Abstract

Red color coverage (RCC) is a commercial trait developed and refined to improve the appearance of many ornamental fish commodities. In neon tetra, the status of variation of RCC is not yet investigated or reported. This study aimed to analyze the RCC variation of cultured neon tetra as a basis for breeding strategies. A total of 900 neon tetras (standard length, SL, of 2.29 ± 0.16 cm) were collected from Bojongsari, Curug, and Pondok Petir fish farms located in Depok Districts, West Java. All fish were relocated and reared in a fish farm specialized in culturing neon tetra for two weeks using nine aquariums with photoperiod set up of 12 hours bright and 12 hours dark. The RCC traits were determined according to the percentages of RCC length (%LRCC), RCC width (%WRCC), and RCC area (%ARCC) and quantified using the digital image method. The result showed that the RCC varied by sex, size, and original location (P<0.05) in a low coefficient of variation (1.89%-11.41%). The RCC values in the male group were higher than that of females based on %LRCC and %ARCC parameters (P<0.05). Males had the highest %LRCC at size LXL, which was correlated with SL (r 0.25, P<0.1), of females at M size. The %LRCC values of the neon tetra population from the Bojongsari farm were higher than those from the other locations. Based on these findings, breeding strategies of the RCC traits should consider sex, size, and population (farm location) variations. Specifically for neon tetra, this strategy should be based on selecting the SL or %LRCC parameter of M for females and LXL for males.

Abstract

Betta rubra merupakan salah satu spesies ikan cupang alam endemik dari Aceh. Keberadaannya yang hampir dinyatakan punah sebelum ditemukan kembali pada tahun 2007. Tujuan dari penelitian ini adalah mengkaji keragaman genetik dan potensi genetik dari ikan Betta rubra dari tiga generasi yang sudah dibudidayakan untuk perbaikan genetik di Balai Riset Budidaya Ikan Hias (BRBIH), Depok, Jawa Barat, Indonesia. Jumlah sampel yang digunakan pada populasi G-0 adalah enam ekor, sedangkan pada populasi G-1 dan G-2 masing-masing 10 ekor. Ikan uji yang digunakan diambil sirip ekornya untuk analisis secara genotipe dengan randomly amplified polymorphic DNA (RAPD) menggunakan primer yaitu OPZ-9, OPB-6, dan OPZ-13. Sebelum diambil sirip ekornya ikan terlebih dahulu difoto di atas millimeter block untuk data truss morfometrik (fenotipe). Hasil menunjukkan ikan Betta rubra populasi alam (G-0) memiliki nilai heterozigositas 0,1872 dan derajat polimorfisme 47,06% yang lebih rendah dibandingkan generasi G-1 dengan heterozigositas 2,421 dan derajat polimorfisme 64,71%. Populasi G-2 memiliki nilai heterozigositas 0,1577 dan derajat polimorfisme 44,12%. Koefisien keragaman secara fenotipe populasi G-1 memiliki variasi lebih tinggi dibanding populasi G-0 dan G-2. Hubungan kekerabatan antara G-1 dengan G-0 dan G-2 berbeda nyata (P<0,05), sedangkan hubungan antara G-1 dengan G-2 tidak berbeda nyata (P>0,05), sehingga antara populasi G-0 dan G-2 membentuk cluster terpisah dengan G-1. Keragaman genetik pada tiga generasi Betta rubra memiliki pola yang sama baik secara fenotipe maupun genotipe.Betta rubra is one of the endemic species of Betta fish from Aceh. The fish was almost declared extinct before it was rediscovered in 2007. The purpose of this study was to examine the genetic diversity and genetic potential of Betta rubra from three generations which have been reared for genetic improvement at the Research Institute for Ornamental Fish Culture, Depok, West Java, Indonesia. The number of fish for G-0 population used in the study was six fish whilst G-1 and G-2 populations were 10 fish. Tail fins from each fish were sampled for genotype analysis using randomly amplified polymorphic DNA (RAPD) using primers OPZ-9, OPB-6, and OPZ-13. Before tail fin collection, the fish was photographed on a millimeter block for truss morphometric data measurement (phenotype). The results showed that the Betta rubra wild population (G-0) had heterozygosity of 0.1872 and polymorphism of 47.06% which were lower than the G-1 population with heterozygosity of 2.421 and polymorphism of 64.71%. The G-2 population had heterozygosity of 0.1577 and polymorphism of 44.12%. The phenotype coefficient of variation in the G-1 population higher than the G-0 and G-2 populations. The kinship relationship between G-1 with G-0 and G-2 was significantly different (P<0.05), while the relationship between G-1 and G-2 was not significantly different (P>0.05). This research concludes that the populations of G-0 and G-2 have formed a separate cluster to G-1. The genetic diversities in the three Betta rubra populations have similar phenotype and genotype patterns.

Abstract

Peacock goby (Tateurndina ocellicauda) merupakan salah satu ikan hias yang mendiami perairan gambut di wilayah Papua hingga bagian Timur Papua New Guinea. Data dan informasi mengenai aspek biologi ikan peacock goby untuk mendukung kegiatan budidaya masih terbatas. Penelitian bertujuan untuk mengkaji aspek pemijahan, perkembangan embrio dan larva ikan peacock goby. Induk ikan yang digunakan yaitu sembilan ekor induk jantan (bobot tubuh 0,69 ± 0,08 g) dan enam ekor induk betina (bobot tubuh 0,65 ± 0,03 g) dipelihara dalam wadah plastik berukuran 20 cm x 25 cm x 15 cm. Perbandingan induk jantan dan betina yaitu 1:1 (perlakuan A) dan 2:1 (perlakuan B), dengan ulangan sebanyak tiga kali. Wadah pemijahan dilengkapi dengan potongan pipa PVC berbentuk setengah lingkaran sebagai tempat menempelnya telur. Telur diinkubasi pada wadah penetasan berbeda, yaitu water bath (30°C-31°C) dan non-water bath (26°C-30°C). Hasil penelitian menunjukkan perbedaan rasio jantan dan betina tidak memengaruhi banyaknya jumlah pasangan induk yang memijah. Jumlah rata-rata telur pada perlakuan A (133,5 ± 41,72 butir) lebih banyak dibanding perlakuan B (108 ± 24,04 butir). Inkubasi telur menggunakan water bath menghasilkan jumlah telur yang menetas 215% lebih tinggi dibandingkan inkubasi non-water bath. Embrio ikan peacock goby mulai menetas pada hari kelima dan berakhir pada hari ketujuh setelah pemijahan, dan mencapai fase benih pada umur 33 hari setelah menetas. Survival activity index larva ikan peacock goby diperoleh nilai sebesar 10,6-35,45.Peacock goby (Tateurndina ocellicauda) is one of the ornamental fish inhabited peat waters stretched from the Papua region to the eastern part of Papua New Guinea. Despite its increasing exploitation and population pressure, there are currently limited studies and information regarding the biological aspects of the peacock goby fish. The research objective was to determine the spawning, embryogenesis, and larval ontogeny characteristics of the peacock goby in cultured setting. Nine of mature male broodstock (body weight 0.69 ± 0.08 g) and six of mature female broodstock (body weight 0.65 ± 0.03 g) resulted from gonad selection were reared in a plastic container measuring 20 cm x 25 cm x 15 cm in size. The ratios of male and female fish were 1:1 (treatment A) and 2:1 (treatment B), with three replications. The spawning container was equipped with a semicircular piece of PVC pipe for eggs shelter. The fertilized eggs were incubated in two hatching containers fitted with a water bath (30°C-31°C) and without a water bath (26°C-30°C). The results showed that the difference in the ratio of males and females did not affect the number of broodstock pairs to spawn. The average number of eggs in treatment A (133.5 ± 41.72 eggs) was higher than treatment B (108 ± 24.04 eggs). Egg incubation using water bath resulted in a higher number of hatched eggs by 215% than that of without a water bath. Peacock goby fish embryos began to hatch on the fifth day and ended on the seventh day. The larvae reached the juvenile phase at 33 days after hatching. Survival activity index (SAI) of peacock goby fish larvae were between 10.6-35.45.
