Abstract

Esherichia coli ESBLs bertanggungjawab terhadap terjadinya wabah infeksi nosokomial, peningkatan morbiditas dan mortalitas, serta peningkatan biaya kesehatan. Tujuan penelitian ini yakni untuk mendeteksi keberadaan gen SHV pada isolat klinik E.coli penghasil ESBLs dari urine pasien yang merupakan koleksi Laboratorium Mikrobiologi Klinik RSUD Dr. Soetomo Surabaya pada bulan Januari - Februari 2014. Jenis penelitian yang digunakan adalah observasional deskriptif dengan pendekatan molekul genetik. Metode yang digunakan untuk deteksi gen SHV menggunakan PCR (Polymerase ChainReaction) kemudian dielektroforesis dan divisualisasikan pada gel agarose 1.5%. Hasil penelitian menunjukkan bahwa dari tiga puluh isolat, sebanyak dua belas isolat (40%) positif mengandung gen SHV.

Abstract

Escherichia coli producing Extended-Spectrum Beta-Lactamases (ESBLs) responsible for the high number of disease and death from nosocomial infections because of an enzyme encoded by genes CTX-M. This aim of the research to identify isolates E.coli of urine patients, which is a collection of Clinical Microbiology RSUD Dr. Soetomo Surabaya on the January-February 2014. The kind of the research use is observational descriptive with molecule approachment. The sample used in clinical isolate of E.coli producing ESBLs accumulated during January-February 2014 collection Clinical Laboratory RSUD Dr. Soetomo Surabaya that is thirty isolate. The methods used for the detection of genotypic by PCR thus electrophoresis and visualized on agarose gel 1.5%. The results show that twenty-seven isolate (90%) positive containing a gen CTX-M with the highest number found on the Interna Departement. Detection Bacteria producing ESBLs in genotype important that therapy an antibiotic the patients are given more effective and efficient.

Abstract

Extended Spectrum Beta Lactamases (ESBLs) merupakan enzim yang mampu menonaktifkan antibiotik golongan beta laktam generasi ketiga dan keempat, serta monobaktam. Escherichia coli merupakan kelompok Enterobacteriaceae yang paling banyak ditemukan menghasilkan enzim ESBLs. Hal tersebut menyebabkan semakin meningkatnya biaya perawatan di rumah sakit, angka kesakitan, dan angka kematian di berbagai negara, termasuk di Indonesia. Penelitian ini bertujuan untuk mengetahui aktivitas nanokomposit ZnO-Ag dalam menghambat pertumbuhan bakteri Escherichia coli penghasil ESBLs. Nanokomposit ZnO-Ag dibuat dengan metode refluks secara one pot synthesis menggunakan minyak cengkeh sebagai reduktor melalui green synthesis. Nanokomposit ZnO-Ag yang terbentuk yakni 73,21 nm kemudian diujikan pada bakteri E.coli penghasil ESBLs dengan metode Kirby-Bauer. Konsentrasi nanokomposit 100 µg/ml merupakan konsentrasi yang efektif dalam menghambat pertumbuhan E.coli penghasil ESBLs.

Abstract

Participants of the Chronic Disease Management Program (PROLANIS) at the Trosobo Health Center are a group of patients with diabetes mellitus and hypertension whose condition needs to be monitored regularly. Diet is one of the important factors that affect the stability of the health condition of patients in the Prolanis group. The purpose of this program is to provide education on healthy eating patterns for Prolanis participants. The method of activities carried out is counseling and workshops with a total of 30 patients. Counseling activities and workshops are carried out by involving families who care for patients. Evaluation of the success of the program is determined by the patient's level of understanding about healthy eating patterns and fasting blood sugar levels and the patient's blood pressure to normal and stable levels over time. Based on the results of the program evaluation that has been carried out, 90% of Prolanis patients understand about healthy eating patterns and apply them in their daily lives.

Abstract

Escherichia coli is a group of Enterobacteriaceae bacteria that often contaminate food so that it can cause diarrhea. These bacteria are very difficult to treat if they are able to produce the Extended-Spectrum Beta-Lactamases (ESBLs) enzyme. The purpose of this study was to identify ESBLs-producing E. coli in food samples in Krian Sidoarjo. Food samples (fried foods, cilok tempura and chili sauce) were collected from ten different places. The sample was then grown on Eosin Methylene Blue (EMB) medium and purified by the 16 streak method, as well as biochemical character tests. The ESBLs phenotypic E. coli method was carried out by screening test and confirmation test using a Double Disk Synergy Test (DDST). Thirty colonies were able to grow on EMB media, but after microscopic identification and biochemistry testing only four samples were E. coli positive and were able to produce ESBLs from the phenotypic test that had been carried out. ESBLs-producing E. coli testing is important not only for nosocomial infections but also for the community so it needs attention to the spread of ESBLs resistance among microorganism species Escherichia coli termasuk kelompok bakteri Enterobacteriaceae yang sering mengkontaminasi makanan sehingga dapat menyebabkan diare. Bakteri ini sangat sulit diobati apabila mampu memproduksi enzim Extended Spectrum Beta-Lactamases (ESBLs). Tujuan penelitian ini adalah untuk mengindetifikasi E. coli penghasil ESBLs pada sampel makanan di Krian Sidoarjo. Sampel makanan (gorengan, cilok, tempura, dan saus sambal) dikumpulkan dari sepuluh tempat berbeda. Sampel kemudian ditumbuhkan pada medium Eosin Metilen Blue (EMB) dan dimurnikan dengan metode streak 16, serta dilakukan karakteristik uji biokimia. Metode fenotipik E. coli penghasil ESBLs dilakukan dengan uji skrining dan uji konfirmasi menggunakan double disk synergy test (DDST). Sebanyak tiga puluh koloni mampu tumbuh pada media EMB, namun setelah diidentifikasi mikroskopis dan uji biokiomia hanya empat sampel positif E. coli dan mampu menghasilkan ESBLs dari uji fenotipik yang telah dilakukan.
