Abstract

Background. Regenerative medicine is a new therapeutic modality using cell, stem cell and tissue engineering technologies. Purpose. To describe the regenerative capacity of dental pulp mesenchymal stem cell. Review. In dentistry, stem cell and tissue engineering technologies develop incredibly and attract great interest, due to the capacity to facilitate innovation in dental material and regeneration of dental and oral tissues. Mesenchymal stem cells derived from dental pulp, periodontal ligament and dental follicle, can be isolated, cultured and differentiated into various cells, so that can be useful for regeneration of dental, nerves, periodontal and bone tissues. Tissue engineering is a technology in reconstructive biology, which utilizes mechanical, cellular, or biological mediators to facilitate regeneration or reconstruction of a particular tissue. The multipotency, high proliferation rates and accessibility, make dental pulp as an attractive source of mesenchymal stem cells for tissue regeneration. Revitalized dental pulp and continued root development is the focus of regenerative endodontic while biological techniques that can restore lost alveolar bone, periodontal ligament, and root cementum is the focus of regenerative periodontic. Conclucion. Dentin-derived morphogens such as BMP are known to be involved in the regulation of odontogenesis. The multipotency and angiogenic capacity of DPSCs as the regenerative capacity of human dentin / pulp complex indicated that dental pulp may contain progenitors that are responsible for dentin repair. The human periodontal ligament is a viable alternative source for possible primitive precursors to be used in stem cell therapy.

Abstract

Background: Angiogenesis is an important and fundamental process for new blood vessels to provide nutrients and oxygen needed by tumor cells to grow, develop, and in case of cancer also to metastasize into other organs. This study aims to evaluate the intensity of angiogenesis within benign (papillomas) and malignant (squamous cell carcinoma) epithelial tumors.Materials and methods:  This analytic observational study with cross-sectional design using histopathology slide sample that were clinically diagnosed as squamous cell carcinoma (n=3) and papilloma (n=3). Microscopically, the angiogenesis characterized with lumen lined by endothelial cells with or without red blood cells inside within sub epithelial connective tissue of papilloma and squamous cell carcinoma by Hematoxylin Eosin stain. Angiogenesis intensity was counted from four areas under magnification of (10x10), each area was scored under (10x40) magnification.Results: Angiogenesis intensity of papilloma and squamous cell carcinoma are (45.17±14.573) and (55.18±6.26041) respectively. T-test analysis showed there was no significant difference (p=0.336>0.05).Conclusions: Angiogenesis intensity of papilloma is less than those of squamous cell carcinoma.Keywords: angiogenesis, oral epithelial tumor, benign, malignant

Abstract

Squamous cell carcinoma (SCC) is the most common malignant tumour in oral cavity. This cancer arises from oral mucosal epithel and also it often causes high rate of morbidity and mortality. Metastasis is one of the major causes of mortality in patient with cancer. This is because metastasis has often occurred before the primary tumor it self is detected. Lymphogenous metastasis is specific for carcinoma. In the late stage, cancer cells from the lymphatic system may enter blood circulation. In brief, steps of the metastatic process may be outlined as follows: genetic alteration, modulation of extracellular matrix and proteolysis, damage cell adhesion, damage tumour cell migration, and also angiogenesis. The involved process factors are genetic factors such as nm23, p16, Lin-7C; proteolitic enzyme such as MMP-1, MMP-2, MMP-3, MMP-8 MMP-9, MMP-14, MMP-15, MMP-17, cathepsin B, D, and O; adhesion between adhesive proteins for example integrin, laminin, cadherin, maspin. In conclusion, the whole process from genetic alterations until the occurence of independent cell is a complex process that involves interactions among the related factors.

Abstract

Sarang semut (Myrmecodia pendans) is one of traditional herbs used to treat various diseases. Myrmecodia pendanscontain chemical compound flavonoid which has anti inflammation effects. Pulpitis is a dental pulp inflammation thatcan be caused by bacteria invasion, physical trauma or iatrogenic factor which affecting blood vessels in dental pulp.This research is an in vivo laboratory experimental pre and post treatment with control. The purpose of this study is tofind out the effects of Myrmecodia pendans ethanol extract on blood vessels appearance of Sprague Dawley rats’dental pulps which had been inducted with 0,01ml Porphyromonas gingivalis for 48 hours. The subjects were dividedinto 3 groups with Group I representing the negative control, group II representing treated group with Myrmecodiapendans ethanol extract, and group III representing positive control with Ca(OH)2. Group II and III were divided intosubgroups based on the treatment periods of 48, 96, 168, 336 hours. The statistic results showed that there weresignificant differences (p<0.05) among the pulpitis, negative control, positive control Ca(OH)2 and the Myrmecodiapendans treatment subgroups of 48, 96, 168, 336 hours. Myrmecodia pendans ethanol extract has comparable antiinflammation effect with Ca(OH)2 which is used to pulpitis commercial medicament.

Abstract

Monosit akan masuk kedalam jaringan dan menjadi makrofag ketika terjadi peradangan. Makrofagsering hanya dihubungkan dengan fungsinya melakukan fagositosis benda-benda asing. Namunmakrofag memiliki fungsi lain yaitu angiogenesis dan fibrosis. Pada proses angiogenesis makrofagakan mensekresikan tumor necrosis factor-alfa (TNF-α), vascular endothelial growth factor (VEGF),angiogenin, urokinase dan platelet derived growth factor (PDGF) yang akan menginduksiterbentuknya pembuluh darah baru. Pada proses fibrosis makrofag akan mensekresikan fibroblastgrowth factor, platelet derived growth factor (PDGF) dan TGF-β (transforming growth factor beta)yang akan merangsang fibroblas menghasilkan kolagen dan elastin.

Abstract

Sarang semut (Myrmecodia pendans) merupakan tumbuhan yang dapat digunakan sebagai obat tradisional. Myrmecodia pendansmemiliki kandungan flavonoid yang memiliki efek antiinflamasi. Pulpitis adalah inflamasi pulpa yang disebabkan invasi bakteri dan mempengaruhi gambaran sel odontoblas pulpa. Penelitian observasional in vivo rancangan potong silang ini bertujuan untuk mengetahui efek ekstrak etanol tumbuhan Sarang semut (Myrmecodia pendans) terhadap sel odontoblas pada pulpitis. Penelitian ini menggunakan sedian histopatologi yang berasal dari pulpa gigi tikus Sprague Dawley yang diinduksi pulpitis dengan Porphyromonas gingivalis selama 48 jam. Gambaran sel odontoblas dievaluasisecara mikroskopis pada kelompok I (pulpa normal), kelompok II kontrol negatif (pulpitis tanpa treatment), kelompok III perlakuan (pulpitis dengan Sarang semut), kelompok IV kontrol positif (pulpitis dengan CaOH2). Kelompok II, III dan IV dibagi menjadi sub kelompok pengamatan dengan waktu perlakuan 48, 96, 168, 336 jam. Efek antiinflamasi tertinggi didapatkan dari kelompok perlakuan (pulpitis dengan Sarang semut) pada jam ke-48, 96, dan 168 dibandingkan dengan kelompok kontrol positif (pulpitis dengan CaOH2). Hasil uji Kruskal Wallis menunjukkan efek signifikan (p<0.05) antara kelompok pulpa normal, kontrol negatif, positif, dan perlakuan pada 48, 96, 168, 336 jam. Hasil penelitian menunjukkan ekstrak tumbuhan sarang semut mempunyai efek antiinflamasi setara dengan Ca(OH)2 yang biasa digunakan sebagai obat komersial pada pulpitis.

Abstract

Binahong atau Anredera cordifolia {Tenore) Steenis merupakan suatu tanaman obat yang berasal dari daratan China dan juga ditemukan di Lembang dan daerah sekitarnya. Daun tanaman ini mengandung saponin dan alkaloid yang merupakan kandungan aktif anti jamur. Candida a/bicans merupakan organisme penyebab infeksi rongga mulut yang paling umum. Penelitian inibertujuan untuk mengetahui efek anti-jamur ekstrak etanol 70% daun binahong (BLEE) terhadap Candida albicanssecara in vitro. Ujiinhibisipada penelitian ini menggunakan metode difusi dari pelbagai konsentrasi BLEE mulai 0.2% sampai 100% dengan melakukan streakingpada media agar, sementara suspensi oral nystatin dan akuades digunakan sebagai kontrol positif dan negatif. Ujidengan metode streakingdilakukan pada konsentrasiBLEE yang dengan metode dilusi memberikan hasil positif atau suspensilarutan tetap keruh.Hasil penelitian ini menunjukkan bahwa koloni Candida albicanstidak  ditemukan pada BLEE 25% sehingga diasumsikan bahwa konsentrasi inimerupakan nilaiminimum antijamur (MFC).Jumlah koloni terkecil ditemukan pada konsentrasi BLEE 12,5% yang merupakan konsentrasi hambat mini­ mal (MIC). Juga tidak ditemukan perbedaan bermakna dari efek hambat Candida a/bicans antara pelbagai konsentrasi BLEE dan dengan suspensi oral nystatin (p>0.05 ;t test=O)

Abstract

Minuman probiotik yang mengandung mikroorganisme hidup berperan penting bagi kese­ hatan dengan mempengaruhi sistem imun sehingga dapat mengurangi peradangan. Lac­tobacillus  casei mempengaruhi sistem imun melalui mekanisme selular dan humoral.  Tu­ juan penelitian  ini adalah  untuk mengetahui  efek minuman  probiotik  yang mengandung Lactobacillus  casei strain Shirota terhadap proses penyembuhan lesi mukosa mulut. Se­jumlah 36 tikus Sprague dawley digunakan dalam penelitian ini dibagi secara radom dalam 4 kelompok (n=9): K- (kelompok kontrol negatif), P-1 (kelompok perlakuan dengan pembe­ rian larutan probiotik secara sistemis), P-2 (kelompok perlakuan dengan pemberian larutan probiotik secara topikal), K+ (kelompok kontrol positif dengan aplikasi topikal gel asam hialuronik). Lesi mukosa mulut dibuat dengan menggosok mukosa labial menggunakan larutan H202 3%. Proses penyembuhan diamati secara histopatologi pada hari ke 2, 4, dan 7. Berdasarkan uji Kruskal Wallis, ada perbedaan bermakna antara kelompok perlakuan (p=0,001<0,05). Pada hari ke 2, tidak ditemukan perbedaan bermakna antar kelompok (MRD=4,67<5,33). Pada hari ke 4, ditemukan perbedaan bermakna antara kelompok per­ lakuan (P-1 dan P-2) dengan kelompok kontrol negatif (K-) dan kontrol positif (K+) dengan MRD=14 dan 9,33 (>5,33). Pada hari ke 7, kelompok P-1 menunjukkan nilai MRD yang sama dengan kelompok K+, namun menunjukkan perbedaan bermakna dengan kelompok K- dan P-2 (MRD=6>5,33). Penelitian ini menyimpulkan bahwa aplikasi minuman probiotik yang mengandung Lactobacillus  casei strain shirota baik secara sistemik maupun topikal dapat mempercepat penyembuhan lesi mukosa mulut. Aplikasi sistemis lebih efektif pada stadium yang lebih lanjut (hari ke 7) sedangkan aplikasi topikal lebih efektif pada stadium awal (hari ke 4).
