Abstract

Penelitian ini bertujuan untuk mengkaji reproduksi bulubabi (Tripneustes gratilla) yang hidup pada ekosistem lamun dan ekosistem berpasir di perairan Pulau Barrang Lompo, dengan menganalisis aspek reproduksinya antara lain Indeks Kematangan Gonad dan Diameter Telur. Pengambilan sampel dilakukan mulai akhir juli hingga desember 2017 Jumlah bulubabi yang diperoleh pada ekosistem lamun sebanyak 273 jantan dan 239 betina pada ekosistem berpasir sebanyak 217 jantan dan 160 betina. Hasil penelitian menunjukkan Indeks Kematangan Gonad dan diameter telur bulubabi (Tripneustes gratilla) pada ekosistem lamun dan ekosistem berpasir di Pulau Barrang Lompo. menunjukkan tipe pemijahan bulubabi Tripneustes gratilla termasuk dalam tipe pemijahan sebagian atau partially spawner yaitu mengeluarkan telur matang secara bertahap pada satu kali periode pemijahan atau tipe pemijahan partial spawner.

Abstract

Tujuan Penelitian ini untuk menganalisis toksisitas daun miana sebagai antibakteri pada udang vanamei. Penelitian ini dilaksanakan pada Maret sampai Oktober 2019, di Laboratorium Sekolah Tinggi Teknologi Kelautan (STITEK) Makassar. Sampel daun miana berasal dari beberapa daerah di Sulawesi Selatan. Uji aktivitas antibakteri ekstrak daun miana dengan tiga fraksi, yaitu ethanol, n-hexane, dan chloroform menggunakan metode difusi kertas cakram dengan konsentrasi ekstrak 500 µg/mL dan 1000 µg/mL, sebanyak tiga kali ulangan. Uji toksisitas metode BST (LC70) dengan konsentrasi uji 2500, 1250, 625, 312.5, 156.25, 78.125 µg/mL. Data yang diperoleh dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa uji aktivitas ekstrak daun miana tertinggi pada fraksi chloroform, dengan diameter hambat 28 mm. Sementara pada antibiotik oxytetracycline diameter hambat sebesar 31 mm. Nilai toksisitas pada tingkat mortalitas 30% pada konsentrasi 1250 µg/mL. Ekstrak daun miana fraksi chloroform mampu menggantikan peran antibiotic oxytetracycline dengan penggunaan konsentrasi dibawah 1250 µg/mL.

Abstract

ABSTRACT: The present study aimed to determine the effect of antibiotic on the survival and growth performance of K. alvarezii using in vitro technique. This research was conducted for 2 months in Pangkep State of Agricultural Polytechnic at Tissue Culture Laboratory. The seaweed samples were taken from waters of Pangkep Regency. The fresh and healthy thallus branches were used as explants. The culture medium was conwy liquid media added vitamin mix with the treatment of different antibiotic concentrations. The parameters observed were survival rate, number of shoots, shoot length. A completely randomized design and 5 treatments with 3 replications were applied. One way ANOVA was performed for data analysis. The results showed that survival rate and shoot length and the highest number of shoots were found at 10 ppm antibiotic treatment with 56.6% of survival rate, the average number of shoots was 6, the average length of shoots was 1.86 mm. while a total 30% of survival rate was found at 12 ppm treatment with a number of shoots reaching 5.4 and a shoot length of 1.79. ABSTRAK:Tujuan penelitian ini adalah untuk mengetahui pengaruh perbedaan dosis antibiotik pada media kultur jaringan terhadap kelangsungan hidup dan pertumbuhan rumput laut K. alvarezii secara in vitro. Penelitian ini dilakukan selama 2 bulan di Laboratorium Kultur Jaringan Politeknik Pertanian Negeri Pangkep. Bahan uji yang digunakan adalah bibit rumput laut jenis Kappaphycus alvarezii yang diperoleh dari perairan Kabupaten Pangkep. Bibit yang digunakan adalah bibit segar, muda, memiliki percabangan yang banyak dan sehat. Media kultur adalah media cair conwy dengan campuran vitamin mix dengan perlakuan perbedaan konsentrasi antibiotik yang berbeda. Wadah percobaan adalah botol kultur dengan volume 200 ml sebanyak 15 buah. Parameter yang diamati sintasan, jumlah tunas, panjang tunas. Rancangan yang digunakan adalah Rancangan Acak lengkap (RAL) dan 5 perlakuan sebanyak 3 ulangan. One way ANOVA digunakan untuk analisis data. Hasil penelitian memperlihatkan bahwa sintasan dan pertumbuhan panjang tunas serta jumlah tunas tertinggi adalah pada perlakuan antibiotik 10 ppm. Sintasan eksplan rumput laut mencapai 56.6%, rata-rata jumlah tunas mencapai 6, rata-rata panjang tunas mencapai 1.86 mm. sedangkan perlakuan sintasan untuk perlakuan 12 ppm hanya 30%, dengan jumlah tunas mencapai 5.4 serta panjang tunas 1.79.

Abstract

This study aims to determine the type of seaweed that is good and can produce maximum growth and survival of Siganus guttatus. This research was carried out from June-September 2021, at at the Shrimp Hatchery Installation (IPUW), Barru, South Sulawesi. This study used a completely randomized design (CRD) used three treatments with three replications. The treatments used were treatment A (100% control of commercial feeding), treatment B (feeding Gracilaria sp. 100 %), treatment C (feeding Ulva sp.). The parameter measured were Specific Growth Rate (SGR), Absolute Growth, Survival and Palatability. The data was analyzed using Analysis of Variance (ANOVA) and with the W-Tukey test if there were differences between treatments. 180 Siganus guttatus juveniles (average weight of 5.65 g) were reared in a tanks (62x41x51 cm) for 75 days. The highest daily growth was obtained in commercial feed A (PF 1000) which was 1.53 g, the highest absolute growth was obtained in treatment A (Commercial Feed) 12.29 g, both treatments B (Gracilaria sp.) 4.30 g and the lowest was treatment C (Ulva sp.) .) 2.82 g. The highest survival rate (survival rate) was obtained in treatment A (Commercial Feed) and B (Gracilaria sp.) 98.33%, and the lowest was in treatment C (Ulva sp.) with an average value of 85.00%. Based on the results of the ANOVA test, it can be concluded that the research conducted by providing commercial feed (PF 1000), Gracilaria sp. and Ulva sp. significant effect (P<0.05) on specific growth, absolute growth, survival and palatability of baronang fish (Siganus guttatus). The results showed that the highest feed consumption value was in treatment A of 144.57 g, followed by Treatment B (Gracilaria sp.) of 142.19 g and the lowest of treatment C (Ulva sp.) of 100.01 g.
