the local onion roots of Palu (Allium cepa Var Aggregatum L.) with various polarity solvents has been carried out. This study aims to determine the potential of Palu local shallot root extract as an antioxidant and determine the IC50 value of the Palu local shallot root antioxidant activity. The method applied in this research is maceration using n-hexane, ethyl acetate, and ethanol. The extracts from each solvent were tested for their antioxidant activity by the DPPH method. The results of the study showed that the IC50 value in n-hexane extract, ethyl acetate extract, and ethanol extract were 579.98 ppm, 518.28 ppm 315.83 ppm and as a comparison used ascorbic acid obtained IC50 of 53.69 ppm. Based on the results obtained, the best antioxidant activity is in polar solvents (ethanol extract) compared to other solvents. Abstract

Cellulose from Pennisetum purpureum has been used as the main ingredient in the synthesis of carboxymethyl cellulose (CMC). The purpose of CMC synthesis from the cellulose of Pennisetum purpureum is to obtain CMC compounds with the best degree of substitution (DS) value and the highest yield. The study was conducted using a completely randomized design with a factorial pattern consisting of two independent variables, namely the concentration of trichloroacetic acid (10%, 20%, and 30%) and reaction time (3 and 4 hours). The results showed that the CMC with the highest DS value (0.839) was obtained using 20% trichloroacetic acid with a reaction time of 3 hours. The highest CMC yield was 30.15% which was obtained using 30% trichloroacetic acid with a reaction time of 4 hours. Keywords: Pennisetum purpureum, carboxymethyl cellulose, substitution degree.

Abstract

Coconut pulp contains up to 18.2% protein which is one of the wastes from coconut oil processing. Protein from coconut pulp can be used in the manufacture of protein concentrates which are widely used in the food industry. The purpose of this research is to determine the concentration of NaOH that can produce the yield and protein content of coconut pulp protein concentrate. Coconut pulp protein was extracted with NaOH at various concentrations of 0.5, 1, 1.5, 2, and 2.5 M, then continued with salting out using salt (NH4)2SO4 with a saturation of 65%. The crude protein content of coconut pulp protein concentrate was determined by the Kjeldahl method. The results showed that the use of 0.5 M NaOH resulted in the highest protein content of coconut pulp protein concentrate, which was 71.30% with a yield of 14.42%.

Abstract

Liquid smoke contains acid, carbonyl, and phenolic compounds, so it is very appropriate to be used as a preservative. Traditional smoking can be replaced with liquid smoke because it is safer and more practical. The study of the use of coconut shell liquid smoke has been used in skipjack fish to determine the effect of liquid smoke concentration and storage time on dissolved protein content and total bacteria in smoked fish. The quality of smoked fish was determined by two treatments, namely the variation in the concentration of liquid smoke (2, 3, 4, and 5%) and storage time (2, 4, 6, 8, and 10 days). The results showed that the best concentration of liquid smoke to produce smoked skipjack fish is 3% with a water content of 62.41% and dissolved protein content of 17.58%. The maximum storage time of smoked skipjack tuna is 6 days with a total plate count (TPC) value or total bacteria 1.8 x 105.

Abstract

Black sea cucumber is one type of sea cucumber whose protein content is quite high at 74-82%, not yet widely used by the community as a source of protein. One way to use it is to process the protein into a protein hydrolyzate using the bromelain enzyme which was isolated from pineapple stems. This study aims to determine the concentration of the bromelain enzyme extract and the optimum hydrolysis time to the value of the hydrolysis degree of the hydrolyzate of black sea cucumber protein. Protein hydrolysates are produced by variations of the crude bromelain concentration of 9%, 10%, 11%, and 12% (v/v) and hydrolysis time of 120, 180, 240, and 300 minutes. The results showed that the optimum degree of hydrolysis was obtained with the use of bromelain extract with a concentration of 12% with a value of 11.43% and a hydrolysis time of 240 minutes with a value of 19.17%.

Abstract

Coconut pulp wasted from processed coconut oil has a protein content of up to 18.20%. The potential coconut pulp protein can be processed in protein concentrate which has many benefits. This research was conducted to determine the effect of NaOH and (NH4)2SO4 concentrations in isolating protein and producing protein concentrates that have the highest yield and protein content. Isolation of coconut pulp protein was carried out using NaOH at several concentrations, namely 0, 0.2, 0.4, 0.6, 0.8, and 1 M, while the protein concentrate was prepared using the salting out method with a saturation level of the ammonium sulfate salt of 55%, 60%, and 65%. Analysis of protein content was carried out using the Kjeldahl method. The use of NaOH 0.2 M and (NH4)2SO4 with a saturation level of 60% was the chosen treatment with crude protein content and yield of 75.63% and 33.83%, respectively.
