Abstract

Background: The application of Regulation of the Minister of Health Number 72 of 2016 is very important to be implemented in every Pharmacy Installation in a hospital, one of which is in the clinical pharmacy service section, namely dispensing sterile preparations to avoid the possibility of medication errors. Objective: The research aimed to find out how the implementation of Regulation of the Minister of Health Number 72 of 2016 stipulated in the basic guidelines of aseptic techniques related to space, equipment, and the implementation of dispensing sterile preparations with its practice in Hospital "X" Gianyar, Bali. Methods: This research used Sequential Explanatory design by combining quantitative and qualitative methods. Results: The results of quantitative research with a checklist found the percentage of completeness of the room and equipment by 100%, while the conformity of the practice with the guidelines of 76.48% is appropriate. The results of qualitative research with triangulation matrices that practice is by the guidelines. Conclusion: Dispensing sterile preparations at the Hospital "X" is following the basic guidelines of aseptic techniques that refer to Regulation of the Minister of Health Number 72 of 2016. Abstract

Penuaan kulit adalah proses biologi kompleks yang dipengaruhi oleh adanya paparan berlebih radiasi ultraviolet (UV) dari sinar matahari. Photoaging kulit dapat diatasi dengan agen anti-photoaging yang mempunyai aktivitas menghambat matrix metalloproteinases (MMPs). Degradasi kolagen kulit dikatalisis oleh enzim MMPs yaitu MMP-1. Senyawa ellagic acid dilaporkan secara in vitro memiliki aktivitas antioksidan melalui induksi SOD dan GSH yang berperan dalam peredaman ROS. Penelitian ini bertujuan untuk mengetahui potensi ellagic acid sebagai agen anti-photoaging melalui penentuan afinitas dan mekanisme molekuler terhadap enzim MMP-1 secara in silico. Uji in silico dilakukan secara docking molecular dengan tahapan yaitu preparasi protein target enzim MMP-1 dilakukan mengunaankan software Chimera 1.11.1. Validasi dan docking dilakukan dengan menggunakan Autodock tools 1.5.6. yang dilengkapi dengan program Autodock4 dan Autogrid4. Metode docking molecular dapat dinyatakan valid apabila nilai RMSD (root mean square distance) yang diperoleh < 3 Å. Hasil pengujian molecular docking menunjukkan bahwa ellagic acid dengan protein target enzim MMP-1 memiliki ikatan yang lebih stabil dibandingkan dengan native ligant RS2 dengan protein target enzim MMP-1. Sehingga berdasarkan hasil penelitian ini dapat diprediksi bahwa ellagic acid memiliki aktivitas sebagai agen anti-photoaging karena memiliki afinitas dengan protein target enzim MMP-1.
