Abstract

BACKGROUND: Cancer stem cells (CSCs) eradication might serve as a robust approach for cancer eradication. MCF-7 as breast cancer continuous cell line is known to contain breast CSCs (BCSCs) for its capability to maintain its original tumor population. CSCs enriched culture is a fundamental tool for CSCs targeted therapy development. Effective and unsophisticated CSCs dedifferentiation protocol for producing CSCs enriched culture is needed.METHODS: MCF-7 cells were cultured initially in Dulbecco's Modified Eagle Medium (DMEM) low glucose medium then changed to DMEM:F12. Serum starvation was performed during each medium refreshment gradually with fetal bovine serum (FBS) concentration of 10%, 5%, 2.5% until reaching 1% FBS concentration. Stable MCF-7 culture was then adapted to serum free culture system, containing DMEM:F12, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), and B27 supplement as dedifferentiation protocol for 18 days. Cluster of differentiation (CD)44 and CD24 double staining immunocytochemistry was performed to evaluate cell stemness.RESULTS: The population of cells expressing BCSCs markers (CD44+/CD24low) in non-adherent single cells subpopulation was significantly increased after the dedifferentiation procedure (70.39%) compared to control groups (0.71%) (p<0.05). In contrast, the expression of BCSCs marker in adherent single cells subpopulation and for both adherent and non-adherent mammosphere the BCSCs markers showed a stable expression.CONCLUSION: BCSCs enrichment of breast cancer cell cultures from MCF-7 breast cancer cell line can be performed. Breast cancer cell plasticity is observed during the dedifferentiation protocol. Development of dedifferentiation inducing protocols can serve as an important foundation for breast cancer therapy development through BCSCs elimination.KEYWORDS: breast neoplasms, cell line, dedifferentiation, immunohistochemistry, neoplastic stem cells

Abstract

Highlights:Neutrophil to lymphocyte ratio (NLR) has been reported by several studies for its role as a biomarker in various diseases, however, the role of NLR in testicular GCT is still unclear. The characteristics and responses of testicular GCT patients among Indonesian men show similarity to other reports worldwide. As a parameter, NLR shows promise to be used as a potential biomarker for prognosis in testicular GCT.Abstract:The global incidence of testicular cancer is 1-2% from all cancers. The attempts to maintain high therapeutic rates while decreasing the treatment-related side effects and toxicity have become the current concern. However, the reports regarding testicular germ cell tumors (GCT) in Indonesia are limited. Thus, we aimed to evaluate the clinical characteristics of testicular GCT patients undergoing bleomycin, etoposide, and cisplatin (BEP) chemotherapy, as well as their chemotherapy response and side effects. We reported the data of patients with Testicular Germ Cell Tumor from January 2015 to December 2019. Several data were retrieved, including patient demographics, tumor characteristics, treatment, and prognosis outcome. A total of 67 patients with testicular germ cell tumors were included in this study. The mean age was 28.9 years old. The chemotherapy regimens used were four cycles of (BEP) in 36 patients (53.7%), followed by three cycles of BEP in 22 patients (32.8%). Patients with seminoma GCT mostly had a complete response (54.1%), whereas most patients with non-seminoma GCT had progressive disease (47.8%). The multiple logistic regression analysis showed that NLR and S staging were independently associated with the patient’s response to chemotherapy (OR 2.14, 95% CI 1.22, 3.78, p <0.01,OR 9.43, 95% CI 1.81, 49.14, p < 0.01). The clinical characteristics and response of testicular GCT patients among Indonesian men showed similarity with the current worldwide data. The NLR could be used as a potential biomarker for prognosis.

Abstract

Background: In 2017, about 1.2 million people died because of Chronic Kidney Disease (CKD). Patients with CKD are known to have increased levels of oxidative stress which leads to decrease in NO production. NO is a highly reactive signaling molecule and a major determinant of vascular homeostasis. Thus, the decreased NO can be a risk factor for the development of atherosclerosis and increased cardiovascular risk. Meanwhile, Malondialdehyde (MDA) is known as excellent biomarker for oxidative stress. This study aims to determine the correlation of serum total nitric oxide (NO) and urine MDA levels in non-hemodialysis CKD patients.Materials and Methods: This study was an observational clinical study with a cross sectional design. Fourty-nine CKD subjects were selected by consecutive sampling. The samples for laboratory tests were collected from urine. MDA concentration was measured using the High-Performance Liquid Chromatography (HPLC) kit. NO concentration was measured with Griess reaction method and Total Nitric Oxide Parameter kit. The data were analyzed using the Statistic Package for Social Science (SPPS) software version 16.Results: The data showed significant negative correlations between MDA with NO (r=-0.294; p=0.041).Conclusion: There was a correlation between serum total NO and urine MDA levels in non-hemodialysis CKD patients. Keywords: chronic kidney disease, malondialdehyde, nitric oxide, non-hemodialysis
