Abstract

ABSTRAK                                                                        Penelitian bertujuan untuk mengetahui efektivitas nira aren sebagai pengencer alternatif dalam proses pembekuan (kriopreservasi) semen kambing boer.Kriopreservasi semen kambing boer menggunakan pengencer tris-gliserol-kuning telur (P1 73-7-20%), nira aren-gliseol-kuning telur(masing-masing P2 74-6-20%, P3 73-7-20%, dan P4 72-8-20%) dan andromed (P5 tanpa mengandung kuning telur dan gliserol). Parameter evaluasi meliputi motilitas, viabilitas, dan membrane plasma utuh setelah pengenceran, ekuilibrasi dan thawing.  Evaluasi motilitas pasca thawing menunjukkan P5 52% berbeda nyata (P<0.05) dengan P1 42%, selanjutnya P5 dan P1 berbeda sangat nyata (P<0.05) dengan P2 8%, P3 6% dan P4 12%.  Viabilitas pasca thawing menunjukkan P5 65,4% tidak berbeda nyata (P>0,05) dengan P1 61,8%, akan tetapi P5 dan P1 berbeda sangat nyata (P<0.05) dengan P2 26,2%, P3 29,8%, dan P4 34%.  Membran plasma utuh (MPU) pasca thawing menunjukkan P5 66,2% tidak berbeda nyata (P>0,05) dengan P1 65,4%, akan tetapi keduanya berbeda sangat nyata (P<0.05) dengan P2 39%, P3 38%, dan P4 36,2%.  Disimpulkan kriopreservasi semen kambing boer dengan pengencer nira aren dan gliserol pada konsentrasi berbeda belum dapat dipergunakan sebagai sumber bibit berdasarkan standar nasional Indonesia.Kata Kunci : Kambing boer, semen, nira arenABSTRACTThe experiment was conducted to determine the effect of sugar palm juice as alternative extender for cryopreservation process of boer semen.Tris-glycerol-egg yolk (P1 73-7-20%), Sugar palm juice-glyserol-egg yolk (P2 74-6-20%, P373-7-20%, dan P4 72-8-20%), and andromed (P5) used as a extender  in the cryopreservation process of boer semen.  Sperm motility (%), live sperm (%) and sperm membrane integrity (%) were recorded after diluted, equilibration and freeze-thawing.  Result of post thawing motility showed that P5 52% was significantly different (P <0.05) with P1 42%, then P5 and P1 were significantly different (P <0.05) with P2 8%, P3 6% and P4 12%. Viability after thawing showed P5 65.4% was not significantly different (P> 0.05) with P1 61.8%, but P5 and P1 significantly different (P <0.05) with P2 26.2%, P3 29.8 %, and P4 34%. Abstract

Sugar palm juice (Arenga pinnata Merr.) can be used as semen extender because its various nutrientscontent which were needed for spermatozoa preservation. The objective of this research was to examineeffectivity of sugar palm juice as swamp buffalo semen extender in artificial insemination (AI) program.Semen of swamp buffalo were collected by using an artificial vagina. Fresh semen were evaluated anddivided in equal volume into four tubes and diluted with lactose extender containing 20% egg yolk (lactoseor control), 85% sugar palm juice + 15% egg yolk (PJEY15), 80% sugar palm juice + 20% egg yolk (PJEY20),and 75% sugar palm juice + 25% egg yolk (PJEY25), respectively. Diluted-semen were preserved inrefrigerator at 5oC, and quality of spermatozoa including percentages of motile spermatozoa, livespermatozoa, and intact plasma membrane (IPM) were evaluated every day for four days. A total of 13female swamp buffaloes were estrous synchronize with PGF2a, and six buffaloes were inseminated withlactose and seven buffaloes were inseminated with PJEY20. Results of this research showed that at dayfourpreservation, mean percentages of motile and live spermatozoa for lactose (36.67 and 51.83%) andPJEY20 (35.83 and 51.5%) were significantly (P<0.05) higher than PJEY15 (31.67 and 44%) and PJEY25 (30 and 45.33%), but no difference significant between treatment for IPM parameter. Pregnancy rate forlactose was 66.67% and 71.43% for PJEY20. Percentage of birth for lactose and PJEY20 were 66.67% and71.43%, respectively. In conclusion, sugar palm juice could be used as an alternative extender in semenpreservation process and AI of swamp buffalo.

Abstract

Ethylene diamine tetraacetic acid (EDTA) is a molecular structure that binds to heavy metals andother toxins which needed to protect the spermatozoa during the cryopreservation process. The objectiveof this research was to examine EDTA in maintain the quality of garut ram frozen semen. Semen werecollected from mature Garut ram by artificial vagina. Fresh semen were evaluated and divided in equalvolume into three tubes and diluted with Tris extender containing 20% egg yolk (TEY-20) as control, TEY-20 + 0.01% EDTA (EDTA0.01), and TEY-20 + 0.02% EDTA (EDTA0.02), respectively. Semen was loaded in 0.25 mL mini straw with the concentration of 100 million motile spermatozoa. Semen was equilibrated at5oC for three hours, then frozen and stored in liquid nitrogen container. Quality of spermatozoa includingpercentages of motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) wereevaluated after diluting, equilibrating, and thawing. Data were analyzed using completely randomizeddesign with three treatments and five replicates. Means were compared significant difference test at 0.05significant level. Results of this research showed that mean volume, color, consistency, pH, mass movement,spermatozoa concentration, percentage of MS, percentage of LS, percentage of abnormal spermatozoa,and percentage of IPM of garut ram fresh semen were 0.87 mL, cream, thick, 6.98, 3, 4,296 million cell/mL,75%, 86.33%, 4.4%, and 85%, respectively. Percentages of MS, LS, and IPM after thawing in control (42.5,52.5, and 52.17%) was not significantly difference with EDTA0.01 (44.17, 52.5, and 52.5%) and EDTA0.02(39.17%, 51%, and 48.33%). In conclusion, the addition of EDTA in Tris extender can maintain quality ofGarut ram frozen semen, and its suitable using in artificial insemination program. Addition of EDTA cannot enhancing quality of garut ram frozen semen.
