Abstract

Ten isolates of Lactobacillus plantarum were selected for its survival after heating. Two isolates which showed the highest survival rate were then encapsulated with skim milk, Arabic gum and mixtured of skim milk - Arabic gum. The probiotic cultures were prepared in the form of suspension and biomass and then spray dried. Survival of the probiotic  encapsulated in the  form of biomass after spray drying, and the viability after one month of storage at room temperature were higher than the suspension. It was found that the survival of probiotic bacteria in different encapsulation materials varied after spray drying. The number of probiotic survivals decreased after spray drying for all encapsulation materials tested ranged from less than 1 log cycle to 2 log cycles. Encapsulation of probiotic with three types combination of encapsulating materials showed that no significant difference of survivals after spray drying and viability after one month of storage were observed. The remained number of the probiotic after one month of storage at 4oC and room temperature was 104-107 cfu/gand 102-105 cfu/g, respectively. Abstract

Fermentation of rice by Monascus purpureus could produce red yeast rice (angkak) that can be used as drugs to decrease cholesterol and triglycerides increase thrombocyt in patients of dengue fever. To increase the immune system is needed the compounds that can be produced by microorganisms. The aim of study is to know that several strains of Monascus purpureus could produce the immunomodulatory effects or not. The study is about the invitro of phagocytosic activities of macrophage cell and polymorphonuclear of mice. This study had been used the fermented rice extract of five concentration such as 1 mg/ml, 0.1 mg/ml, 0.01 mg/ml, 0.001 mg/ml and 0.0001 mg/ml. As positive control was used 5 mg/ml extracts of meniran herbal, suspension of Staphylococcus aureus and the liquid of mice peritoneum. The results showed that the extract of fermented rice of Monascus purpureus with 1 mg/ml concentration was the same effectiveness as positive control. The conclusion was the extract of fermented rice could increase the fagocyticyt activities of can be concluded that administration of extract some kind of Monascus purpureus to increase the phagocytosis activities of macrophages and polymorphonuclear cells of mice.

Abstract

The need of medicine product are increasing now.The fermentation of rice by using Monascus purpureus produce lovastatin, pigments and citrinin as secondary metabolites. The aim of this study was to know the concentration of lovastatin, red and yellow pigments and citrinin in rice which fermented by Monascus purpureus that had been mutated by using ultraviolet irradiation.Three strains were used in this research, such as AsK, yellow Jmba and red Jmba.The result were analyzed by using HPLC and Spectrofotometer.The result showed that the yellow Jmba strain which treated by UV mutagenesis for 3 minutes contain higher lovastatin (1.798%) and pigment and lower citrinin (0.019%) compare to without mutagenesis.On the other hand ASK strain produced higher lovastatin (1.903%) but higher citrinin (1.270%).This research is expected to provide scientific information to the public.That M. purpureus contained lovastatin that is beneficial,red and yellow pigments for natural food colorant.

Abstract

The pyrene dioxygenase coded by gene is an indicator of bacterial isolates capabilities in pyrene degradation. The encoded gene of pyrene dioxygenase can be amplified and detected in Pseudomonas sp. PyrA2 and Burkholdoriasp. PyrA4 isolates, using primary specific Diox which designed based on PhdF gene sequence, the coding gene of pyrene dioxygenase in Mycobacterium vanbalenii PYR-1. The sequence alignment of pyrene dioxygenase putative gene in both of bacterial isolates with the sequence of pyrene dioxygenase coding gene in M. vanbalenii PYR-1 shows the similarity percentage of 41 % and 42% with Pseudomonas sp. PyrA2 and Burkholdoria sp.PyRA4.

Abstract

Pseudomonas bacteria plays essential role in soil ecology such as decomposer and biological control. The bacteria were isolated on selective media and identified from five soil samples taken within area of Wamena Biological Gardens.There are six species Pseudomonas were indentified based on morphological characters and biochemical reaction.P. striata was found to be common in soil of the area.No pathogen Pseudomonas was indentified in all soil samples. Preliminary study on biological control for fungal pathogen Fusarium indicated that P. fluorescens, P. striata and P. cepacia have ability to inhibit Fusarium growth in vitro.
