Abstract

Background: Sea cucumber had been acknowledged to have some medical properties Sticophus hermanii and Holothuria atra are species of sea cucumber which has been known to have antifungal properties thus potentially explored as therapeutic agent in oral candidiasis. Purpose: The aim of this study was to examine the antifungal property Sticophus hermanii and Holothuria atra extract against Candida albicans and its cytotoxicity to human gingiva-derived mesenchymal stem cell. Methods: The study was an experimental laboratories research with post test only control group design. Methanolic extract of Sticophus hermanii and Holothuria atra in concentrations of 1%, 0.5%; 0.25%; 0.13%, 0.07%; 0.03%, 0.02% and 0.01%; were tested its cytotoxicity on gingiva-derived mesenchymal stem cell. Cell viability were measured by MTT assay. The antifungal property against Candida albicans was tested by disk diffusion method. Data were analyzed by ANOVA followed by LSD. Results: Extract of Sticophus hermanii showed no cytotoxicity in all concentrations (p>0.05), while Holothuria atra showed toxicity in the concentration of 1% and not cytotoxic in the concentrations below (p<0.05). Both sea cucumber extract could inhibit the growth Candida albicans, in vitro, proved by the clear zone around the disc in all concentrations (p<0.05). Conclusion: Stichopus hermanii and Holothuria atra extract had the antifungal effect against Candida albicans. Sea cucumber extract were not cytotoxic togingiva-derived mesenchymal stem cell in the concentration of Sticophus hermanii ≤ 1% and Holothuria atra ≤ 0.5%.Latar belakang: Teripang telah diketahui mempunyai berbagai khasiat medis. Sticophus hermanii dan Holothuria atra adalah spesies teripang yang telah diketahui mempunyai sifat anti jamur sehingga santat potensial untuk diekplorasi sebagai agen terapeutik pada infeksi di rongga mulut. Tujuan: Tujuan dari penelitian ini adalah untuk meneliti sifat anti jamur ekstrak Sticophus hermanii and Holothuria atra terhadap Candida albicans dan sitotoksisitasnya terhadap stem sel mesenkimal yang berasal dari gingiva manusia. Metode: Penelitian ini merupakan penelitian eksperimental laboratoris dengan rancangan post test only control group design. Ekstrak metanol Sticophus hermanii dan Holothuria atra pada konsentrasi 1%, 0,5%; 0,25%; 0,13%, 0,07%; 0,03%, 0,02% and 0,01% diuji sitoksisitasnya terhadap stem sel mesenkimal yang berasal dari gingiva. Viabilitas sel diukur dengan menggunakan metode MTT. Sifat anti jamur terhadap Candida albicans diuji dengan metode difusi. Data dianalisis dengan ANOVA dan LSD. Hasil: Ekstrak Sticophus hermanii tidak menunjukkan adanya toksisitas pada seluruh konsentrasi (p>0,05), sedangkan Holothuria atra menunjukkan adanya toksisitas pada konsentrasi 1% dan tidak toksik pada konsentrasi di bawahnya (p<0,05). Kedua ekstrak mampu menghambat pertumbuhan Candida albicans in vitro yang ditunjukkan dengan adanya zona jernih disekitar disk pada semua konsentrasi (p<0,05). Simpulan: Ekstrak Stichopus hermanii dan Holothuria atra mempunyai daya anti jamur terhadap Candida albicans. Ekstrak teripang bersifat tidak toksik terhadap stem sel mesenkimal yang berasal dari gingiva masing-masing pada konsentrasi Sticophus hermanii ≤ 1%, dan Holothuria atra ≤ 0,5%.

Abstract

Background: Oral candidiasis is the most prominent oral fungal infection with Candida albicans (C. albicans) as 75% of ethiologic factor. Golden sea cucumbers (Stichopus hermanii) have been consumed by Asian community as folk medicine. It has been known to have antifungal and immunomodulator agent thus potential to be explored as treatment in oral candidiasis. Purpose: The aim of this study was to examine the potency of Stichopus hermanii extract as oral candidiasis treatment. Method: The study was an experimental laboratories research with post test only control group design. Thirty male wistar rats were divided into 5 groups i.e negative control, positive control and 3 treatment groups. Oral candidiasis condition were induced by spraying C. albicans suspension on dorsal tongue of wistar rats, once in 2 days for 14 days. The treatment groups were given Stichopus hermanii extract on the dose of 4.25 ml/kgBW, 8.5 ml/kgBW, 17 ml/kgBW once daily for 14 days. The expression of anti C. albicans antibody and TNF-α were examined by immunohistochemistry on ephitelial tongue. Data was analyzed by Manova and LSD test. Result: Anti C. albicans antibody expression were higher in positive control group than in negative control group while TNF-α expression were lower in positive control group than in negative control group (p<0.05). Treatment with Stichopus hermanii extract on all doses decreased the expression of anti C. albicans antibody and increased the expression of TNF-α (p<0.05). Conclusion: Stichopus hermanii extract decreased the expression of anti C. albicans antibody and increased the expression of TNF-α in epithelial rat tongue.

Abstract

Background: There is strong evidence to support the claim that periodontitis may be more prevalent among diabetic individuals. Collagen degradation represents one of the key events in periodontal destructive lesions. The level of matrix metalloproteinase 8 (MMP-8) and tissue inhibitor of metalloproteinase 1 (TIMP-1) are key to periodontal collagenolysis and associated with the severity of periodontal inflammation and disease. Host modulatory therapy has been proposed as a treatment for periodontal diseases. Sardinella longiceps (lemuru) fish oil containing polyunsaturated fatty acids (PUFAs), including omega 3 and 6, has been shown to possess therapeutic anti-inflammatory and protective properties effective against inflammatory diseases, including periodontitis. Purpose: The study aimed to examine the effect of dietary supplementation of Sardinella longiceps fish oil on protection against periodontal destruction resulting from the expression of MMP-8 and TIMP-1. Methods: Wistar rat samples are divided into four groups: a negative control group and three groups receiving Sardinella longiceps fish oil treatment (4 ml/ weight (Kg), 8 ml/ weight (Kg) and 16 ml/ weight (Kg). One week before treatment, all groups were administered with streptozotocin (STZ) 65 ml/ weight (Kg) and nicotinamide 110 ml/ weight (Kg) to induce diabetic conditions. Immunohistochemistry slides of periodontal tissues were prepared after three weeks of treatment. The expression of MMP-8 and TIMP-1 was counted using the HSCORE index, data was analyzed by means of non-parametric methods using Kruskal-Wallis, and Mann-Whitney tests. Results: Statistical analyses confirmed a significant increase in MMP-8 expression and a reduction in TIMP-1 expression in the negative control group compared to the treatment group (p<0.05). Meanwhile, the treatment group showed a significant reduction in MMP-8 expression and a marked increase in TIMP-1 expression, with the best result produced by the administering of 16 ml/ weight (Kg) Sardinella longiceps fish oil to the treatment group (p<0.05). Conclusion: Dietary supplementation of Sardinella longiceps fish oil can protect against periodontal destruction under diabetic conditions, by decreasing MMP-8 expression and increasing TIMP-1 expression.

Abstract

Background: Periodontitis have a bidirectional relationship with diabetes mellitus, both can increase oxidative stress that trigger an increase of pro-inflammatory cytokines, such as TNF-α. Sea cucumbers have anti-inflammatory component that act to inhibit the release of inflammatory mediators. Hyperbaric oxygen therapy reduce oxidative stress. Purpose: The aim of this study is to investigate the effect of the sea cucumber powder and hyperbaric oxygen therapy on the expression of TNF-α in rats with diabetes mellitus induced by P.gingivalis bacteria. Materials and Methods: The study was an experimental laboratories research with factorial design. Twenty wistar rats were divided into 5 groups. K0 negative control, K1 positive control. K1-K4 groups were induced with streptozotocin for diabetes condition and P.gingivalis bacteria for periodontitis condition. K2 group was treated with seacucumber 3% for 7 days, K3 with OHB 2.4 ATA 3 x 30’ interval 5’ for 7 days, and K4 group was treated with combination of sea cucumber and OHB. At the 51 st day all rats were sacrified, then the expression of TNF-α on periodontal macrophages were examined by immunohistochemistry stain. Data were analized by Kruskal Wallis followed by Mannwhitney. Result: Expression of TNF-α were increased in K1 (11.50±1.291) compare K0 (p<0.05). Sea cucumber treatment, OHB, and combination treatment decreased expression of TNF-α significantly in amount of 2.50±0.577 (K2), 8.25±2.217 (K3), 3.00±0.816 (K4). Conclusion: Sea cucumber powder and OHB therapy affected the expression of TNF-α in rats with diabetes mellitus induced by P. gingivalis bacteria.

Abstract

Background: Diabetes mellitus (DM) with Periodontitis cause severe alveolar bone resorption. Osteoprotegerin prevent osteoclastogenesis process so that inhibited alveolar bone resorption. Stichopus hermanii powder and hyperbaric oxygen therapy (HBO) have content and good effects in wound healing. Purpose: The aim of this reseach was to analyze the effect of Stichopus hermanii powder and HBO therapy in the increasing of osteoprotegerin expression on DM with periodontitis. Materials and Methods: The research was an experimental laboratories post test only control group design. Twenty Wistar rats were divided into 5 groups. K0 was negative group while K1 was positive group K1-K4 groups were induced with 65mm/kg STZ single dose and 2ml bacterium P. gingivalis., K2 was treated with 3%golden sea cucumber powder in gel form. K3 was treated with OHB therapy 2.4 ATA for 7 days and K4 given a combination of both. Osteoprotegrin expression on osteoblast of alveolar mandible bone were examined by immunohistochemical staining. Data were analyzed by Kruskal-Wallis and Mann-Whitney test. Result: Kruskal-Wallis result showed significant differences from each treatment group (p <0.05). Mann-Whitney test showed the decreasion in the expression of osteoprotegerin between K1(2:50 ± 0.577) compare with K0 (p<0.05). Golden sea cucumber powder, HBO therapy and both combination had increased osteoprotegerin expression.significancy in K2(8.25 ± 1.258), K3 (5.75 ± 0.957) and K4(12.50 ± 2.082) (p<0,05). Conclusion: Stichopus hermanii powder 3% and 2.4 ATA OHB therapy for 7 days increased the expression osteoprotegrin on DM with periodontitis.

Abstract

Background: Enterococcus faecalis is one of the bacteria which have ability to form biofilms that can cause persistent endodontic infections. Biofilm is a communities of well-organized bacteria adhere to the surface and coated by layer of extracellular polysaccharide matrix. The extracts of mangrove Acanthus ilicifolius leaves have been known to have potentially bioactive compounds as antibacterial such as saponins, alkaloids, terpenoids, and tannins. Purpose: The aim of this study was to determine the antibacterial effectiveness of mangrove Acanthus ilicifolius leaves extracts to Enterococcus faecalis biofilm. Materials and Methods: This study was true experimental laboratory with post test only control group design. The sample in this study was Enterococcus faecalis, divided into 6 groups consisted of control positive (NaOCl 2.5%) group, and 5 treatment groups of mangrove Acanthus ilicifolius leaves extract with different concentration of 60 mg/ml; 70 mg/ml; 80 mg/ml; 90 mg/ml; and 100mg/ml and be repeated 8 times. Enterococcus faecalis bacteria were cultured in mediaTSBglu incubated for 24 hours. Amount of 0.1 ml of bacteria Enterococcus faecalis with concentrations of 106were loaded on microtiter plate then stained with crystal violet. Biofilms checked by measuring optical density (OD) using an ELISA reader. Data were analyzed by One-way ANOVA followed by Post Hoc test. Result: Acanthus ilicifolius leaves extract decrease the OD of Enterococcus faecalis biofilm (p<0,05) in all groups. Conclusion: The extract of mangrove Acanthus ilicifolius leaves extract have antibacterial efficacy against Enterococcus faecalis biofilm.

Abstract

The aging process is a process of slowly disappearing the ability of tissues to repair themselves or replace and maintain their normal functions. Tooth decay, tooth mobility, caries, halitosis, gingivitis, gingival recession, loss of periodontal attachment, and alveolar bone are changes in periodontal tissue that are commonly found in the elderly. If this is not treated, it can cause mobility and loss of teeth which will interfere with the function and activity of the oral cavity and will have an impact on the quality of life. Dental and oral health problems become one of the problems that have negative implications for overall health that in the end, it affects the quality of life in the elderly. Based on this analysis, in implementing the Tridarma of Higher Education, especially community service, the strategy is to increase public access to dental health services. One form of community service that can be carried out is promotive and preventive efforts. This activity is an educational approach to increase the ability and participation of the elderly community in maintaining dental health which can affect a person's quality of life.
