Abstract

This study aims to determine the phylogenetic position of rotifer Brachionus sp. sample originated from Tumpaan coastal water, North Sulawesi in the phylogenetic tree of Brachionus spp. from 27 samples of rotifer selected from 100 samples available in NCBI Genbank. The phylogeny tree was constructed on the basis of COI (Cytochrome Oxidase I) gene sequences using MEGA7 software and the neighbor-joining method. Results show that the phylogeny tree was divided into 2 clades, the group (closely related group) and out-group (distantly related group). It is confirmed that the position of the rotifer originating from Tumpaan Coastal Water is within in group, which has closely related to the B. plicatilis complex BUS06 indicated by a bootstrap value of 80%. The group belongs to SS-type rotifers with an average body size of 149±1.3 μm. This rotifer group has been known as live prey for fish larvae with a mouth-opening size of 150 μm.

Abstract

Sea cucumbers are one of the marine biotas which consist of high nutritional and protein content. One bioactive compound in sea cucumbers is triterpenoid with antifungal and anti -bacterial properties. This study aimed to obtain fungal symbionts derived from marine cucumbers from Molas Waters, North Sulawesi continued by extraction and testing their antibacterial property towards bacterial strains Escherichia coli and Staphylococcus aureus. The antibacterial activity test was carried out by using the Kirby-Bauer disc diffusion method. In the results, we found 3 species of sea cucumbers from Molas Waters, e. g. Holothuria atra, Bohadschia argus and Holothuria scabra. From these species, three isolates of fungal symbionts were isolated and tested for their antibacterial property. These three extracts array antibacterial activity against E. coli and S.aureus. The most potent antibacterial activity was found in the extract of fungal symbiont from H. atra with an average of 29.3 mm against E.coli and 28.0 mm against S.aureus. Keywords: Sea cucumber, antibacterial activity test, Escherichia coli ABSTRAK Teripang adalah salah satu biota yang memiliki kandungan gizi dan protein yang juga cukup tinggi dalam tubuh teripang Salah satu senyawa yang terkandung dalam teripang yaitu senyawa triterpenoid yang memiliki aktivitas sebagai anti jamur dan anti bakteri. Tujuan dari penelitian ini yaitu untuk mendapatkan isolat jamur dan ekstrak jamur yang bersimbiosis dengan teripang dan menguji aktivitas antibakteri terhadap strain Escherichia coli dan Staphylococcus aureus dengan metode difusi agar (disc diffusion Kirby-Bauer). Hasil penelitian didapatkan dari 3 jenis teripang dari Pantai Molas yaitu Holothuria atra, Bohadschia argus dan Holothuria scabra. Dari ketiga spesies teripang ini telah diisolasi tiga isolat jamur simbion untuk diuji aktivitas antibakterinya. Ketiga ekstrak jamur simbion asal teripang ini memiliki aktivitas antibakteri terhadap strain bakteri Escherichia coli dan Staphylococcus aureus. Aktivitas antibakteri yang paling kuat dari terdapat pada ekstrak jamur simbion asal Holothuria atra dengan rata-rata 29,3 mm terhadap bakteri Escherichia coli dan 28,0 mm terhadap bakteri Staphylococcus aureus.
