Abstract

The purpose of this study was to observe the effect of extender and cooling rate on the quality of frozen thawed semen of Bali bull (Bos sondaicus). The experiment was conducted at Laboratory of Animal Biotechnology, Faculty of Animal Husbandry, Andalas University. A completely randomized design of factorial 2 x 3, 2 different extenders and 3 cooling rates with 3 bulls as the replicate. Data were analyzed using analysis of variance (ANOVA). The first factors was extender such as tris citrate egg yolk (TCEY) and tris citrate soy milk (TCSM). The second factors was the cooling rate such as: 15ºC/min, 10ºC/min and 5ºC/min. Variables were: motility, viability, abnormality, and membrane integrity. The results of treatment of TCEY extender on the average of motility 42.67±5.17%, viability of 53.78±3.79%, abnormality of 18.89±1.07%, and membrane integrity of 35.44±3.01%. The effect of cooling rate of 10°C /min has the highest semen motility of 44.35±5.28%, viability of 57.17±1.18%, abnormality of 17.84 ± 0.23 % and membrane integrity of 36.83±2.12%. The interaction between extender and cooling rate was founded significant different (P<0.05). Otherwise of extender and cooling rate on post-thawed of motility, viability and membrane integrity, had no difference (P>0.05) on semen abnormality. It can be concluded that tris-citrate egg yolk (TCEY) extender with cooling rate of 10ºC / min were the best semen motility, viability, and membrane integrity of spermatozoa.

Abstract

This study aimed to evaluate different recovery techniques of oocytes and the duration time between ovary collection and processing in the laboratory on the quality and quantity of Simmental cross bovine oocytes in vitro maturation. A total of 75 bovine ovaries were divided into three groups. Experiment-1;  the storage duration of ovaries were 6, 9 and 12 h. Experiment -2;  the techniques for oocytes recovery were slicing, aspiration and slicing + aspiration. The ovaries were collected from slaughterhouse and kept in saline solution during transportation to laboratory. A total of 136 oocytes were used for experiment-1, and 246 oocytes for experiment-2. This study found that the number and quality of oocytes were significantly different (P<0.05) as the effect of storage time. The average number of oocytes were 15.90±2.48; 17.10±1.38 and 13.60±3.00, in 6 h; 9 h and 12 h of storage time, respectively. The techniques of oocytes collection significantly (P<0.05) affect the quantity of oocytes but no significant (P>0.05) on oocytes quality. The average of oocytes of groups slicing, aspiration and slicing plus aspiration were 14.66±2.09; 9.46±2.99 and 16.40±6.86, respectively. Both experiment was significant effect (P<0.05) on immaturation rate of bovine. Conclusion of this study the storage time of ovaries was suitable in 9 h and the technique of oocytes collection was in slicing plus aspiration.

Abstract

AbstractThe purpose of this study was to know the quality of Boer liquid semen during storage by adding sweet orange essential oil and gentamicin into the tris yolkextender.The semenlongevity test was carried out by storing semen in a closed tube at room temperature and refrigerator, and evaluated motility and Viability every 3 hours at room temperature and 12 hours in refrigerator. The results showed that the characteristics of the liquid semen, with the addition of 1% of essential oils showed percentage of motility and Viability significantly higher than 0.5% and without the addition of essential oil, either stored at room temperature or in the refrigerator. This may be related to the content of essential oils of sweet orange peel containing flavonoids and antibacterials that are capable of maintaining liquid semen quality of Boer Goat.In addition, gentamicin contains antibacterials capable of suppressing the growth of bacteria which can damage and durabilitythe spermatozoa.Keywords: Boer Goat,Gentamicin, Liquid semen, Sweet orange

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian PGF2 (Capriglandin dan Lutalyse) terhadap service perconception, conception rate dan  ovarium, folikel, dan corpus luteum pada induk sapi simmental. Penelitian menggunakan 24 ekor induk sapi Simmental yang tidak bunting di BPTUHPT Padang Mengatas, Sumatera Barat dengan kriteria tidak estrus, siklus estrus normal dan tidak mengalami gangguan reproduksi. Metode eksperimen dengan Rancangan Acak Lengkap Pola Faktorial 2 x 2 x 6 bila terdapat hasil berbeda nyata maka dilanjutkan uji lanjutan DMRT. Faktor A adalah preparat hormon prostaglandin (Capriglandin dan Lutalyse). Faktor B adalah dosis (3 ml dan 5 ml). Perlakuan A1B1 = Capriglandin 3 ml; A1B2 = Capriglandin 5 ml; A2B1 = Lutalyse 3 ml; dan A2B2 = Lutalyse 5 ml yang masing-masing terdapat enam ulangan. Baik Capriglandin dan Lutalyse diberikan secara single dosis intra muskuler. Parameter yang diukur service perconception, conception rate dan ukuran diameter panjang ovarium. Hasil analisis antara preparat hormon Capriglandin dan lutalyse terhadap jumlah dosis 3 ml dan 5 ml menunjukan hasil berbeda nyata (P kecil dari 0.05) terhadap conception rate dan ovarium. Hasil analisis tidak berbeda nyata (P besar dari 0,05) terhadap service perconception. Prostaglandin dengan zat aktif dinoprost tromethamine hanya efektif bila ada CL fungsional, yaitu antara hari ke-7 sampai hari ke-18 siklus estrus. Penggunaan terbaik pada pemberian Capriglandin dengan dosis 5 ml.

Abstract

This study aimed to find  the best combination between pre-freezing method and layer of post sexing on semen quality of Pesisir Bulls. This study used 2 Pesisir Bulls. The semen was evaluated using Computer Assisted Sperm Analyzer (CASA). This study used factorial randomized block design with 2 factors. Factor A was sexed semen layer of BSA column with 2 level, whereas Factor B was pre-freezing method with 3 level. Analysis of Variance (ANOVA) were used in this study for data Analysis. According to the analysis of variance, there is no interaction of the combination of two factors on each paramters. Pre-freezing method showed highly statistically significant effect (P<0.01) on the quality of motility on upper layer and bottom layer which was about (52.86-72.93%; 52.32-65.50%) and about (40.17-56.99%; 40.33-53.18%) on progressive motility. The value of Distance Curve Line on  (39.15-43.24 μm; 49.25-53.86 μm), Distance Average Path (22.38-24.60 μm; 26.52-29.94 μm), Distance Straight Line (17.32-19.07 μm; 20.93-24.25 μm). The value of Velocity Curve Line (88.67- 97.50 μm/s; 109.85-117.90 μm/s), Velocity Average Path (50.98- 55.63 μm/s; 59.32- 64.69μm/s), Velocity Straight Line (34.45-42.37 μm/s; 46.80-50.62 μm/s), Beat Cross Frequancy (23.91-25.17 Hz; 25.63-27.35 Hz), Straightness (0.75-0.77; 0.77-0.80), and Wobble (0.56-0.57; 0.53-0.55), highly statistically significant (P<0.01) on post sexing layers. Meanwhile, the value of Amplitude of Lateral Head (4.24-4.94 μm; 4.25-4.44 μm) showed highly significant effect (P<0.01) on factor A and factor B treatment, and value Linearity was about (0.42-0.44; 0.43-0.44) showed not significant effect (P>0,05) on every factor. According to the study, it can be concluded that the best treatment was treatment modified procedure layer by space the straw 16 cm every layers and The quality of post sexing X- and Y- spermatozoa motility effect on motility characteristic such as DCL, DAP, DSL, VCL, VAP, VSL, STR, WOB, ALH and BCF.
