Abstract

Amentoflavone (1), a biflavonoid was isolated from leaves of Calophyllum inophyllum Linn. Isolation and purification of the compound used maceration and chromatography methods using Si-gell and sephadex LH-20. The structure was determined by UV, IR, 1H and 13C NMR spectroscopy includes HMQC/HMBC analysis and comparison with references.

Abstract

A xanthone derivative namely trapezifolixanthone has been isolated and identified from EtOAc extract of stem bark ofCalophyllum soulattri. The maceration method was used to isolates the trapezifolixanthone then followed by fractionation method using vacuum liquid chromatography and purification process by flash chromatography. A 14 mg of yellowish oily solid was extracted. The compound was elucidated by 1H NMR, 13C NMR, and 2D NMR (HSQC, HMBC, COSY) supported by UV and IR spectroscopy. Base on spectral data and comparison with some references, it is  known that the compound is trapezifolixanthone.

Abstract

Satu senyawa biflavonoid yaitu amentoflavon (1) berhasil diisolasi dari daun tumbuhan nyamplung (Calophyllum inophyllum Linn). Ekstraksi menggunakan metode maserasi dan pemurnian dengan kromatografi kolom dengan fasa diam Si-gel dan sephadex LH-20. Elusidasi struktur menggunakan metode spektroskopi UV, IR, 1H dan 13 C NMR beserta HMQC dan HMBC. Abstract

Telah dilakukan uji toksisitas secara Brine Shrimp Lethality Test (BSLT) ekstrak buah merah (Pandanus Conoideus Lamk.). Uji toksisitas dilakukan pada ekstrak etanol dan n-heksana. Hasil uji BSLT menunjukkan rata-rata prosentase kematian ekstrak n-heksan lebih tinggi dibandingkan dengan ekstrak etanol, sebesar 54 % diukur pada konsentrasi 400 (μg/mL). Ekstrak n-heksan selanjutnya dilakukan pemisahan secara kromatografi kolom dengan elusi secara gradien menggunakan pelarut campuran n-heksana-etil asetat untuk memperoleh fraksi aktif. Uji toksisitas secara BSLT menunjukkan fraksi aktif mempunyai nilai LC50 sebesar 138,05 μg/mL. Identifikasi fraksi aktif diperoleh 3 komponen senyawa asam lemak yaitu asam palmitat, asam oleat dan asam miristat.Kata Kunci : Buah merah (Pandanus conoideus Lamk.), identifikasi, toksisitas, Brine Shrimp LethalityABSTRACT

Abstract

Chemical isolation and identification of root bark of Calophyllum soulattri Burm. f. have been conducted. Isolation was conducted by maceration with ethyl acetate as solvent. Ethyl acetate extract was separated and purified by vacuum liquid chromatography and flash chromatography which was guided by thin layer chromatography to obtain yellowish white solid. Isolated compounds were determined by spectroscopy methods such as UV, FTIR, 1H NMR, 13C NMR, DEPT, HSQC, HMBC and were compared to the references. Based on the analysis result it’s known that the compounds are the mixture of β-sitosterol and stigmasterol.

Abstract

A xanthone, named ananixanthone (1) has been isolated and identified from the ethyl acetate extract of the root barks of Calophyllum soulattri. Structure of the compound was determined based on spectroscopic data, including UV, IR, NMR 1D, NMR 2D and by comparison with references.

Abstract

The purpose of this research was to evaluate antibacterial activity of Neem (Azadirachta indica A. Juss) leaves fraction against some pathogenic bacterial and to identificate the most active fraction. Leaves powder of Neem was macerated with ethanol and factionated by vacuum liquid chromatography using hexane, ethyl acetate and ethanol as eluent, respectively. The antibacterial activity of the fraction was evaluated by diffusion method, then the most active fraction of antibacterial was evaluated by zone of inhibition. The most active fraction of antibacterial was evaluated for Minimum Inhibitory Concentration (MIC) and equivalent value, compared with amoxicillin and chloramphenicol, then identified using phytochemical screening and Gass Chromatography-Mass Spectroscopy (GC-MS).The fraction of Neem leaves fractionated by vacuum liquid chromatography had antibacterial activity againstStaphylococcus epidermidis, Bacillus cereus, and Shigella flexneri. The ethyl acetate fraction showed the most active fraction of antibacterial against all bacterial tested. Ethyl acetate fraction had MIC 0.075% againstStaphylococcus epidermidis 0.05% against Bacillus cereus and Shigella flexneri. The antibacterial activity of ethyl acetat fraction compared to amoxicillin was 0.01% for S. epidermidis, 0.02% for B. cereus, and 0.02% for S. flexneri. Then compared to chloramphenicol was 0.04% for S. epidermidis, 0.02% for B. cereus, and 0.06% for S. flexneri. The result of GC-MS showed that the ethyl acetate fraction contained palmitic acid, ethyl linoleolate, stearic acid, trans-phytol, DOP (di octyl phthalate), and suggested 1 class of compound fatty acid and 5 classes of compound triterpenoids.
